ABSTRACT: Use RNA-seq to obtain transcriptional signature of Tsc2 KO neonatal mouse dermal fibroblasts +/- sirolimus. The resulting mTORC1 transcriptional signature is tested in the TCGA database of bladder cancers with non-silent mutations in TSC1/2. Overall design: n = 3 WT vehicle, n = 3 KO vehicle, n = 3 WT sirolimus, n = 3 KO sirolimus

Increased mTORC1 signaling from TSC1/TSC2 inactivation is found in cancer and causes tuberous sclerosis complex (TSC). The role of mesenchymal-derived cells in TSC tumorigenesis was investigated through disruption of Tsc2 in craniofacial and limb bud mesenchymal progenitors. Tsc2cKOPrrx1-cre mice had shortened lifespans and extensive hamartomas containing abnormal tortuous, dilated vessels prominent in the forelimbs.
