Astrocyte-enriched pan-brain aging transcriptome

Project description:Normal brain aging is marked by a cognitive decline, spurred by changes in cellular metabolism and homeostatic dysregulation, as well as modifications in synapses and neuronal connectivity. Astrocytes are well positioned as an effector of these changes, although how properties of astrocytes change with age remains unclear. Here, we address this question by profiling astrocytic gene expression from multiple brain regions of adult (4 month-old) and aged (2 years-old) mice. We isolated mRNA from transgenic mice where ribosomes within astrocytes were genetically tagged (GFAP-cre x RPL22-HA, astrocyte ribotag). We then used RNA sequencing to identify and quantify the astrocyte-enriched mRNA, analyzing 4 different brain regions: visual cortex, somatomotor cortex, hypothalamus, and cerebellum. Overall, we find that astrocyte expression of inflammatory and immune response factors increase with age, as well as changes in genes associated with metabolism, cholesterol synthesis, and synaptogenesis. Going forward, these data contribute to an understanding of astrocyte diversity and provide insight into the role of astrocytes in normal aging.

In this study, we analyzed the impact of ciliary deficient astrocytes on astrocytes’ intercellular communications and neuronal transcriptomes in the brain. Our analysis revealed a broad disturbance of astrocyte intercellular connectomes and profound changes in neuronal transcriptomic profiles induced by ciliary deficiency in astrocytes.

Injury-specific factors in the cerebrospinal fluid regulate astrocyte plasticity in the human brain

Project description:The glial environment determines the outcome of neurological disease progression, yet much of our knowledge still relies on preclinical animal studies, especially regarding astrocyte heterogeneity. In murine models of traumatic brain injury, beneficial functions of proliferating reactive astrocytes on disease outcome have been unraveled, but little is known if and when they are present in human brain pathology. Here, we examined a broad spectrum of pathologies with and without intracerebral hemorrhage and found a striking correlation between lesions involving blood-brain barrier rupture and astrocyte proliferation that was further corroborated in an assay probing for neural stem cell potential. Most importantly, proteomic analysis unraveled a crucial signaling pathway regulating this astrocyte plasticity with GALECTIN3 as a novel marker for proliferating astrocytes and the GALECTIN3-binding protein LGALS3BP as a functional hub mediating astrocyte proliferation and neurosphere formation. Taken together, this work identifies a therapeutically relevant astrocyte response and their molecular regulators in different pathologies affecting the human cerebral cortex.

Cross-talk between monocyte invasion and astrocyte proliferation regulates scarring in brain injury

Project description:Scar formation after brain injury is still ill understood. Here we examined the interplay between astrocyte proliferation and monocyte invasion. Using genetic mouse models to decrease or increase reactive astrocyte proliferation, we demonstrate inverse effects on monocyte numbers in the injury site, with e.g. reduced monocyte numbers when astrocyte proliferation was increased. Conversely, reducing monocyte invasion using CCR2-/- mice causes a strong increase in astrocyte proliferation, demonstrating an intriguing negative cross-regulation between these cell types at the vascular interface. CCR2-/- mice showed reduced scar formation with less extracellular matrix deposition, smaller lesion site and increased neuronal coverage. Surprisingly, also the GFAP+ scar area was significantly decreased despite increased astrocyte proliferation. Proteomic analysis at the peak of increased astrocyte proliferation revealed a decrease in ECM synthesizing enzymes in the injury sites of CCR2-/- mice already at this stage, highlighting how early key aspects of scar formation are initiated. Taken together we provide key novel insights into the cross-regulation of juxtavascular proliferating astrocytes and invading monocytes as a key mechanism of scar formation upon brain injury.

Identification of region-specific astrocyte subtypes at single cell resolution

Project description:Astrocytes, a major cell type found throughout the central nervous system, have general roles in the modulation of synapse formation and synaptic transmission, blood-brain-barrier formation and regulation of blood flow, as well as metabolic support of other brain resident cells. To investigate the true extent of astrocyte molecular diversity across forebrain regions, we used single cell RNA sequencing and generated a database containing ~2000 astrocytes. Our analysis identifies five transcriptomically distinct astrocyte subtypes in adult mouse cortex and hippocampus. Validation of our data in situ reveals distinct spatial positioning of defined subtypes, reflecting the distribution of morphologically and physiologically distinct astrocyte populations. Our findings are evidence for specialized astrocyte subtypes between and within brain regions.

Carcinoma-astrocyte gap junctions promote brain metastasis by cytosolic dsDNA response transfer

Project description:Brain metastasis represents a substantial source of morbidity and mortality in various cancers, and is characterized by high resistance to chemotherapy. Here we define the role of the most abundant cell type in the brain, the astrocyte, in brain metastasis. Cancer cells assemble of carcinoma-astrocyte gap junctions composed of connexin 43 (Cx43). Cx43 in cancer cells support brain metastatic colonization. We employ translating ribosome affinity purification (TRAP) to isolate translating mRNA from cancer cells in mixed asrtocyte co-cultures to determine the mechanism behind this Cx43-mediated brain metastatic growth. Once engaged with the astrocyte gap-junctional network, brain metastatic cancer cells employ these channels to transfer the cytosolic dsDNA response messenger cGAMP to astrocytes, activating the cGAS-STING pathway and production of inflammatory cytokines IFNα and TNFα. As paracrine signals, these factors activate the STAT1 and NF-κB pathways in brain metastatic cells, which support tumour growth and chemoresistance.
