Unraveling the immunological roles of murine serum amyloid A3 in aortic immune cell subsets during atherosclerosis progression

Project description:Atherosclerosis is a growing concern in developed nations, necessitating the identification of therapeutic targets for advancing personalized medicine. Serum amyloid A3 (Saa3) has been linked to accelerated plaque progression by affecting cholesterol metabolism and modulation of inflammation. We hypothesize that knocking out Saa3 (Saa3-/-) could mitigate plaque development by regulating aortic immune cell subsetscompositions during atherosclerosis progression. Using a murine model, we induced atherosclerosis via a gain-of-function mutant PCSK9-encoding adeno-associated viral vector (AAVmPCSK9) in wild-type (WT) and Saa3-/- mice. Single-cell RNA sequencing revealed that Saa3-/- mice developed smaller plaques than WT mice, with and single-cell RNA sequencing revealed significant differences in aortic immune cell populations, particularly among aortic macrophages. Saa3-/-Trem2hi macrophages, characterized by high Gpnmb, Lpl, and Spp1 expressions, predominated over the typical resident foamy macrophages in WT mice.Saa3-/- compared to WT mice. SAA3 regulates cholesterol metabolism and inflammatory responses in foamy macrophages. Notably, aortic immune cells in atherosclerotic Saa3-/- mice also showed enhanced intercellular immune communication and increased signaling interactions, suggesting a shift towards an anti-inflammatory and tissue-repairing phenotype. Our study highlights the potential of Saa3 as a therapeutic target forkey modulator of aortic immune cells that impact atherosclerosis progression.

Microarray gene expression profiling of aortic genes of APOE-deficient mice receiving atherosclerosis treatment with the antioxidant vitamin E

Project description:Hypercholesterolemic APOE-deficient mice are a widely used experimental model of atherosclerosis and increased generation of reactive oxygen species (ROS) is a prominent feature of atherosclerosis development. To study the impact of ROS on atherogenesis, we treated APOE-deficient mice for 7 months with the antioxidant vitamin E (2000 IU/kg diet) and performed whole genome microarray gene expression profiling of aortic genes. Microarray gene expression profiling was performed of whole aortas isolated from vitamin E-treated APOE-deficient relative to untreated APOE-deficient mice with overt atherosclerosis, and nontransgenic B6 control mice. Microarray gene expression profiling revealed that vitamin E treatment prevented atherosclerosis-related gene expression changes of the aortic intima and media. Microarray gene expression profiling was performed of whole aortas isolated from APOE-deficient mice with atherosclerosis relative to vitamin E-treated APOE-deficient mice, and nontransgenic B6 control mice. Three study groups were analyzed, i.e. 8 months-old untreated APOE-deficient mice with overt atherosclerosis, age-matched APOE-deficient mice treated for 7 months with the antioxidant vitamin E (2000 IU/kd diet), and nontransgenic B6 control (C57BL/6J) mice. Two biological replicates were made of each group, and total RNA of three aortas was pooled for one gene chip. The study complements microarray study GSE19286.

Glutamine supplementation alleviated aortic atherosclerosis in mice model and in vitro

Project description:This study aimed to clarify the role of glutamine in atherosclerosis and its participating mechanism. Forty C57BL/6J mice were divided into wild control, ApoE-/- control, glutamine+ApoE-/-control, ApoE-/- high fat diet (HFD) and glutamine+ ApoE-/- HFD groups. The degree of atherosclerosis; plasma biochemistry and protein profiles, and western blotting and untargeted metabolomics of aortic tissue were detected at 18 weeks. An in vitro study was also performed. Glutamine treatment significantly decreased the degree of aortic atherosclerosis compared with that in the ApoE-/- HFD group (p=0.03). O-GlcNAc, IL-1β, IL-1α, GLUT1 and PKM2 in the ApoE-/- HFD group were significantly higher than those in the ApoE-/- control group (p < 0.05). These differences were reversed by glutamine treatment (p < 0.05), and aggravated by OGT overexpression in the in vitro study (p < 0.05). Multiomics showed that the ApoE-/- HFD group had higher levels of oxidative stress regulatory molecules (GUAD, XDH, PSB1 and PSB4), proinflammatory regulatory molecules (myristic acid, HMGB3, vinculin, DNJC2 and SPRC) and stress granules regulatory molecules (caprin-1 and DEAR) (p < 0.05). These differences were reversed by glutamine treatment (p < 0.05). We conclude that glutamine supplementation might alleviate atherosclerosis through downregulation of O-GlcNAc, glycolysis, oxidative stress and proinflammatory pathway.

An intersegmental single-cell profile reveals aortic heterogeneity [aorta]

Project description:The continuous inter-segmental aortic profile has been rarely reported at the single-cell level. To reveal aortic heterogeneity, we identified 15 cell subtypes from five continuous aortic segments by marker genes and functional definitions. In conclusion, our study provides a more comprehensive segmental single-cell transcriptome of the aorta at the physiological level, inspiring new ideas for diagnosis and intervention targets in the pathogenesis of aortic diseases. Furthermore, a novel construction system of VSMCs was proposed to replace the traditional classification.

Differential gene expression in human abdominal aortic aneurysm and atherosclerosis

Project description:The aim of this study was to assess the relative gene expression in human AAA and AOD. Genome-wide expression analysis of abdominal aortic aneurysm (AAA) and aortic occlusive disease (AOD) specimens obtained from 20 patients with small AAA (mean maximum aortic diameter=54.3±2.3 mm), 29 patients with large AAA (mean maximum aortic diameter=68.4±14.3 mm), and 9 AOD patients (mean maximum aortic diameter=19.6±2.6 mm). Relative aortic gene expression was compared with that of 10 control aortic specimen of organ donors.

Intermittent childhood hyperlipidaemia alters resident arterial macrophages and accelerates atherosclerosis

Project description:Mice deficient in the LDL receptor (LDLR KO mice) fed with high fat diet (HFD) feeding have been widely used as a model to mimic human atherosclerosis. RNA-seq using aortic macrophages to investigate the effects of aortic macrophages on the plaque size progression. LDLR KO mice, 6-8 weeks old, were used, and fed with either an alternative HFD or a regular HFD. After aorta macrophages were isolated, RNeasy Mini Kit (Qiagen) was used to isolate mRNA. Single end (50bp), unstranded, SMART-Seq v4 plus nextera (SMART-Seq v4 Ultra Low Input RNA Kit (Takara) ). The RNA-seq libraries were prepared with SMART-Seq v4 Ultra Low Input RNA Kit (Takara).
