Exogenous miRNA expression profiling in serum of pig and rat fed with different diets

ABSTRACT: We performed carefully designed and controlled animal feeding studies. We devided 9 adult rats in three group and each group fed with one type of diets in 28 days: rice, potatoes and chow. We fed piglets in triplicate either with cow milk or let them feed from sows milk for 4 weeks followed by 7 weeks of maize diet. Based on the small RNA sequencing data from 9 rat and 6 pig serum samples, we detected no transfer of plant miRNAs into rat blood, or bovine milk sequences into piglet blood.

Milk- and soy-based diets have differential effects on colitis and colon gene expression in the Il10-/- mouse

Project description:Milk and soy are reported to contain bioactive molecules with antibacterial and immunomodulatory actions, which may be beneficial to people with IBD. The aim of this study was to determine whether diets containing ruminant milk or soy solids reduce intestinal inflammation in Il10-/- mice. Male Il10-/- mice and C57BL/6J mice were fed diets containing 40% (w/w) sheep, goat, or cow whole milk powder, 40% (w/w) soy solids (NOW® Foods Soy Milk Powder, Instant), or one of two control diets (casein-free modified-AIN76A or standard AIN76A) from 4 to 11 weeks of age. Diets were based on AIN76A, which was included as an inter-experimental control for inflammation. For all diets except AIN76A, total protein, fat, carbohydrate and energy were kept as similar as possible. Weight and food intake were measured throughout the experiment (three times weekly), and intestinal tissue was taken for histopathology evaluation of inflammation and analysis of gene expression. Analysis of mouse weight and feed intake both showed a significant strain-diet interaction: Il10-/- mice fed the cow and goat milk diets ate less and gained less weight than all the other diet groups. This diet effect was not evident for the C57BL/6J mice. Il10-/- mice on the cow and goat milk diets had reduced colon histological injury scores relative to those on the other diets. Il10-/- mice on the cow and goat milk diets also had reduced expression of many immune/inflammatory-related genes and pathways.

Inclusion of Oat and Yeast Culture in Sow Gestational and Lactational Diets Alter Certain Immune and Antimicrobial Associated Proteins in Milk

Project description:probiotics and prebiotics to maternal diets is related to decreased incidence of diarrhea and greater weight gain during lactation. Our objective was to determine the impact of adding whole ground oat as a prebiotic alone or in combination with postbiotic yeast culture (YC) (Saccharomyces cerevisiae) to sow gestation and lactation rations on milk composition, piglet growth, and incidence of post weaning diarrhea (PWD). Diets: control (CON), CON + yeast culture (YC) [5g/kg], CON + oat (15% inclusion rate) (Oat) or CON+ YC [5g/kg] + Oat (15%) were fed during the last 30 days of gestation and throughout lactation (18-21 days). Shotgun proteome analysis of day 4 and 7 postpartum milk samples found 36 differentially abundant proteins (P-adj <0.1) in both Oat and YC supplemented sows relative to CON. Notable was increased expression of antimicrobial proteins, lactoferrin and chitinase. IgG in milk of Oat supplemented sows was lower than YC supplemented sows (p<0.05) but had greater E. coli-antigen reactivity. Piglet weights at birth were similar. At weaning YC + Oat piglets weighed less and gained less weight (p<0.05) postweaning than CON. The incidence of PWD was lowest in the YC and Oat groups compared to CON and YC+ Oat groups. These data suggest that Oat or YC culture supplementation alters milk immune and antimicrobial associated proteins that can impact piglets but may have negative effects on piglet growth when given in combination.

Project description:To further knowledge of piglet maturity, we have developed a microarray analysis to describe biological processes and to find candidate genes for key roles in piglet maturity. The objective was to identify which genes and biological processes are specifically involved in the difference between two extreme breeds: Large White (LW) and Meishan (MS). The LW breed is a selected breed known to show an increased rate of mortality at birth, while the MS breed presents more robust piglets at birth. MS and LW sows were inseminated with mixed semen (LW and MS) hence each litter was composed of pure fetuses (LW or MS) and crossed fetuses (LWMS from MS sows and MSLW from LW sows).

Effects of cold stress changes on subchondral bone and cartilage of tibial knee joint in rats and molecule related to hypothermia and arthritis

Project description:Forty eight-week-old male SD-rats were randomly divided into four groups: control group (4 weeks), hypothermia group (4 weeks), control group (8 weeks) and hypothermia group (8 weeks). The hypothermia group was fed at 4±2℃, and the normal group was fed at 25±2℃. After 4 weeks and 8 weeks, Micro-CT and saffron solid green staining were performed to observe the conditions of subchondral bone and cartilage. At 4 weeks, rat subchondral bone tissue was cut for second generation high-throughput sequencing, differential gene screening and gene enrichment analysis, and PCR validation.

Different Diets Change the Bovine Milk Extracellular Vesicle-miRNA Expression

Project description:Given that different diets could alter cow milk yield and composition, the effects of different feed formula on milk extracellular vesicle (EV) miRNAs were detected. Cow milk EVs contained various small RNAs, including miRNAs, snRNAs, tiRNAs, Cis-regulatory elements, and piRNAs. Two hundred and seventy-six known bos taurus miRNAs were identified by sequencing in bovine milk EVs. There were 13 immune-related miRNAs in the top 20 miRNAs in milk EVs. Nine differently expressed known miRNAs were detected in responding to different feed formulations. Cow milk EVs are abundant of small RNAs, especially miRNAs, which might be closely related to the development of maternal mammary gland and neonatal immune maturity.

Proteomic changes associated with maternal dietary low ω6:ω3 ratio in piglets supplemented with seaweed Part I: Serum proteomes

Project description:This study examines whether maternal low ω6:ω3 ratio diet and offspring seaweed (SW) supplementation can improve offspring immunity and performance by elucidating the effects on piglet serum proteome. A total of 16 sows were given either a standard (CR, 13:1) or low ω6:ω3 ratio diet (LR, 4:1) during pregnancy and lactation and their male weaned piglets were supplemented with SW powder (4 g/kg, SW) or not (CT) in a 21-day post-weaning (PW) diet. Four PW piglet groups were then identified based on dam and piglet treatment, namely CRCT, CRSW, LRCT, and LRSW (n = 10 each). Piglet serum collected at weaning and d21 PW were analyzed (n = 5 each) using TMT-based quantitative proteomics and validated by appropriate assays.

Transcription profiling by array of pig liver from neonatal pigs fed milk and soy formula

Project description:The majority of babies in the US are formula-fed instead of breast fed. There are major differences in the composition of formulas and breast milk and yet little is known about metabolic differences in babies as the result of feeding these very different diets and how that might affect development or disease risk in later life. One concern is that soy-based formulas might have adverse health effects in babies as a result of the presence of low levels of estrogenic phytochemicals genistein and daidzein which are normally present in soy beans. In the current study, we used a piglet model to look at this question. Piglets were either fed breast milk from the sow or were fed two different infant formulas (cow's milk-based or soy-based) from age 2 days to 21 days when pigs are normally weaned onto solid food. Blood glucose and lipids were measured. Formula-fed pigs were found to have lower cholesterol than breast fed piglets and in addition had larger stores of iron in their liver.Microarray analysis was carried out to see if changes in liver gene expression could explain these effects of formula feeding. It was found that overall gene expression profiles were influenced by formula feeding compared to breast fed neonates. Gender-independent and unique effects of formula influenced cholesterol and iron metabolism. Further, soy formula feeding in comparison to milk-based formula failed to reveal any estrogenic actions on hepatic gene expression in either male or female pigs. Piglets (female, male) were either fed breast milk from the sow or were fed two different infant formulas (cow's milk-based or soy-based) from age 2 days to 21 days when pigs are normally weaned onto solid food.
