Gene expression profile among undifferentiated iPS cells (Und. iPSCs) , mesoderm cells (KDR+PDGFRα+ (KP)) at day 3(d3) to (d5) , and d5 KP derived-cardiomyocytes (CMs d5-19).

ABSTRACT: Despite numerous reports of human induced pluripotent stem cell (iPSC)-derived cardiac progenitor cells which potentially differentiate into cardiomyocytes (CMs), defined cellular population which definitively differentiate into CMs under diverse conditions is not reported so far. To identify cardiomyocyte-fated progenitors (CFPs) which exclusively differentiate into CMs under various culture conditions in vitro, we performed microarray analysis to detect candidate genes related to CFPs by examining the increase of temporal gene expression level during differentiation days 3 to 5.

We identified 197 known and 78 new candidate microRNAs during reproductive development of B. juncea. Large conserved domains of low DNA methylation maintained by 5-hydroxymethycytosine and Dnmt3a (CMS bisulfite-seq)

Project description:Gains and losses in DNA methylation are prominent genomic features of all mammalian cell types. To gain insight into mechanisms that could promote shifts in DNA 5-hydroxymethylation (5hmc) patterns and thus contribute to cell fate, including malignant transformation, we performed genome-wide mapping of 5hmc in purified wild type and Dnmt3a conditional knockout hematopoietic stem cells (HSCs) using cytosine-5-methylenesulphonate sequencing (CMS-seq). Comparing anti-CMS pulldown to input control, we identified 107,549 and 175,682 peaks of 5hmc enrichment in control and Dnmt3a knockout HSCs, respectively. Genome-wide transcriptomes of adult mouse cardiomyocytes (CMs) with ectopic expression of reprogramming factors

Project description:We carried out bulk RNA sequencing of normal adult mouse CMs (Control-CMs) and adult mouse CMs with ectopic overexpression of Yamanaka factors (Oct4, Sox2, Klf4, Myc, OSKM-CMs), based on a hypothesis that OSKM would induce at least partial CM dedifferentiation in vivo. More than 2,000 genes were differentially expressed between Control- and OSKM-CMs, which were enriched for gene ontology (GO) terms related to multiple key aspects of CM dedifferentiation. This study provides a genome-wide transcriptional profile of dedifferentiating CMs induced by OSKM.
