Transcriptome profiling of SUM159 and derivatives with different metastasis capability

Project description:Triple negative breast cancer cell line SUM159 labeled with triple reporter (TR, TK-GFP-fLUC) injected into Nude mice via mammary fat pad and tail vein injection. Tumor progression is monitored by in vivo bioluminescent imaging. Tumors from mammary and lung were isolated and cultured. These derivatives cells are further tested for their capability to develop mammary primary tumor and metastasis in lung and bone in mice. The total RNA samples are extracted from the panel of SUM159 and derivatives and the transcriptome profiles are determined by microarray. The correlation of transcriptome of the cells to metastasis capability indicated candidate genes and regulatory pathways of metastasis of TNBC.

Global transcriptome analysis of paracancerous lung tissues from the B16 metastasis mice model under the influence of lactoferrin knockout

Project description:Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of cellular pathways. The goals of this study are using transcriptome profiling (RNA-seq) to evaluate the effects of lactoferrin deficiency on the global transcriptome of the lung microenvironment of the B16 metastasis mice model. Methods: 2×10^5 B16-F10 cells were injected into WT and lactoferrin kncoutout (Lf-/-) mice (each group has 10 eight-weeks-old male mice) through tail vein. At 3 weeks post injection,the paracancerous lung tissue were isolated (not including any metastatic colonies). RNAs were extracted by Trizol and sequenced by Solexa high-throughput sequencing service (Oebiotech, Shanghai, China). Data were extracted and normalized according to the manufacturer’s standard protocol.Each group has two mice paracancerous lung tissues be tested. Results: There are significant gene expression profile changes between WT and Lf-/- tumor-bearing mice. Conclusions: Our study describes the global transciptome changes of paracancerous lung tissues from the B16 metastasis mice model under the influence of lactoferrin kncokout.

Pulmonary melanoma metastasis

For information on the proper use of pre-publication data shared by the Wellcome Trust Sanger Institute (including details of any publication moratoria), please see http://www.sanger.ac.uk/datasharing/In this experiment, we have injected mice with fluorescent melanoma cells and then collected their lungs 1-3 days later and run them through a flow cytometer to separate the fluorescent cells (i.e., the melanoma cells) from the non-sluprescent cells (i.e., the lung cells) so we can sequence their transcriptomes to identify the transciptional profiles associated with pulmonary melanoma metastasis.

RNA-seq expression profiling of metastasis-associated endothelial cells from murine bone marrow

Project description:Breast tumour cells were found to remodel the murine bone marrow vascular microenvironment to support metastatic expansion. To gain insight into the host vasculature in response to tumor colonization, cells from the isogenic murine mammary tumour cell line 4T1.2 were intracardially injected into immunocompetent BALB/c mice. We analysed the transcriptional profiles of endothelial cells extracted from heavily infiltrated (Metastatic) bone marrow stroma and the adjacent tumor-free bone marrow stroma (Non-metastatic), as well as from healthy bones (Control). Methods: Immunocompetent BALB/c mice were intracardically injected with 4T1.2 tumor cells. The bones of these tumor-burdened mice were examined under fluorescence stereomicroscope to microdissect heavily-infiltrated (Metastatic) and the adjacent tumour-free (Non-metastatic) areas. Bones from healthy mice were used as controls. The dissected bone material from both burdened and control mice was processed to isolate EMCNhi/CD31hi endothelial cells using flow cytometry. RNA was extracted for 75bp paired-end RNA-seq profiling using an Illumina NextSeq 500 sequencer.

Expression data from breast cancer bone metastasis [EO771]

Project description:The goal of this study was to evaluate genes that are differentially expressed in the bone stroma when breast cancer metastasis are present. We focused our attention on bone stroma cells to understand how the dissemination and growth of tumor cells can impact on the bone environment. Moreover, we aimed to identify potential targets to inhibit the cross talk between cancer cells and tumor microenvironment in bone metastasis. EO771 breast cancer cell line were engineered to express luciferase and GFP, and injected in C57BL/6 mice by the intracardiac route to obtain bone metastasis. Tumor growth was monitored in-vivo by bioluminescence for 2 weeks. Endothelial cells and osteoblasts were isolated from the long bones of tumor-bearing and tumor-free mice to identify genes differentially expressed.
