Developmental Dynamics of Skeletal Muscle Can Be Recapitulated In Vitro from Pig Embryonic Stem Cells

Project description:Understanding skeletal muscle development in pigs is required for fundamental developmental biology and its application in biomedical and agricultural industries. Here, we employed a directed myogenic differentiation approach to guide pig embryonic stem cells (ESCs) through presomitic mesoderm (PSM) and somite (SM) stages, ultimately generating myogenic progenitors (MPs) and mature muscle cells. The core signaling pathways, including WNT/β-catenin and BMP, along with several growth factors, were involved in sequential myogenic lineage induction in pigs. During differentiation, single-cell RNA sequencing revealed stage-specific transcriptional dynamics with three distinct ‘Myogenic states’, reflecting the in vivo developmental trajectory: PSM progenitors commitment into posterior PSM, posterior PSM transitioning into anterior PSM, and anterior PSM/SM giving rise to dermomyotome/muscle cells. These findings provide a comprehensive in vitro roadmap of pig skeletal muscle development, advancing developmental biology and biotechnology in livestock.

Gene expression profiling of the 414C2 iPSC line during hypertrophic chondrocyte induction

Project description:In order to better understand the mechanisms of chondrocyte hypertrophy, we induced hypertrophic chondrocytes from 414C2 iPSCs and analyzed their gene expression profile. This dataset includes the expression data obtained from iPSC-derived sclerotome and cartilage on days -1, 28, and 56 of induction. Day 56 samples were compared to day 28 samples to analyze pathways involved in chondrocyte hypertrophy.

The bone derived from scl-progenitors micromass forms bone marrow [scRNAseq]

Project description:Sclerotomal progenitors derived from pluripotent stem cells hold promises for modeling skeletal development and recreating embryonic bone microenvironment to restore hematopoiesis and immunity in ageing. Current strategy to derive mouse sclerotomal progenitors suffered from low differentiation efficiency and heterogenous cell progenies. Here, we developed a fast and efficient strategy to generate sclerotomal progenitors from murine embryonic stem cells (mESCs). Naïve mESCs were differentiated through epiblast-like cells (EpiLCs) and primitive streak, and the sclerotomal fate was directly induced from primitive streak by modulating BMP and SHH signaling, achiving an 85.8% differentiation efficiency. Moreover, the resulting progenitors showed similar global gene expression profiles as primary sclerotome, possessed strong osteochondral bipotential and could recapitulate key features of endochondral ossification upon micromass-mediated osteogenic induction, including bone marrow niche regeneration. To investigate the cellular component of the bone marrow derived from scl-progenitors micromass , we performed droplet-based scRNAseq (10X Genomics) of the differentiated bone tissue.

Genome-wide analysis of the regulatory function mediated by the small regulatory psm-mec RNA of methicillin-resistant Staphylococcus aureus

Project description:Several methicillin resistance (SCCmec) clusters characteristic of hospital-associated methicillin-resistant Staphylococcus aureus (MRSA) strains harbor the psm-mec locus. In addition to encoding the cytolysin, phenol-soluble modulin (PSM) mec, this locus has been attributed gene regulatory functions. Here we employed genome-wide transcriptional profiling to define the regulatory function of the psm-mec locus. The immune evasion factor protein A emerged as the primary conserved and strongly regulated target of psm-mec, an effect we show is mediated by the psm-mec RNA. Furthermore, the psm-mec locus exerted regulatory effects that were more moderate in extent and possibly mediated by the PSM-mec peptide. For example, expression of PSM-mec limited expression of mecA, thereby decreasing methicillin resistance. Our study shows that the psm-mec locus has a rare dual regulatory RNA and encoded cytolysin function, both with the potential to enhance MRSA virulence. Furthermore, our findings reveal a specific mechanism underscoring the recently emerging concept that S. aureus strains balance pronounced virulence and high expression of antibiotic resistance.

We performed single cell RNA sequencing (scRNA-seq) on sclerotome-derived fibroblasts from 52 hpf embryos to determine population heterogeneity and plasticity.

Project description:We sequenced RNA from embryonic mouse tissue directed to form sclerotome in culture. Using Crispr/Cas9, we generated a mouse line bearing a complete deletion of the PEAT, a LncRNA adjacent tp Pax1 and highly enriched in sclerotome. We perform RNA-seq on samples from 4 stages of maturation of the Dll1 lineage.
