Project description:Astrogliosis is a hallmark of the response to brain ischemia, comprised of changes in gene expression and morphology. Hsp72 protects from cerebral ischemia, and although several mechanisms of protection have been investigated, effects on astrocyte activation are unknown. To identify potential mechanisms of protection, gene expression was assessed in mice subjected to middle cerebral artery (MCAO) or sham surgery, of either wildtype (WT) or Hsp72-overexpressing (Hsp72Tg) mice. After stroke, both genotypes exhibited genes related to cell death, stress response, and immune response. Furthermore, genes indicative of astrocyte activation, including cytoskeletal proteins and cytokines, were upregulated. To measure astrocyte activation after stroke, detailed histological and morphological analyses were performed in the cortical penumbra after stroke using unbiased stereology. Consistent with other reports, we observed a marked and persistent increase in glial fibrillary acidic protein (GFAP ) as soon as 3 hours after MCAO. In contrast, vimentin immunoreactivity appeared 12-24 hours after stroke, peaked at 72 hours, and returned to baseline after 30 days. Surprisingly, no change in overall astrocyte number was observed based on glutamine synthetase (GS) immunoreactivity. To determine if Hsp72Tg mice exhibited altered astrocyte activation compared to WT controls, morphological evaluation by fractal analysis was used. Overexpression of Hsp72 reduced astrocyte cell area, arbor area, and to a lesser extent fractal dimension, 72 hours following stroke. In conclusion, in vivo overexpression of Hsp72 alters gene expression following stroke, including genes involved in astrocyte activation, and decreases astrocyte activation acutely following MCAO. Thus, modulation of astrogliosis may be a neuroprotective mechanism exerted by Hsp72 after ischemia. A total of 10 samples were analyzed, with 5 of each genotype, wildtype (WT) and Hsp72-overexpressing (Hsp72Tg) mice. Of the 5 in each group, 3 received middle cerebral artery occlusion (MCAO) and 2 received a sham surgery. The sham samples serve as the controls for the MCAO samples in each genotype. All samples were taken from the ischemic or control hemisphere 24 hours after surgery.

Project description:Reactive astrocytes play critical roles after brain injury, but their precise function is not well defined, particularly in humans and nonhuman primates (NHPs). Here, we utilized single nuclei transcriptomics to characterize astrocytes after ischemic stroke in the primary visual cortex (V1) of the marmoset monkey. We observed nearly complete segregation between stroke and control astrocyte clusters. Screening the top 30 differentially expressed genes to identify candidates that might limit stroke recovery, we discovered that RTN4A/ NogoA, a neurite-outgrowth inhibitory protein previously associated specifically with oligodendrocytes but not astrocytes, was expressed in a majority of reactive astrocytes. Robust NogoA upregulation on reactive astrocytes following stroke was confirmed in both the marmoset and human brain, whereas rodents exhibited only a marginal change in NogoA expression following stroke. Further, in vivo and in vitro studies determined that NogoA mediated an anti-inflammatory response which likely contributes to limiting deeper infiltration of peripheral macrophages into the surviving parenchyma during the subacute post-stroke period. These findings are directly relevant to the development of NogoA-targeting therapies designed for clinical use shortly after ischemic stroke. In addition, our data have uncovered the complexity of astrocyte responses in primates, which highlights the importance of preclinical model selection for discovery research designed to identify novel therapeutics for brain injury.

Project description:N,N-Dimethyltryptamine (DMT) is a psychoactive molecule naturally present in the brains of mammals, including humans. Due to its neuroprotective effect in experimental stroke, DMT is under clinical evaluation, yet its mechanism of action remains poorly understood. Here we show that DMT protects against stroke by stabilizing the blood-brain barrier (BBB) and by mitigating neuroinflammation. In a transient middle cerebral artery occlusion (tMCAO) model of stroke in rats, treatment with DMT reduced infarct volume, and restored tight junction integrity and BBB function in vitro and in vivo. DMT also reduced cerebral edema, attenuated astrocyte dysfunction and shifted serum protein composition towards an anti-inflammatory, neuroprotective state. Finally, DMT suppressed the release of proinflammatory cytokines and chemokines in brain endothelial cells and peripheral immune cells, and reduced microglial activation, all in a sigma-1 receptor-dependent manner. Together, our findings demonstrate that the protective effect of DMT relies on its interaction with the vascular and immune systems, which can be harnessed to complement current stroke therapy.

Project description:Stroke involves in the interaction between central and peripheral immune systems. Skull bone marrows serve as reservoirs for immune cells in brain borders, and can rapidly respond to perturbations in the brain environment. Hence, targeting the skull bone marrow to modulate neuroimmune communications along the calvaria-meninges-brain axis would potentially improve stroke prognosis. Here, we successfully achieved cranial immunomodulation via ultraviolet (UV) irradiation of the interparietal region, which was characterized by rich marrow cavities and channels connecting the skull and meninges. Utilizing the recently-developed long-term clearing cranial window that ensured the integrity of skull, we discovered that the cranial photo-immunologic regulation (CPR) could promote cerebrovascular regeneration and aid in neurovascular repair post ischemic stroke. Single-cell transcriptome analysis revealed that meninge could be a crucial neuroimmune interface for ischemic stroke-induced immune responses. And, CPR could restore the stroke-induced alterations in cellular gene expression, especially meningeal B cells. Further we demonstrated that CPR could effectively alleviate the excessive suppression of meningeal B cell activation caused by ischemic stroke. This work opens avenues for immunoregulation through the skull-meninges-brain axis and provides valuable insights for immunomodulatory therapies in brain diseases.
