GNPS - Metabolites of psoriasis vs healthy skin_Pilot study

ABSTRACT: MS/MS data were collected from skin swab samples of one volunteer. Samples were collected from psoriasis sites located on elbows, arms, lower back and on the neck hairline. Nearby healthy skin was also sampled as a control.

Transcriptional Profile of Aging in Healthy Human Skin

Project description:Gene expression changes were assessed from the non sun-exposed skin of the lower back of 98 healthy males aged 19-86. We show that contrary to previous thought, genome wide transcriptional activity does not display an exclusively linear correlation with ageing, but rather, in human skin, undergoes a period of significant transient change between 30 and 45 years of age. The identified transient transcriptional changes suggest a period of heightened metabolic activity and cellular damage mediated primarily through the actions of TP53 (tumour protein 53) and TNF (tumour necrosis factor). We also identified a subgroup of the population characterised by increased expression of a large group of hair follicle genes that correlates strongly with a younger age of onset and increasing severity of androgenetic alopecia. Skin was collected from the lower back at the level of the belt, aproximately 5cm lateral to midline from healthy males, (defined as; non-smoking, no hospital admissions in the previous 5 years, no significant medical conditions or medications). Each sample was individually hybridised to an exon 1.0 ST array.

Healthy volunteers gene expression study of CD4 and NK lymphocytes

Project description:Sixteen volunteers, that were age, gender and BMI matched, were included in the study. Blood samples were taken one per each volunteer on three different days to also assess variability of gene expression markers in one individual. In all datasets variability between volunteers was higher than variability of measurables in samples collected from the same volunteer on different days. We have identified genes in individuals that are highly stable during one month period and are thus suitable as markers of individual disease/therapy progress. A list of genes with low overall variability is also available. Interestingly, data show that important components of immune response are down-regulated with BMI in T helper and NK cells. This paper also gives the first insight into the gene expression profile of T helper and NK cells in healthy population.

Skin regeneration is enabled in the absence of fibroblast inflammatory priming

Project description:Mammalian skin wounds heal by forming fibrotic scars. We report that reindeer antler velvet exhibits regenerative wound healing, whereas identical injury to back skin forms scar. This regenerative capacity was retained following ectopic transplantation of velvet to scar-forming sites. Single-cell mRNA/ATAC-Sequencing revealed that while uninjured velvet fibroblasts resembled human fetal fibroblasts, back skin fibroblasts were enriched in pro-inflammatory features resembling adult human fibroblasts. Injury elicited site-specific immune polarization; back skin fibroblasts amplified the immune response, whereas velvet fibroblasts adopted an immunosuppressive state leading to restrained myeloid maturation and hastened immune resolution ultimately enabling myofibroblast reversion to a regeneration-competent state. Finally, regeneration was blunted following application of back skin associated immunostimulatory signals or inhibition of pro-regenerative factors secreted exclusive to velvet fibroblasts. This study highlights a unique model to interrogate mechanisms underlying divergent healing outcomes and nominates both decoupling of stromal-immune crosstalk and reinforcement of pro-regenerative fibroblast programs to mitigate scar.
