Project description:Spalax, an underground fossoral rodant is exposed to chronic hypoxic environments. Measurements of Spalax burrows indicated 7% oxygen and up to 15% CO2. In the laboratory Spalax can survive 3% oxygen up to 14 hours as compared to rats that survive less than 4 hours at these oxygen levels. we have proposed Spalax as a model species for the investigation of hypoxia tolerance. this research has implications for many biomedical phenomena involving ischemic disease, notably heart disease and cancer. In this work we have subjected Spalax to 3%, 6% and 10% O2 levels for varying lengths of time and have profiled expression patterns and differential expression in muscle and brain, two tissues with high energy requirements. For comparison with a terrestrial mammal we have profiled RNA from rats exposed to either 21% or 6% oxygen.

Project description:In the site of infection, the human fungal pathogen Aspergillus fumigatus faces dynamic changes in oxygen concentrations. The ability to survive in severely hypoxic environments during host invasion is one of important virulence traits of A. fumigatus. Therefore, investigation of hypoxia adaptation mechanisms of this pathogen may suggest potential targets for the development of antifungal therapies. Currently, an increasing number of reports has demonstrated that elevated production of intracellular reactive oxygen species (ROS) under low oxygen conditions plays a regulatory role in modulating cellular responses for adaptation to hypoxia. Our results confirmed raised amounts of intracellular ROS in A. fumigatus exposed to decreased oxygen levels. Moreover, nuclear accumulation of the oxidative stress response transcriptional factor AfYap1 was observed after two hours of low oxygen cultivation. For further analysis, we applied iodo-TMT labeling of redox-sensitive cysteine residues to identify proteins, which get reversibly oxidized and therefore regulated by hypoxic ROS after shifting oxygen content in the culture from 21% to 0.2%. For instance, proteins with important roles in maintaining redox balance and protein folding were modified after one hour of hypoxic cultivation. Redox proteomic investigation also revealed that a mitochondrial protein, designated as Coa6, was regulated by hypoxic ROS. A homologous protein in Saccharomyces cerevisiae was shown to be important for a respiratory chain complex IV assembly. Deletion of the gene encoding this protein in A. fumigatus resulted in complete absence of hypoxic growth, suggesting the significance of complex IV during adaptation of A. fumigatus to oxygen limiting conditions.

Spalax animals, captured in the field and held in captivity were subjected to 3 different levels of hypoxia. (1) 3% O2 for 6 hrs (2) 6% O2 for 6 hrs (3) 10% O2 for 44hrs (4) 21% O2 (normoxia). For comparison, Rats were subjected to 21% and 6% oxygen. Animals were euthanized according to approved protocols and RNA extracted from muscle and brain. Samples were labeled using the Agilent low input 2-color labeling kit and hybridized to a custom Spalax microarray (8x15K format). Results were normalized and analyzed using Limma from the R bioconductor package. Differentially expressed gens were identified as well as over-represented gene onotolgy defined functions.

Project description:To determine hypoxia mediated changes in whole blood, normal C57Bl/10 mice were gradually exposed to a chronic hypoxic environment, equivalent to an altitude of 6500m, for 2 weeks in vivo. Control, age-matched mice were maintained under normoxic, normobaric conditions by exposing them to ambient air in Philadelphia (c. 50 mts above sea level). Purpose: To determine the expression profile of genes differentially expressed in mouse whole blood upon exposure to normobaric hypoxia in vivo. Methods: The hypoxic group consisting of 6 C57/BL10 mice was exposed to normobaric hypoxia, in a specially designed and hermetically closed hypoxic chamber, using a gas mixing system Pegas 4000 MF (Columbus Instruments, Ohio, USA). The oxygen level was decreased from 21% to 8% over a period of 14 days. 6 control mice were kept at normal oxygen and pressure levels by exposing them to ambient air in Philadelphia (c. 50 mts above sea level). RNA from whole blood was isolated, processed and used for microarray-based expression profiling. Profiles were generated for genes differentially expressed at control versus normobaric hypoxia in whole blood using a false discovery rate (FDR) of 0%. The Profile was validated by real-time quantitative reverse transcription-polymerase chain reaction (qPCR) on 2 biologically independent samples, not used for generating the profile. Results: The transcriptomes associated with normobaric hypoxia in whole blood were identified. We found 4723 genes that were differentially expressed in normobaric hypoxic whole blood compared to control with a 0% FDR and a 2 fold cutoff. Conclusion: Transcriptome level differences exist in the whole blood of animals subjected to normobaric hypoxia. Our definition of the normobaric hypoxia blood transcriptome provides insight into the functioning and response to hypoxia in whole blood.

Project description:Cytochrome bd (CydAB), a high-affinity oxidase that performs terminal oxygen reduction in cellular respiration, is used by various bacteria to grow in hypoxic environments, form nonreplicating persister cells, or resist antimicrobial killing. It is thought that oxygen concentration solely determines the usage of CydAB in bacteria, with the tradeoff being that CydAB-containing respiratory chains generate less ATP than those containing low-affinity proton-pumping cytochrome oxidases, as CydAB does not pump protons when generating a proton motive force (PMF). However, CydAB has roles in acid, nitrosative and oxidative stress that suggest the existence of oxygen-independent CydAB usage. Here, we show that an obligate aerobe, the soil actinomycete Mycobacterium smegmatis, uses CydAB to normoxically maintain a homeostatic balance of the PMF, involving an oxygen-independent interaction with the classical hypoxia regulator (DosR). We show that overpotentials of the PMF apply feedback inhibition on the proton-pumping, low-affinity, cytochrome c oxidase supercomplex bcc-aa3, and that CydAB can immediately escape this inhibition and promote a PMF-responsive programme.
