Prepubertal vs pubertal lung gene expression during influenza in male mice

ABSTRACT: During the 1918 influenza pandemic, children experienced substantially lower mortality than adults, a striking but poorly understood finding. Whether this was due to enhanced resistance (reduced virus load) or better tolerance (reduced impact of infection) has not been defined. We found that prepubertal mice infected with H1N1 influenza virus also showed greater survival than infected pubertal mice, despite similar virus loads. To explore mechanisms for the survival differences, we compared gene expression profiles in lung tissue from mice of both age groups during the course of influenza pneumonia.

In order to validate the RNA-seq data, the expression patterns in the samples of lncRNAs and mRNA genes were confirmed by qRT-PCR. This research provides an expression profile of lncRNAs to further research on puberty.

Transcription profiling of bronchus from macaques infected with either the 1918 influenza virus or a humanized contemporary H1N1 influenza virus

Project description:The 1918 influenza pandemic was unusually severe, resulting in about 50 million deaths worldwide. A reconstructed version of the 1918 (H1N1) virus has been shown to also highly pathogenic in mice; however, the potential virulence and pathogenicity of the 1918 virus in nonhuman primates in unknown. In these studies, we demonstrate that the 1918 virus caused a highly pathogenic respiratory infection in a cynomolgus macaque model that culminated in acute respiratory distress and a fatal outcome. To characterize the global gene expression host response, oligonulceotide microarray analysis was performed on RNA isolated from the bronchus of macaques infected with either the 1918 virus or a humanized contemporary H1N1 influenza virus (A/Kawasaki/173/01). These experiments showed that infected animals mounted an immune response, characterized by dysregulation of the antiviral response, that was insufficient for protection, suggesting that atypical host innate immune responses may contribute to lethality.

Project description:Segment 3 of influenza A virus contains a second open reading frame accessed via robosomal frameshifting. The frameshift product, PA-Z, comprises the endonuclease domain of viral PA protein with C-terminal demain encoded by the X-ORF and functions to repress cellular gene expression. PA-X also modulates IAV virulence in a mouse infection model. The effect of PA-X deltions were examined using 2 separate mutant influenza viruses (1918-FS/1918-PCT1) and the results compared to wild-type 1918 influenza virus. Balb/c mice were infected and samples harvested at days 3, 5 and 8 days. Four animals/condition were used. Gene expression levels from infected animals were compared to mock animals.
