Comparison of the RNA expression profiles of CD14+ monocytes from human peripheral blood with derived macrophages (MACs) and osteoclasts (OCs) obtained by exposure with M-CSF and M-CSF/RANKL, respectively, during 5 and 20 days

ABSTRACT: The expression profiles of RNA of human CD14+ monocytes were compared with derived macrophages (MACs) and osteoclats (OCs) following M-CSF and M-CSF/RANKL incubation for 5 and 20 days using the SurePrint G3 Human Gene Expression Array (Agilent Technologies, CA, USA). This platform allows the interrogation of >36,000 transcrits and variants per sample. The samples were hybridized in the array following the manufacturer’s instructions.

Gene expression of Dcir-/- osteoclasts induced by M-CSF and RANKL

Project description:Purpose: DCIR is an inhibitory type of C-type lectin receptor that regulates osteoclast (OC) formation and functions. This study aimed to find the genes of OCs that could be regulated through DCIR. Method: Bone marrow cells from WT and Dcir-/- mice were cultured in M-CSF for 2 days and the the cells were further cultured in M-CSF and RANKL. RNA profiles were generated by deep sequencing, in triplicate, using Illumina NovaSeq. Results: We detected xxx gense that were higher expression in Dcir-/- OCs, compared with WT OCs, with a fold change ≥1.5 and p value <0.05. Conclusion: Our data showed that Dcir deficiency in OCs changed the gene expression pattern, compared to WT OCs, which are induced by M-CSF and RANKL.

Project description:The methylation profiles of bisulfite(BS)- and oxidative-BS-modified DNA of human CD14+ monocytes were compared with derived macrophages (MACs) and osteoclasts (OCs) following M-CSF and M-CSF/RANKL incubation for 5 and 20 days, using the Infinium HumanMethylation450 BeadChips (Illumina, Inc., San Diego, CA,). This platform allows the interrogation of >485,000 methylation sites per sample at single-nucleotide resolution, and comprises an average of 17 CpG sites per gene in the 99% of RefSeq genes. 96% of CpG islands are covered, with additional coverage in CpG island shores and the regions flanking them. The samples were hybridized in the array following the manufacturer’s instructions.

Parenchymal border macrophages regulate CSF flow dynamics [leptomeningeal macs]

Project description:Microglial cells are the resident macrophages of the brain. Perivascular macrophages (PVM) are also myeloid resident cells that are located at the interface between blood, CSF and brain in the perivascular space. Due to their strategic location and their ability to sample CSF content, we hypothesized that PVM might play a role in CSF flow. Using different fluorescent tracers injected either into the CSF (influx) or the brain parenchyma (efflux) we assessed their spatio-temporal distribution and found that ablation (using clodronate liposomes) or dysfunction (using genetic tools) of PVM alters CSF dynamics and patterns. Using single cell RNA sequencing, we found that PBMs can be divided in two sub-populations, and that one population of PBMs could interact with vascular smooth muscle cells, which allow arterial pulsations and subsequently CSF flow. Interestingly, aging results in altered PVM phenotype, and reversing this phenotype using macrophage specific grown factors reversed some of aging-associated dysfunctions of CSF flow. In summary, our results identify a new role of PVM in CSF flow and open new avenues for therapeutical applications targeting PVM in neurodegenerative diseases such as Alzheimer’s disease.

Comparative characterization of osteoclasts derived from murine bone marrow macrophages and RAW 264.7 cells using quantitative proteomics

Project description:Osteoclasts (OCs) are bone-resorbing cells differentiated from macrophage/monocyte precursors in response to M-CSF and RANKL. In vitro models are principally based on primary bone marrow macrophages, but RAW 264.7 cells are frequently used because they are widely available, easy to culture, and more amenable to genetic manipulation than primary cells. Increasing evidence, however, has shown that the vastly different origins of these two cell types may have important effects on cell behavior. In particular, M-CSF is prerequisite for the differentiation of BMMs, by promoting survival and proliferation and priming the cells for RANKL induction. RAW 264.7 cells readily form OCs in the presence of RANKL, but M-CSF is not required. Based on these key differences, we sought to understand their functional implications and how it might affect osteoclast differentiation and related signaling pathways. Using a robust and high-throughput proteomics strategy, we quantified the global protein changes in OCs derived from bone marrow macrophages and RAW 264.7 cells at 1, 3, and 5 days of differentiation. Correlation analysis of the proteomes demonstrated low concordance between the two cell types (R2 ≈ 0.13). Bioinformatics analysis indicate that RANKL-dependent signaling was intact in RAW 264.7 cells, but biological processes known to be dependent on M-CSF were significantly different; including cell cycle control, cytoskeletal organization, and apoptosis. RAW 264.7 cells exhibited constitutive activation of Erk and Akt that was dependent on the activity of Abelson tyrosine kinase, and the timing of Erk and Akt activation was significantly different between BMMs and RAW 264.7 cells. Our findings provide the first evidence for major differences between BMMs and RAW 264.7 cells, indicating that careful consideration is needed when using the RAW 264.7 cell line when studying M-CSF-dependent signaling and functions.

Human FcγRIIa mediates osteoclastogenesis through a novel noncanonical pathway independent of RANKL and inflammatory cytokines

Project description:Under physiological conditions, osteoclasts (OCs) are generated from monocytic osteoprecursors under the stimulation of RANKL and M-CSF, requiring co-stimulatory signals from Fc receptor common γ chain (FcRγ) or DNAX-activated protein 12. It has been proposed that immune complexes (ICs) directly potentiate RANKL-mediated osteoclastogenesis by triggering FcγR-coupled Fcγ receptors (FcγRs), thereby linking IC accumulation and pathological osteolysis under various disease conditions. However, whether ICs possess pro-osteoclastogenic potential independent of RANKL is unknown. Here we demonstrate that IgG ICs alone can drive the differentiation of human blood monocytes into nonclassical OCs (NOCs) phenotypically and functionally distinguishable from RANKL-induced classical OCs (COCs). This novel noncanonical osteoclastogenesis pathway is triggered by full crosslinking of human FcγRIIa (hFcγRIIa), or co-ligation of hFcγRIIa and TLR4, and signals through the Src family kinase-STAT5 axis without inducing the expression of NFATc1, a master transcription factor for the previously described osteoclastogenesis pathways. Surprisingly, IgG ICs strongly overrule the generation of COCs driven by RANKL in vitro. More importantly, TRAP+ OCs found in the inflammatory joint tissues of hFcγRIIa-transgenic mice with collagen-induced arthritis bear the NFATc1- phenotype. Our results unmask the “double faces” of IgG ICs in health and disease, and suggest a novel and important pathway for ICs contributing to pathological bone erosion in inflammatory arthritis.
