Expression microarray analysis of mycobacteria-specific CD4 T cells

ABSTRACT: Live antigen-specific CD4 T cells are identified typically by tetramer staining or by staining of cytokines captured on the cell surface by bi-functional antibodies. We attempted identification of mycobacteria specific CD4 T cells using CD154 staining. In this method, splenocytes were stimulated with appropriate antigens in the presence of monensin and fluorochrome-conjugated anti-CD154 antibody. Antigen-specific cells will be stained positive for CD154. We performed microarrays with CD154 positive and negative CD4 T cells.

Effect of CD154:CD11b blockade on antigen-specific CD8 T cell response to murine Epstein-Barr virus infection

Project description:CD154 is a promising target for immunosuppression in transplantation, autoimmunity, and inflammatory diseases. We previously identified CD11b as a novel alternate receptor for CD154 during alloimmunity. However, the impact of specific CD154:CD11b blockade on immune responses to infection has not been well characterized. Here, we have shown that in contrast to its immunosuppressive effect on graft-specific CD8+ T cells, CD154:CD11b blockade unexpectedly improved both the quantity and quality of murine herpesvirus-68 (MHV)-specific CD8+ T cells as measured by an increase in tetramer-positive KLRG1loCD127hi memory precursor effector cells (MPECs). RNAseq of antigen-specific CD8+ T cells identified using an MHV68-specific MHC tetramer reveals specific pathways altered following cM7 treatment during MHV68 infection, including transcripts associated with T cell activation and survival. The differential effect of CD154:CD11b blockade on graft-vs. virus-specific CD8+ T cells was underpinned by differences in phospho-S6 downstream of mTORC1, however differential expression of key transcription factors Eomes and TCF-1 was dictated by the type of antigen stimulus. These data demonstrate that priming conditions play an important role in determining the outcome of immunotherapy and suggest that specific inhibition of CD154:CD11b interactions could be effective for suppressing alloimmune responses while maintaining protective immunity to minimize infectious complications following transplantation.

Single cell RNA and TCR profiling of tetramer sorted mouse mLN and IELs.

Project description:We utilized H2-I-Ab tetramers to isolate CD4 T cells from mice to characterize the functional characteristics of ileal intraepithelial T cells and their lymphoid counterparts in the corresponding draining lymph nodes (mLN). Non-CD4 T cells (B220+, CD11b+, CD11c+, F4/80+, CD8a+) were excluded before FACS-sorting of CD45+CD3+CD4+ tetramer-specific T cells discriminated by double-positive staining with dual-labeled tetramer fluorophores. We recovered tetramer+ and tetramer- IELs and mLN T cells, and profiled them by single-cell RNA and TCR sequencing.

Autoantigen-specific CD4+ T cells acquire an exhausted phenotype and persist in human antigen-specific autoimmune diseases

Project description:Pro-inflammatory autoantigen-specific CD4+ T helper (auto-Th) cells are central orchestrators of autoimmune diseases (AIDs). We aimed to characterize these cells in human AIDs with defined autoantigens by combining human leukocyte antigen (HLA)-tetramer-based and activation-based multidimensional ex vivo analyses. In aquaporin4-antibody-positive neuromyelitis optica spectrum disorder (AQP4-NMOSD) patients, auto-Th cells expressed CD154, but proliferative capacity and pro-inflammatory cytokines were strongly reduced. Instead, exhaustion-associated co-inhibitory receptors were expressed together with FOXP3, the canonical regulatory T cell (Treg) transcription factor. Auto-Th cells responded in vitro to checkpoint inhibition and provided potent B cell help. Cells with the same exhaustion-like (ThEx) phenotype were identified in soluble liver antigen (SLA)-antibody-autoimmune hepatitis and BP180-antibody-positive bullous pemphigoid, AIDs of the liver and skin, respectively. While originally described in cancer and chronic infection, our data point to T cell exhaustion as a common mechanism of adaptation to chronic (self-)stimulation across AID types and link exhausted CD4+ T cells to humoral autoimmune responses, with implications for therapeutic targeting.

Kruppel-like factor 2+ CD4 T cells avert microbiota induced intestinal inflammation [Fluidgm]

Project description:Commensal immune tolerance requires cells that actively sense and suppress inflammation. To understand how commensal tolerance works, mice colonized with recombinant Candida albicans expressing model MHC II peptide 2W1S were used to track CD4 cells with surrogate commensal specificity by tetramer staining and enrichement. Interestingly, half of the commensal CD4 cell lack expression of established T helper lineage defining transcription markers after intranuclear staining and flow cytometry. To more comprehensively investigate commensal cells, FACS sorted commensal CD4 cells with commensal specificity were analysed individually using single cell RNA sequencing.
