Study of gene expression of peripheral blood T cells in 25 healthy male donors of different age from 25 to 97 years

ABSTRACT: In this study we analyse a gene expression dataset, obtained by microarray hybridization, consisting of T lymphocytes from 25 healthy male volunteers with ages ranging over whole adult lifespan (25 to 97 years). Large groups of genes exhibiting non-monotonic expression patterns over time were observed, highlighting similarities in gene expression profiles of young and ‘‘successfully aged’’ individuals.

Project description:In this study we explored the effects of chronological and photoageing on the miRNome of human skin. To this end, miRNA expression was analysed in biopsies collected from sun-exposed (outer arm) and sun-protected (inner arm) skin from fair-skinned (phototype II/III) healthy female volunteers of two age groups: young, 18-25 years and aged, >70 years. Strict inclusion criteria were used for photoageing scoring and for chronological ageing. Microarray analysis revealed that chronological ageing had minor effect on the human skin miRNome. In contrast, photoageing had a robust impact on miRNAs, and a set of miRNAs differentially expressed between sun-protected and sun-exposed skin of the young and aged groups was identified. We have performed miRNome analysis (using microarray) on RNA isolated from sun-exposed and sun-protected skin of young and old females

Longitudinal profiling of the whole blood transcriptional response of human volunteers to repeated malaria challenge

Project description:We took a systems immunology approach to analyse the evolution of the human immune response to the first three infections of life in a re-challenge model of falciparum malaria. Ten volunteers were infected up to three times with Plasmodium falciparum in 4- to 8-month intervals. Remarkably, parasite densities and the dynamics of blood-stage infection were not altered over the course of three homologous infections. To track the development of immunity in real-time we used whole blood RNA-sequencing to analyse each volunteer’s transcriptional response throughout infection and convalescence (up to 9 time points per volunteer per infection).

Keywords: time course This study is a combination of 2 separate YF-17D vaccination trials. Trial 1 has 15 subjects, and Trial 2 has 10 subjects. The two trials occurred 1 year apart and used different lots of the vaccine. Enrolled volunteers were healthy, aged 18 to 45, and signed a written informed consent form. Potential volunteers were excluded from participating in the study if they were pregnant, or if they had been vaccinated previously with YF-17D. Blood samples were taken pre-vaccinaion to serve as a reference point for each individual, and post-vaccination samples were taken a days 1, 3, 7, and 21 after YF-17D vaccination. All subjects received YF-17D. Blood samples for microarray analysis were collected either in blue and black topped citrate buffered cell preparation tubes (CPT). Following PMBC isolation from CPT, 2 x 106 cells were lysed in 1 ml of TRIzol and stored at -80?C (Cat# 15596-026; Invitrogen Life Technologies). After all time points were collected for a subject, the samples were thawed, and the RNA isolation proceeded according to the manufacturer’s protocol.
