Project description:For determination of specificity of the MAbs, rDer p 21, thermally denatured rDer p 21 and rMBP-Der p 21 (0.3 mg/mL) were printed. the slides were incubated for 2 h 5 different MAbs (0.5 µg/mL for specificity analysis). Then the slides were incubated for 30 min goat anti-mouse IgG Fc Alexa Fluor® 647. Human serum albumin, rMBP, HRP and PBS were printed as negative controls.

Transcriptional signature of human mesoangioblasts from different fetal tissues.

Project description:Purpose: We performed comparative transcriptome analysis of mesoangioblasts (MABs) derived from different cardiovascular or skeletal muscle tissues to determine possible gene signatures underlying their specific differentation capacities and define future appropriate cell therapy applications. Methods: RNA was extracted from human fetal mesoangioblasts derived from Aorta, Atrium, Ventricle and Skeletal muscle (Ao, A, V and Sk) in proliferative conditions. Deep sequencing was used to determine quantitative gene expression between the different cell populations. Results: MABs derived from different tissues show differences in their transcriptome profile, despite their common ebryologic origin and isolation using common caracterization markers. The expression by Sk MABs of key cardiogenic genes (i.e. Isl1, TBX2) and the identification of active signaling pathways will serve for future skeletal to cardiac differentiation protocols. Conclusion: Cardiogenic Sk MABs may thus represent a easily accessible cell source for cardiac regeneration.

Differential regulation patterns of anti-CD20 mAbs in MCL

Project description:We investigated the differential regulation patterns of type I anti-CD20 monoclonal antibody (mAb) rituximab and type II obinutuzumab on a transcriptional level. Using a panel of MCL cell lines, we determined the effects of obinutuzumab and rituximab as monotherapies as well as in combination on cell viability and proliferation. Obinutuzumab induced a higher reduction in cell proliferation in each mantle cell lymphoma cell line than rituximab did. Results indicate a common pattern of expression changes after binding of anti-CD20 mAbs, but also reveal a significant difference between type I and type II treatment. Combination treatment resulted in a rituximab-like expression pattern. Many deregulated genes were associated with stress signalling, cell death, immune response and other functional clusters. Our analyses identified different and antibody-specific downstream expression patterns of obinutuzumab and rituximab, which may represent the molecular basis of the superior effect of obinutuzumab in comparison to rituximab.
