Project description:Citrus cultivars are widely spread worldwide, and some of them only differ by specific mutations along the genome. It is difficult to distinguish them by traditional morphological identification. To accurately identify such similar cultivars, the subtle differences between them must be detected. In this study, UPLC-ESI-MS/MS-based widely targeted metabolomics analysis was conducted to study the chemical differences between two closely related citrus cultivars, Citrus reticulata 'DHP' and C. reticulata 'BZH'. Totally 352 metabolites including 11 terpenoids, 35 alkaloids, 80 phenolic acids, 25 coumarins, 7 lignans, 184 flavonoids and 10 other compounds were detected and identified; Among them, 15 metabolites are unique to DHP and 16 metabolites are unique to BZH. Hierarchical cluster analysis (HCA), principal component analysis (PCA), and orthogonal signal correction and partial least squares-discriminant analysis (OPLS-DA) can be used to clearly discriminate between DHP and BZH. 93 metabolites including 36 down-regulated and 57 up-regulated are significantly different in DHP and BZH. They are mainly involved in the biosynthesis of flavonoids, flavones, flavonols, and isoflavonoids. In addition, the relative content levels of flavonoids, alkaloids, and terpenoids are much higher in the peel of DHP than that of BZH, the presence of which may correlate with the quality difference of the peels. The results reported herein indicate that metabolite analysis based on UPLC-ESI-MS/MS is an effective means of identifying cultivars with different genotypes, especially those that cannot be distinguished based on traditional identification methods.

Project description:In the current study, the active flavonoids from Apocynum venetum and Apocynum hendersonii leaf were efficiently characterized using UPLC-ESI-MS/MS, and yielding the highest content of 15.35 mg/g (A. venetum) and 13.28 mg/g (A. hendersonii) respectively. The antioxidant assay in vitro showed that the isolated flavonoid ingredient groups exhibited free radical scavenging activities to DPPH, ABTS and linoleic acid. The antimicrobial assay revealed the isolated flavonoid ingredient from both A. venetum and A. hendorsonii have exerted anti-MRSA and anti-P. aeruginosa effect through disrupting cell integrity and declining ATP. In vivo assay demonstrated that these flavonoid ingredients effectively accelerated MRSA-infected and P. aeruginosa-infected Balb/c mice wound healing. In summary, these results showed that the characterized flavonoid ingredients exhibited great potential as natural antioxidant and antimicrobial agents, and shed light into future potential applications of Apocynum spp.

Project description:The properties of teak wood, such as natural durability and beautiful color, are closely associated with wood extractives. In order to further understand the performance differences between teak heartwood and sapwood, we analyzed the chemical components of extractives from 12 wood samples using an ultrahigh-performance liquid chromatography-electrospray ionization-tandem mass spectrometry (UPLC-ESI-MS/MS)-based metabolomics approach. In total, 691 metabolites were identified, and these were classified into 17 different categories. Clustering analysis and principal component analysis of metabolites showed that heartwood samples could be clearly separated from sapwood samples. Differential metabolite analysis revealed that the levels of primary metabolites, including carbohydrates, amino acids, lipids, and nucleotides, were significantly lower in the heartwood than in the sapwood. Conversely, many secondary metabolites, including flavonoids, phenylpropanoids, and quinones, had higher levels in the heartwood than in the sapwood. In addition, we detected 16 specifically expressed secondary metabolites in the heartwood, the presence of which may correlate with the durability and color of teak heartwood. Our study improves the understanding of differential metabolites between sapwood and heartwood of teak and provides a reference for the study of heartwood formation.

Project description:Chaenomeles species are used for both ornamental decoration and medicinal purposes. In order to have a better understanding of the flavonoid profile of Chaenomeles, the petals of four Chaenomeles species, including Chaenomeles japonica (RB), Chaenomeles speciose (ZP), Chaenomeles sinensis (GP), and Chaenomeles cathayensis (MY), were selected as experimental material. The total flavonoid content of GP was found to be the highest, followed by MY, ZP, and RB. In total, 179 flavonoid metabolites (including 49 flavonols, 46 flavonoids, 19 flavone C-glycosides, 17 procyanidins, 15 anthocyanins, 10 flavanols, 10 dihydroflavonoids, 6 isoflavones, 5 dihydroflavonols, and 2 chalcones) were identified by Ultra-Performance Liquid Chromatography-Electrospray Ionization-Tandem Mass Spectrometry. Screening of differential flavonoid metabolites showed that GP had higher levels of metabolites when compared with the other three Chaenomeles species. Annotation and enrichment analysis of flavonoid metabolites revealed that cyanidin 3,5-diglucoside and pelargonidin-3,5-diglucoside anthocyanins are likely responsible for the color differences of the four Chaenomeles petals. Additionally, a large number of flavonoids, flavonols, and isoflavones were enriched in the petals of GP. This study provides new insights into the development and utilization of Chaenomeles petals and provides a basis for future investigations into their utilization.

Project description:Flavonoids from plants are particularly important in our diet. Buckwheat is a special crop that is rich in flavonoids. In this study, four important buckwheat varieties, including one tartary buckwheat and three common buckwheat varieties, were selected as experimental materials. The total flavonoid content of leaves from red-flowered common buckwheat was the highest, followed by tartary buckwheat leaves. A total of 182 flavonoid metabolites (including 53 flavone, 37 flavonol, 32 flavone C-glycosides, 24 flavanone, 18 anthocyanins, 7 isoflavone, 6 flavonolignan, and 5 proanthocyanidins) were identified based on Ultra Performance Liquid Chromatography-Electrospray Ionization-Tandem Mass Spectrometry (UPLC-ESI-MS/MS) system. Through clustering analysis, principal component analysis (PCA), and orthogonal signal correction and partial least squares-discriminant analysis (OPLS-DA), different samples were clearly separated. Considerable differences were observed in the flavonoid metabolites between tartary buckwheat leaves and common buckwheat leaves, and both displayed unique metabolites with important biological functions. This study provides new insights into the differences of flavonoid metabolites between tartary buckwheat and common buckwheat leaves and provides theoretical basis for the sufficient utilization of buckwheat.

Project description:Endophytic fungi associated with medicinal plants are reported as potent producers of diverse classes of secondary metabolites. In the present study, an endophytic fungi, Aspergillus clavatonanicus strain MJ31, exhibiting significant antimicrobial activity was isolated from roots of Mirabilis jalapa L., was identified by sequencing three nuclear genes i.e. internal transcribed spacers ribosomal RNA (ITS rRNA), 28S ribosomal RNA (28S rRNA) and translation elongation factor 1- alpha (EF 1α). Ethyl acetate extract of strain MJ31displayed significant antimicrobial potential against Bacillus subtilis, followed by Micrococccus luteus and Staphylococcus aureus with minimum inhibitory concentrations (MIC) of 0.078, 0.156 and 0.312 mg/ml respectively. In addition, the strain was evaluated for its ability to synthesize bioactive compounds by the amplification of polyketide synthase (PKS) and non ribosomal peptide synthetase (NRPS) genes. Further, seven antibiotics (miconazole, ketoconazole, fluconazole, ampicillin, streptomycin, chloramphenicol, and rifampicin) were detected and quantified using UPLC-ESI-MS/MS. Additionally, thermal desorption-gas chromatography mass spectrometry (TD-GC-MS) analysis of strain MJ31 showed the presence of 28 volatile compounds. This is the first report on A. clavatonanicus as an endophyte obtained from M. jalapa. We conclude that A. clavatonanicus strain MJ31 has prolific antimicrobial potential against both plant and human pathogens and can be exploited for the discovery of new antimicrobial compounds and could be an alternate source for the production of secondary metabolites.

Project description:Diacyl glycerophospholipids (GPs) belong to the most abundant lipid species in living organisms and consist of a glycerol backbone with fatty acyl groups in sn-1 and sn-2 and a polar head group in the sn-3 position. Regioisomeric mixed diacyl GPs have the same fatty acyl composition but differ in their allocation to sn-1 or sn-2 of the glycerol unit. In-depth analysis of regioisomeric mixed diacyl GP species composed of fatty acyl moieties that are similar in length and degree of saturation typically requires either chemical derivatization or sophisticated analytical instrumentation, since these types of regioisomers are not well resolved under standard ultra-performance liquid chromatography (UPLC) conditions. Here, we introduce a simple and fast method for diacyl GP regioisomer analysis employing UPLC tandem mass spectrometry (MS/MS). This GP regioisomer analysis is based both on minor chromatographic retention time shifts and on major differences in relative abundances of the two fatty acyl anion fragments observed in MS/MS. To monitor these differences with optimal precision, MS/MS spectra are recorded continuously over the UPLC elution profile of the lipid species of interest. Quantification of relative abundances of the regioisomers was performed by algorithms that we have developed for this purpose. The method was applied to commercially available mixed diacyl GP standards and to total lipid extracts of Escherichia coli (E. coli) and bovine liver. To validate our results, we determined regioisomeric ratios of phosphatidylcholine (PC) standards using phospholipase A2-specific release of fatty acids from the sn-2 position of the glycerol backbone. Our results show that most analyzed mixed diacyl GPs of biological origin exhibit significantly higher regioisomeric purity than synthetic lipid standards. In summary, this method can be implemented in routine LC-MS/MS-based lipidomics workflows without the necessity for additional chemical additives, derivatizations, or instrumentation.
