Epitope mapping of 5 monoclonal antibodies directed against meningococcal factor H binding protein by peptide microarray


ABSTRACT: The aim of the study was to determine the epitope targeted by 5 different HumAbs and the cross-reactivity to linear peptide epitopes of 12 different factor H binding protein (fHbp) variants. The HumAbs were diluted at 1:200 and incubated on a custom PepStar Peptide Microarray platform printed with 363 different peptides. Overall design: A panel of 363 peptides (15-mer overlapping peptides with an offset of 4 amino acids) covering the entire sequence of 12 different fHbp variants (1.1, 1.4, 1.13, 1.14, 1.15, 2.16, 2.19, 2.25, 2.77, 3.28, 3.45, NL10A) were printed in triplicate on a custom PepStar Peptide Microarray. Process-control peptide (GGSGGGSDYKDDDDK) and human IgG were distributed in each array copy as controls. HumAbs were profiled for anti-human IgG specific analysis of reactivity to the peptide epitopes

4CMenB is the first broad coverage vaccine for the prevention of invasive meningococcal disease caused by serogroup B strains.
