Changes in gene expression upon DXO (Dom3Z) knockdown

ABSTRACT: Next generation sequencing was performed on polyA RNAs extracted from untreated cells and DXO knockdown cells (knockdown was achieved by expressing an inducible shRNA targeting DXO). Moreover, polyA RNAs from cells that contain the inducible shRNA targeting DXO but do not express this shRNA were also sequenced (n=1 for each condition). Overall design: Total RNA was extracted from cells and mRNAs were purified. cDNA sequencing libraries were generated and multiplexed. Paired-end 100bp sequencing was performed (Illumina HiSeq 4000). Reads were mapped to the human genome using Bowtie 2 and quantified in transcripts per million using RSEM.
