Expression data from mouse lung from mice fed diets containing permethrin

ABSTRACT: Female Crlj:CD1(ICR) mice were fed diets containing 0 (control), 5000 and 10000 ppm permethrin and 2500 ppm isoniazid (positive control for tumor induction) for periods of 7 and 14 days. We used microarrays to evaluate gene expression profiling in mouse lung at the early phase of treatment with permethrin and isoniazid.

Genome-wide expression profiles and identification of upregulated genes in larval Culex quinquefasciatus during exposure to permethrin

Project description:The Southern house mosquito, Culex quinquefasciatus, is a vector of the causative agents of many diseases including West Nile Fever, St. Louis Encephalitis, and lymphatic filariasis. In order to manage the spread of these diseases, vector control efforts rely heavily on insecticides, including pyrethroids, namely permethrin. In our study we investigated the changes in the gene expression profiles of a highly-permethrin resistant strain of Cx. quinquefasciatus during constant exposure to permethrin at the LC-0, LC-50, and LC-70 rates, which killed 0, 50, and 70% of all larvae, respectively. Overall, we identified several genes that were up-regulated including detoxification genes such as cytochrome P450s as well as genes that were down-regulated.

Transcription profiling of rat cerebrocortical tissue following acute exposure to permethrin or deltamethrin.

Project description:Pyrethroids are neurotoxicants that disrupt nervous system function by interacting with a variety of membrane bound ion channels on neuronal plasma membranes. This study is designed to investigate the transcriptional events downstream of pyrethroid-induced disruption of nervous system excitability. Adult, male Long-Evans rats were orally dosed in vivo with a single dose of either permethrin (1, 10, or 100 mg/kg) or deltamethrin (0.3, 1, 3 mg/kg) at levels that produce only modest behaviroal effects in the whole animal (Wolansky et al. 2006). Transcriptional profiles were obtained from frontal cerebrocortical tissue 6 hours after acute exposure. The primary goals were 1) to identify dose-responsive biomarkers of effect for pyrethroids and 2) identify sensitive intracellular signaling or metabolic pathways sensitive to pyrethroid compounds. Experiment Overall Design: In total 60 samples were analyzed in the present study: vehicle control (n = 12), 1 mg/kg permethrin (n=8), 10 mg/kg permethrin (n = 8), 100 mg/kg permethrin (n = 8), 0.3 mg/kg deltamethrin (n = 8), 1 mg/kg deltamethrin (n = 8), 3 mg/kg deltamethrin (n = 8). Transcriptional profiles for each test subject were obtained independently, without sample pooling. Experiment Overall Design: The publication will include an Appendix Table comparing the variability of RMA values (see Sample VALUE columns) to that of GCOS values (see Sample .CHP files).

Epigenome-Wide Association Study for Pesticide (Permethrin and DEET) Induced DNA Methylation Epimutation Biomarkers for Specific Transgenerational Disease

Project description:Permethrin and N,N-diethyl-meta-toluamide (DEET) are the pesticides and insect repellent most commonly used by humans. These pesticides have been shown to promote the epigenetic transgenerational inheritance of disease in rats. The current study was designed as an epigenome-wide association study (EWAS) to identify potential sperm differential DNA methylation regions (DMRs) for specific transgenerational disease. Outbred Sprague Dawley gestating female rats (F0) were exposed to the pesticide combination including Permethrin and DEET. Their great-grand offspring (F3) were only transgenerationally exposed to pesticides. The transgenerational adult male rat sperm were collected from individuals with single and multiple diseases and compared to non-diseased animals to identify DMRs associated with transgenerational disease transmission. The exposure of gestating female rats to a permethrin and DEET pesticide combination promoted testis disease, prostate disease, kidney disease, and the presence of multiple disease in the subsequent great-grand offspring F3 generation. Interestingly, the majority of the disease specific sperm DMR associated genes were previously found to be linked to relevant disease specific genes.

A whole transcriptome approach to investigate the genes involved in permethrin resistance in the Southern house mosquito, Culex quinquefasciatus

Project description:Illumina HiSeq 2000 was conducted on the mRNA fraction of fourth instar Cx. quinquefasciatus from a highly permethrin resistant strain (HAmCqG8) and its parental low resistance strain (HAmCqG0). After aligning reads to the Johannesburg genome using the GTF annotations file from Ensembl Metazoa, we found that 367 genes were upregulated in the high resistant strain and 3982 were downregulated. The upregulated genes were associated with cytochrome P450s and proteases while the downregulated genes were mostly found in the “no annotation” category for the Structural Classification of Proteins classifications for Cx. quinquefasciatus. This study identified the upregulated genes in the highly resistant HAmCqG8 strain that may be involved in resistance to permethrin.
