Osteoblast-specific deletion of Hrpt2/Cdc73 results in high bone mass and increased bone turnover

ABSTRACT: RNA-seq within OCN-Cre;Cdc73flox/flox osteoblasts showed that loss of Cdc73 led to a derepression of osteoblast-specific genes, specifically those for collagen and other bone matrix proteins.

TENT5A mediated cytoplasmic polyadenylation in osteoblast supports proper bone formation.

Project description:The bone formation, osteogenesis, is a very complex process in which osteoblasts play a crucial role. The primary function of theses terminally differentiated mesenchymal stem cells is the secretion of fibrillary, dense, highly crosslinked type 1 collagen. Herein we show that the TENT5A gene, which mutations lead to osteogenesis imperfecta, is an active cytoplasmic poly(A) polymerase, which in osteoblast positively regulates the expression of collagen subunits and also other secreted proteins essential for proper bone formation. TENT5A substrates have shorter poly(A) tails in TENT5 KO, as revealed by direct RNA sequencing. In the case of Col1α1 and Col1α2 transcripts, the lack of cytoplasmic polyadenylation by TENT5 leads to drastic decree in the protein production but also aberrant structure. The increase in TENT5A expressing during osteoblasts differentiation positively correlates with the cytoplasmic extension of the poly(A) trials of mRNA targets. The first physiologically relevant regulation of collagen expression at the posttranscriptional level.

Project description:Sp7/Osterix is a master regulator of osteoblast specification. To identify the Sp7-mediated gene regulatory network in osteoblasts, we performed Sp7 ChIP-seq on primary mouse calvarial osteoblasts comparing the DNA binding profile with the transcriptional profile of Sp7-positive osteoblasts. Analysis of these identified a network of Sp7 regulated osteoblast targets and provides a new insight into the mode of Sp7 action in osteoblast. To further study for the Sp7 mode, we performed ChIP-seq for Sp1, Sp7, Dlx5, as a potential Sp7 partner identified in this study and mutated Sp7 which has mutations in the zinc finger domain, by using in vitro system with a pre-osteoblast mouse cell line, MC3T3E1.

To identify transcripts profile in Sp7 positive osteoblast, we performed RNA-seq on primary mouse calvarial cells obtained from Sp7-GFP reporter mice at P1. By hierarchical clustering using transcriptional profiles for chondrocytes and mouse embryonic fibroblasts (MEFs) together with the osteoblast data, we identified cell-type enriched gene expression signatures in osteoblasts, chondrocytes and MEFs. In conjunction with Sp7 ChIP-seq in osteoblast, we identified putative Sp7 targets which underlie the osteoblast regulatory program.

NF-kB regulator Bcl3 modulates bone density and turnover

Project description:B-cell lymphoma 3-encoded protein (Bcl3) is an intimate and context-specific regulator of the NF-kB family of transcription factors, a ubiquitous master regulator involved in many homeostatic and inflammatory processes. NF‑kB signalling determines the fates of bone-forming osteoblasts and bone-resorbing osteoclasts. Herein we show that Bcl3 is a negative regulator of NF‑kB via control of osteoblast and osteoclast function. Mice lacking Bcl3 (Bcl3−/−) have congenitally increased bone density, long bone dwarfism, increased biomechanical strength and altered bone turnover. Bcl3−/− osteoblasts and osteoclasts have accelerated differentiation and increased activity; whereas, rescue overexpression with a Bcl3 mimetic peptide inhibits differentiation. Early-differentiating Bcl3−/− osteoblasts have altered gene transcription favouring bone formation, including perturbation of the RANKL-OPG axis. In a model of osteoarthritic aberrant mineralisation Bcl3−/− mice exhibit decreased pathological osteophyte formation. Cumulatively, these findings identify Bcl3 as a viable target for controlling NF‑kB signalling in the treatment of skeletal pathologies.
