Project description:Hyperosmotic stress caused by drought and salinity is a significant environmental threat that limits plant growth and agricultural productivity. Osmotic stress induces diverse responses in plants including Ca2+ signaling, accumulation of the stress hormone abscisic acid (ABA), reprogramming of gene expression, and altering growth. Despite intensive investigation, no global regulators of all of these responses have been identified. Here, we show that the Ca2+-responsive phospholipid binding BONZAI (BON) proteins are critical for all of these osmotic stress responses. A Ca2+-imaging-based forward genetic screen identified a loss-of-function bon1 mutant with a reduced cytosolic Ca2+ signal in response to hyperosmotic stress. The loss-of-function mutants of the BON1 gene family, bon1bon2bon3, are impaired in the induction of gene expression and ABA accumulation in response to osmotic stress. In addition, the bon mutants are hypersensitive to osmotic stress in growth inhibition. BON genes have been shown to negatively regulate plant immune responses mediated by intracellular immune receptor NLR genes including SNC1. We found that the defects of the bon mutants in osmotic stress responses were suppressed by mutations in the NLR gene SNC1 or the immunity regulator PAD4. Our findings indicate that NLR signaling represses osmotic stress responses and that BON proteins suppress NLR signaling to enable global osmotic stress responses in plants.

Project description:SUMOylation is an essential and highly dynamic post-translational modification that regulates developmental processes and stress adaptations in plants to environmental cues. The global SUMOylation is quickly induced by dehydration and hyperosmotic stresses in plants, while the detailed mechanism underlying such SUMOylation dynamics is largely unknown. Herein, we report key components in osmotic stress and abscisic acid signaling, SNF1-related protein kinase 2s (SnRK2s), phosphorylate SUMO E3 ligase SAP and MIZ1-1 (SIZ1) to stabilize SIZ1 protein under stress conditions. The Ser820 residue is a functional SnRK2 phosphosite and non-phosphorylatable SIZ1S820A is unstable in vivo and in vitro. Under osmotic stress, the rapidly activated protein kinases SnRK2.4 and SnRK2.6 can phosphorylate SIZ1, which enhances the stability of SIZ1, and participates in the response to osmotic stress. However, the instability of SIZ1S820A protein was independent of COP1, indicating that there might be a new degradation mechanism for SIZ1. Our studies suggest that SnRK2s induced phosphorylation regulates SIZ1 stability to precisely modulates SUMOylation dynamics upon osmotic stresses. Additionally, such a reciprocal regulation by SnRK2 and COP1 might be a general regulation machinery that controls the stability of dozens of proteins.

Project description:Elucidating how plants sense and respond to water loss is important for identifying genetic and chemical interventions that may help sustain crop yields in water-limiting environments. Currently, the molecular mechanisms involved in the initial perception and response to dehydration are not well understood. Modern mass spectrometric methods for quantifying changes in the phosphoproteome provide an opportunity to identify key phosphorylation events involved in this process. Here, we have used both untargeted and targeted isotope-assisted mass spectrometric methods of phosphopeptide quantitation to characterize proteins in Arabidopsis (Arabidopsis thaliana) whose degree of phosphorylation is rapidly altered by hyperosmotic treatment. Thus, protein phosphorylation events responsive to 5 min of 0.3 m mannitol treatment were first identified using 15N metabolic labeling and untargeted mass spectrometry with a high-resolution ion-trap instrument. The results from these discovery experiments were then validated using targeted Selected Reaction Monitoring mass spectrometry with a triple quadrupole. Targeted Selected Reaction Monitoring experiments were conducted with plants treated under nine different environmental perturbations to determine whether the phosphorylation changes were specific for osmosignaling or involved cross talk with other signaling pathways. The results indicate that regulatory proteins such as members of the mitogen-activated protein kinase family are specifically phosphorylated in response to osmotic stress. Proteins involved in 5′ messenger RNA decapping and phosphatidylinositol 3,5-bisphosphate synthesis were also identified as targets of dehydration-induced phosphoregulation. The results of these experiments demonstrate the utility of targeted phosphoproteomic analysis in understanding protein regulation networks and provide new insight into cellular processes involved in the osmotic stress response.

Project description:Bacteria respond to osmotic stress by a substantial increase in the intracellular osmolality, adjusting their cell turgor for altered growth conditions. Using E. coli as a model organism we demonstrate here that bacterial responses to hyperosmotic stress specifically depend on the nature of osmoticum used. We show that increasing acute hyperosmotic NaCl stress above ~1.0 Os kg-1 causes a dose-dependent K+ leak from the cell, resulting in a substantial decrease in cytosolic K+ content and a concurrent accumulation of Na+ in the cell. At the same time, isotonic sucrose or mannitol treatment (non-ionic osmotica) results in a gradual increase of the net K+ uptake. Ion flux data is consistent with growth experiments showing that bacterial growth is impaired by NaCl at the concentration resulting in a switch from net K+ uptake to efflux. Microarray experiments reveal that about 40% of up-regulated genes shared no similarity in their responses to NaCl and sucrose treatment, further suggesting specificity of osmotic adjustment in E. coli to ionic- and non-ionic osmotica The observed differences are explained by the specificity of the stress-induced changes in the membrane potential of bacterial cells highlighting the importance of voltage-gated K+ transporters for bacterial adaptation to hyperosmotic stress.

Project description:The euryhaline marine yeast Debaromyces hansenii is a model system for the study of genes related to osmotic stress. To study the transcriptional response of this organism to osmotic stress we have used the two color microarray based gene expression analysis of 6211 genes which constitutes the whole genome. Analysis was done at three time points after induction with salt (0.5h, 3h and 6h.) The mRNA level of 64 genes significantly increased at least 3-fold after induction with 2M NaCl whereas that of 45 genes was 3-fold diminished. The induced as well as the repressed genes were grouped into functional categories to identify biochemical processes possibly affected by osmotic shock. 53% of the induced genes encode for ribosomal proteins, 12.5% for mitochondrial and redox, 6% for aminoacid, cell wall and carbohydrate, and 1.5%for heat shock, protein transport and glycerol proteins. The function of 31% of repressed genes is currently unknown. 15% of the repressed genes are involved in carbohydrate metabolism and protective function, 6% are associated with cell wall, redox and signal transduction, and 4% in vacuolar and lipid functions. Surprisingly, the activity of NAD+ glycerol 3-phosphate dehydrogenase gene (GPD1) involved in the glycerol biosynthesis in response to osmotic stress did not show induction. Then, 3 replicates were pooled and total RNA was extracted and prepared for smRNA deep sequencing. After normalization and annotation, we selected potential osmotic stress responsive smRNAs.
