Project description:The Aster-C protein (encoded by the Gramd1c gene) is an endoplasmic reticulum (ER) resident protein that has been reported to transport cholesterol from the plasma membrane to the ER . Although there is a clear role for the closely-related Aster-B protein in cholesterol transport and downstream esterification in the adrenal gland, the specific role for Aster-C in tissue cholesterol homeostasis is not well understood. Here, we have examined whole body cholesterol balance in mice globally lacking Aster-C under low or high dietary cholesterol conditions.transport and metabolism under divergent dietary cholesterol conditions. These results strongly suggest that Aster-C alone is not sufficient to control whole body cholesterol balance, but can modestly impact circulating cortisol and bile acid levels when dietary cholesterol is limited.

Project description:Cholesterol is one of the key molecules in mammals and the most striking examples of its deficiency are the inborn errors of cholesterol biosynthesis that manifest in severe whole body phenotypes. Liver, the principal site of cholesterol homeostasis, has rarely been investigated in these defects. We thus focused on the hepatocyte-specific deletion of lanosterol 14α-demethylase (CYP51) catalyzing the rate-limiting step in the post-squalene part of cholesterol synthesis. Liver-specific Cyp51 KO (LKO or K) and littermate control (LWT or W) mice (129/Pas (10%) × C57BL/6J (90%)) of both sexes (F and M) were investigated in the context of different nutritional availability of fat and cholesterol (standard laboratory diet without cholesterol (LFnC or L), high-fat diet without cholesterol (HFnC or H) and high-fat diet with cholesterol (HFC or C) due to the known sexual dimorphism in hepatic gene expression, where lipid metabolic pathways are among the most biased. 3 condition experimental design: 3 diets (LFnC or L, HFnC or H, HFC or C), 2 genotypes (LWT or W, LKO or K), 2 sexes (F, M), 3 biological replicates per condition, 36 mice altogether

Project description:Apolipoprotein E-knock out (apoE-KO) mouse is known as a model animal for atherosclerosis accompanied by spontaneous hypercholesterolemia. When apoE-KO mice were fed a chow supplemented with 1.25% cholesterol (high-Chol diet), cholesterol and bile acids were highly increased in the liver within a week. However, the amount of triacylglycerol (TG) in very low-density lipoprotein (VLDL), but not in the liver, was reduced by 78%. The epididymal adipose tissue was almost diminished in the long term. Cholesterol metabolism is tightly regulated by both cholesterol and its metabolites in the mammalian liver, but the regulatory mechanism of TG synthesis remains to be elucidated. To evaluate the impact of high-Chol diet for 1 week on gene expression in the liver of apoE-KO mice, DNA microarray analysis was performed with comparison to apoE-KO mice fed a normal chow. We found that mRNA expression related to lipid metabolism was suppressed by the high-Chol diet in the liver of apoE-KO mice, which includes beta-oxidation and glycerol-3-phosphate (G3P) pathway for TG synthesis. In particular, the mRNA and protein expression of lipin-1 and lipin-2 was markedly decreased. PGC-1?, which up-regulates the transcription of lipin-1, was also suppressed. Lipin is reported to function as a coactivator of PGC-1? and is an inducible amplifier of PPAR?, indicating that the suppression of genes involved in beta-oxidation could be induced by suppression of the lipins. These data using apoE-KO mice indicate that cholesterol and its metabolites are involved in regulating TG metabolism through a suppression of lipin-1 and lipin-2 in the liver. This research provides evidence for the mechanism of lipin expression in the liver. Apo E-KO mice at 10 weeks of age were fed normal chow (control) or high-Chol diet containing 1.25% cholesterol for 1 week. Three independent experiments were performed at each diet.

Project description:Postprandial dyslipidemia is a significant risk factor for atherosclerotic cardiovascular diseases. High-density lipoprotein (HDL)-driven reverse cholesterol transport (RCT) plays a crucial role in clearing postprandial lipids. In this study, we aimed to investigate the role of esculetin, a hypolipidemic substance, in regulating postprandial cholesterol metabolism. We measured postprandial cholesterol levels and conducted metabolomics and transcriptomics studies to understand the effect and underlying mechanism of esculetin on postprandial cholesterol metabolism in mice fed a lipid-rich meal. We found that esculetin significantly elevated the postprandial HDL cholesterol levels and induced alterations in postprandial serum metabolomics and hepatic transcriptomics which involved in the key steps of HDL-driven RCT including lipid metabolism, ABC transporters and bile acid biosynthesis and secretion. Additionally, the ratio and the phagocytic activity of an adipose tissue macrophage subset highly expressing CD36 and Tim4 increased following esculetin administration in mice. CCAAT enhancer-binding protein beta (C/EBPβ), a key transcription factor, was involved in the effect of esculetin on macrophages phagocytosis in ox-LDL-stimulated macrophages cells. Therefore, esculetin appears to elevate postprandial HDL levels and regulate postprandial cholesterol metabolism which are associated with the increased phagocytosis of adipose tissue macrophages and C/EBPβ.

Project description:<p>In recent years, the immunomodulatory function of adipose tissue has attracted much attention; however, the mechanism of steady-state regulation of the immune microenvironment in adipose tissue remains unclear. Catecholamines (CAs) activate adipocyte surface β3-AR to promote lipid metabolism, and cholesterol metabolism is bi-directionally immunoregulated. However, it remains unknown whether CAs and cholesterol metabolism interactions are a key pathway in the regulation of adipose tissue immune function. In this study, chickens were used as model animals, and dexamethasone and NDV vaccine were used to induce different immune states in chicks. qRT-PCR and metabolomics techniques were used to investigate the dynamic characteristics of lymphocytes, CAs and cholesterol metabolism in adipose tissues of chicks in different immune states, and to further characterize the miRNA regulatory mechanisms of cholesterol metabolism. The results indicate that B cells are present in chicken adipose tissue and respond positively to different immune states through up-regulation.Adipose tissue responds positively to immunosuppression and immune response through alterations in cholesterol and CAs metabolism, and differences in cholesterol and CAs metabolism are a key pathway by which dexamethasone suppresses the immune response in adipose tissue. Adipose tissue synthesizes bile acids and steroid derivatives, and secondary immunization immunity may be the stage of active changes in cholesterol metabolism derivatives in adipose tissue. A “lymphocyte-catecholamine-adipocyte-cholesterol-cholesterol metabolism derivative-lymphocyte” pathway exists in adipose tissue, and this pathway may be a potential mechanism for the homeostatic regulation of the immune microenvironment in chicken adipose tissue. In addition, adipose tissue miR-206 is actively involved in the process of dexamethasone affecting the immune response, and the miR-206/MSMO1 pathway is a potential mechanism for regulating cholesterol metabolism in adipose tissue. This study may provide new insights into the mechanisms of adipose tissue immunomodulation as well as new directions for the development of cholesterol derivatives for immunomodulation.</p>

Project description:This single-cell RNA-sequencing dataset was generated to investigate cellular and transcriptional changes following endothelial PLVAP loss in the adult mouse retina–choroid. Control and inducible endothelial-specific PLVAP knockout (PLVAPiECKO) mice were analyzed 10 days after PLVAP deletion. Single-cell transcriptomic profiling was used to characterize major retinal and choroidal cell populations and to examine PLVAP-loss-associated changes in choriocapillaris endothelial cells, retinal pigment epithelial cells, and photoreceptors. The dataset provides a cellular framework for understanding how disruption of PLVAP-dependent endothelial fenestration affects vascular–tissue exchange and metabolic homeostasis in the outer retina.

Project description:Hypercholesterolemia and hyperlipidemia are major contributors to the pathogenesis and progression of atherosclerosis and cardiovascular diseases (CVDs). Nuclear factor erythroid-derived 2-like 2 (NRF2) exerts antioxidant and anti-inflammatory effects while regulating glucose and lipid metabolism. However, the precise role and molecular mechanisms of NRF2 in hepatic cholesterol metabolism remain incompletely understood. Approach and Results: ApoE-knockout (ApoE-KO) mice were crossed with hepatocyte-specific Nrf2-knockout (Nrf2(H)-KO) mice to explore how NRF2 modulated cholesterol metabolism and the progression of atherosclerosis in vivo. Atherosclerotic lesion areas in Nrf2(H)-KO;ApoE-KO mice were significantly reduced compared with control counterparts (Nrf2-LoxP;ApoE-KO). Total cholesterol and LDL-cholesterol levels in plasma of Nrf2(H)-KO;ApoE-KO mice were significantly decreased, consistent with atherosclerotic phenotypes. Furthermore, hepatic triglyceride levels and cholesterol contents were increased in Nrf2(H)-KO;ApoE-KO mice. The hepatic transcriptomic analysis highlighted lipid metabolism pathway alterations in Nrf2(H)-KO;ApoE-KO mice, and carboxylesterase 1 (Ces1s) and lipocalin gene family member major urinary proteins (Mups) clusters markedly reduced expression in Nrf2(H)-KO;ApoE-KO livers. Liver proteomics analysis corroborated the transcriptomic findings. The current findings suggest that NRF2-modulated CES1s and MUPs may regulate cholesterol metabolism in the ApoE-KO model. Conclusions: Hepatocyte-specific Nrf2 deficiency significantly reduced plasma cholesterol levels, thereby attenuating atherosclerotic plaque formation and progression in the ApoE-KO background. This study established NRF2 in hepatocytes as a potential therapeutic target for controlling hypercholesterolemia and preventing atherosclerosis development.
