Genome-wide analysis of HeyA8 human ovarian cancer cells co-incubated with platelets under low attachment conditions

ABSTRACT: Analysis of gene expression changes in cancer cells upon interaction with platelets. The hypothesis tested in the present study was to test whether platelets can influence anoikis resistance and metastasis in ovarian cancer by influencing genes which are important for apoptosis, metastasis and proliferation. Results provide important informations of the response of HeyA8 human ovarian cancer cells upon platelet interaction and provide insights on platelet-regulated gene expression changes related to increased cancer cell survival under low attachment/anoikis conditions.

Project description:Interactions of cancer cells with the primary tumor microenvironment are important determinants of cancer progression towards metastasis but it is unknown whether additional prometastatic signals are provided during the intravascular transit to the site of metastasis. Here, we show that transient platelet-tumor cell interactions are sufficient to prime tumor cells for subsequent metastasis. Platelet-derived TGF-beta and direct platelet-tumor cell contacts synergistically activate the TGF-beta/Smad and NF-kappaB pathways in cancer cells, resulting in their transition to an invasive mesenchymal-like phenotype and enhanced metastasis in vivo. Inhibition of NF-kappaB signaling in cancer cells or ablation of TGF-beta1 expression solely in platelets protects against lung metastasis in vivo. Thus, cancer cells rely on platelet-derived signals outside of the primary tumor for efficient metastasis. MoEx-1_0-st: Five replicates of Ep5 tumor cells cultured alone, five replicates of tumor cells cultured in combination with platelets, and three replicates of tumor cells combined with platelets just prior to RNA isolation to control for the presence of platelet RNAs. MoGene-1_0-st: Three replicates of tumor cells cultured alone, three replicates of tumor cells cultured in combination with platelets, three replicates of tumor cells cultured with centrifuged platelets and three replicates of tumor cells cultured with supernate (releasate) from centrifuged platelets.

Platelets favor the outgrowth of established metastases

Project description:Despite abundant evidence demonstrating that platelets foster metastasis, anti- platelet agents have low therapeutic potential due to the risk of hemorrhages. In addition, whether platelets can regulate metastasis at the late stages of the disease remains unknown. In this study, we subjected syngeneic models of metastasis to various thrombocytopenic regimes to show that platelets provide a biphasic contribution to metastasis. While potent intravascular binding of platelets to tumor cells efficiently promotes metastasis, platelets further support the outgrowth of established metastases via immune suppression. Genetic depletion and pharmacological targeting of the platelet-specific receptor GPVI in humanized mouse models efficiently reduced the growth of established metastases, independently of active platelet binding to tumor cells in the bloodstream. Our study is the first to demonstrate therapeutic efficacy when targeting animals bearing growing metastases. It further identifies GPVI as the first molecular target whose inhibition can impair metastasis without inducing collateral hemostatic perturbations.

In this work we employed exon microarray technology to address platelet gene expression in metastatic non small cell lung cancer versus controls without cancer. We found that 197 of the 200 genes with the most significantly altered expression levels had their expression levels downregulated. Retrospective observational study. Platelet proteome is affected by cancer presence and therapy

Project description:Platelets play an important role in primary hemostasis, inflammation , (tumor) angiogenesis and cancer metastasis. Platelets are produced by megakaryocytes in the bone marrow and contain numerous bioactive proteins (e.g. growth factors, chemokine’s and proteases). These proteins inside platelets are either synthesized by the parental megakaryocytes or absorbed from the (micro)circulation. Therefore, it was postulated that changes in platelet protein content might indicate the presence or recurrence of a malignant tumor. We followed up the notion that both cancer and cancer therapy affect the platelet proteome. The aim of the current study was 1) to identify a pan-cancer proteome signature that could be used to discriminate patients with cancer from healthy individuals, and 2) to investigate the effect of cancer therapy on the platelet proteome. In this study, we included patients with cancer and healthy controls. Using global protein profiling by mass spectrometry-based proteomics we determined that the platelet proteome is affected by both cancer presence and antitumor therapy.
