Identification and characterization of miRNAs in temperature-dependent sex determination (TSD) of Chinese pond turtles (Mauremys reevesii)

ABSTRACT: In the present study, the eggs of Chinese pond turtles (Mauremys reevesii) were incubated at three temperatures (26℃, 29℃ and 32℃). During the thermosensitive period (TSP) of incubation, the adrenal-kidney-gonad (AKG) complexes were sampled, and a comprehensive investigation for miRNAs was performed using next-generation small RNA sequencing.

Transcriptional Control of Male-specific Pathway in Temperature-dependent Sex Determination

Project description:Temperature-dependent sex determination (TSD), wherein the incubation temperature of embryos dictates the sex of the offspring, is a phenomenon documented across diverse reptilian species. In the accompanying study, we demonstrated the thermal switch mechanism within thermosensitive steroidogenic cells (TSSCs) that triggers the male development pathway in turtles. However, the transcriptional regulatory mechanisms linking TSSCs' cold sensitivity with pre-Sertoli cell specification remain elusive. Herein, we elucidate the regulatory schema governing the male determination pathway, including Ca2+-induced aldosterone synthesis in TSSCs and subsequent aldosterone-driven transcriptional modulation in pre-supporting cells. Aldosterone-bound NR3C1, a master regulator of male determination pathway, activates DMRT1 gene expression, culminating in pre-Sertoli specification. Therefore, our findings suggest a conserved male-specific pathway in turtles.

Project description:Sex determination is the process by which and original bipotential gonad differentiate into either a testis or ovaries. While mammals and birds determine their sex solely by genetic clues (genetic sex determination, GSD), other vertebrates like the turtle Trachemys scripta are influenced by environmental factors, like temperature (environmental sex determination, ESD). In both cases an initially bipotential gonad develops into either testes or ovaries in response to GSD or ESD cues. In order to shed light into the differences and similarities between sex determination systems we performed single-cell RNA-seq on Trachemys scripta developing gonads during the sex determination window.

Project description:In our study, differential male nucleus events and development behaviors were revealed from the fertilized eggs in response to the sperm from males of genotypic sex determination (GSD) and temperature-dependent sex determination (TSD) in gibel carp. When the eggs of maternal fish were fertilized by the sperm from males of GSD, the fertilized egg encountered similar sexual reproduction events and behaviors. However, when the eggs of maternal fish were fertilized by the sperm from males of TSD, a typical process of gynogenesis was observed. To reveal the underlying molecular mechanism of differential sperm nucleus development behaviors in the fertilized eggs, iTRAQ-based quantitative semen proteomics were performed on three semen samples from three males of GSD and three semen samples from three males of TSD respectively.
