Allergen-induced activation of NK cell genes represents an early life immune response in development of allergic asthma

ABSTRACT: Peripheral blood samples from children enrolled in the URECA cohort were collected at age 2. In short, at each URECA research center, within 16 hours of blood collection, mononuclear cells were separated and incubated (48 hours, 37C, 5% CO2) in the presence of German cockroach extract (10mg/mL; CR), dust mite extract (Dermatophagoides pteronyssinus, 10mg/mL; DM), tetanus toxoid (5mg/mL; TT), or medium alone (NS).

Project description:RATIONALE: Vaccines may make the body build an immune response to kill tumor cells. Drugs used in chemotherapy use different ways to stop tumor cells from dividing so they stop growing or die. Tetanus toxoid may make tumor cells more sensitive to chemotherapy and vaccine therapy. PURPOSE: Randomized phase II trial to study the effectiveness of chemotherapy and vaccine therapy with or without tetanus toxoid compared with chemotherapy alone in treating patients who have metastatic colorectal cancer.

Project description:Analysis of global gene expression profiles of flow cytometry-sorted, different pathogen-specific CD4+ T cell populations from the same peripheral blood mononuclear cells (PBMC), to identify molecular parameters that regulate differential susceptibilities of these CD4+ T cells to HIV infection. The results reveal distinct gene expression profiles between CMV-specific and tetanus toxoid/Candida-specific CD4+ T cells that involved selective upregulation of comprehensive innate antiviral Total RNA extracted from CFSE-low, flow-sorted pathogen-specific CD4+ T cells were analyzed for global gene expression. Changes in gene expression were compared between CMV to combined TT/Candida, or TT/CMV and Candida/CMV.

Project description:Plasmodium chabaudi infected mice were drug-treated on day 14 post-infection with an i.p. injection of CQ (10mg/kg) and pyrimethamine (10mg/kg) followed by CQ (6 mg/ml) and pyrimethamine (70 mg/ml) containing water for 5 days. Spleens were removed 12 weeks after completion of drug treatment. Spleens from drug-treated naive mice were used as controls.
