ABSTRACT: The dataset contains data from 12 samples hybridized on Illumina HumanHT-12 array. -patient's primary lymphoma cells obtained from malignant ascites (P2-A1, P2-A2) -patient's non-malignant B-cells (CTRL1, CTRL2) -lymphoma cell line named UPF4D established from the patient's primary lymphoma cells after 15 and 75 days of in vitro cultivation (UPF4D-D15A, UPF4D-D15B, UPF4D-D75) -cells established by xenotransplantation of UPF4D cell line isolated ex vivo from infiltrated murine kidney (UPF4D-K) -cells established by xenotransplantation of UPF4D cell line isolated ex vivo from subcutaneously growing lymphoma (UPF4D-SC) -patient-derived xenograft (PDX) cells named VFN-D2 established by xenotransplantation of primary lymphoma cells into immunodeficient mice isolated ex vivo from the infiltrated murine kidneys (VFN-D2-K1, VFN-D2-K2) -patient-derived xenograft (PDX) cells named VFN-D2 established by xenotransplantation of primary lymphoma cells into immunodeficient mice isolated ex vivo from subcutaneously growing lymphoma (VFN-D2-SC). Samples were sorted using CD19-microbeads (Miltenyi). For xenotransplantation, NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ mice (referred to as NSG mice) were used.

Expression data of human clear cell renal cell carcinoma

Project description:Human tumor cell lines are important tools in tumor biological studies, here the authors report the establishment and characterization of 7 new ccRCC stable cell lines with complete clinical data. Gene expression and methylation were profiled with microarrays between the new cells and those had a finite in vitro life span, and the results prompt that genes such as SLC34A2 and VHL play key roles in the continuous in vitro growth and development of ccRCC. In order to find out what genetic features are related to the successful establishment of stable cell lines, early stage cells of 5 stable cell lines (UT14(P2), UT16(P2), UT33a(P3), UT48(P2), UT56(P1) ), named as P group and 5 finite cell lines (UT13a(P3), UT27(P3), UT37(P2), UT41(P2), UT54(P3) ) which could not be cultured more than 10 generations named as N group were chosen to do gene expression chip. Features such as age, gender, tumor size and Fuhrman nuclear grading were matched in these two groups.

De-differentiation and Proliferation of Artery Endothelial Cells Drive Coronary Collateral Development

Project description:We have earlier observed that upon myocardial infarction, pre-existing coronary arteries give rise to new collateral arteries via Artery Reassembly (AR) in neonatal mice, but not in adults. To understand this age dependent phenomenon, Gja5 (Cx40), an established artery endothelial cell marker, was used to lineage trace artery cells with TdTomato expression. For the neonatal mice, Tamoxifen was induced at postnatal (P)0, Myocardial Infarction (MI)/sham was performed at P2 and hearts were harvested at either P4 or P5. Whereas, for the adults, mice aged ~2months were used. Tamoxifen was induced at Day (D)0, MI/sham was performed at D2 and hearts were harvested at either D9 or D11. Ventricles from hearts were digested and FACS sorted for DAPI-, Ter199- and TdTomato+ cells.

Gene expression profile analysis of inducible melanotic and amelanotic melanoma in mice

Project description:We analyzed an inducible melanoma model in TiRP Ink4a/ArfF/F B10.D2 mice based on the conditional deletion in melanocytes of two tumor suppressor genes INK4a/Arf coupled to the expression of the oncogene H-rasG12V and a known tumor antigen (Hujibers et al. Cancer Res. 66:3278,2006; Soudja et al. Cancer Res. 70:3515-3525,2010). About 40% of these mice develop melanomas. Some tumors are heavily pigmented melanotic melanoma and grow slowly (hereafter referred to as “Mela”), but most of the induced tumors are nonpigmented amelanotic melanomas which are more aggressive (hereafter referred to as “Amela”). Gene expression profiles of the two types of tumors and of Amela-derived cell lines established in vitro were analyzed to identify the transcriptional signatures of these two types of tumors (including stromal components and infiltrating cells), as well as that intrinsic to Amela-tumor cells. 4 Amela and 4 Mela ex-vivo tumors were compared in a dye-swap experiment (8 chips). 4 Amela and 4 Mela ex-vivo tumors were compared to healthy skin (CTRL) (8 chips). 4 Amela ex-vivo tumors and Amela cell lines were compared in a dye-swap experiment (8 chips).
