Lipidomics analysis of outer membrane vesicles and elucidation of the ceramide phosphoinositol biosynthetic pathway in Bacteroides thetaiotaomicron

Project description:In this work, we characterized the lipid composition of membranes and OMV from Bacteroides thetaiotaomicron VPI-5482. LC-MS analysis indicate that OMV carry sphingolipids, glycerophospholipids and serine-dipeptide lipids. Sphingolipid species represent more than 50% of the total lipid content of OMV. The most abundant sphingolipids in OMV are ceramide phosphoethanolamine (CerPE) and ceramide phosphoinositol (CerPI). Bioinformatic analysis allowed the identification of the BT1522-1526 operon putatively involved in CerPI synthesis. Mutagenesis studies revealed BT1522-1526 are essential for synthesis of PI and CerPI, confirming the role of this operon in biosynthesis of CerPI. BT1522-1526 mutant strains lacking CerPI produced OMV that were indistinguishable from the wild-type strain, indicating that CerPI sphingolipid species are not involved in OMV biogenesis. Bacteroides sphingolipids are thought to modulate host-commensal interactions, and based on our data, we propose that OMV could act as long distance delivery vehicles for these molecules.

Analysis of a non-obese diabetes-experimental autoimmune encephalomyelitis (NOD-EAE)

Project description:The purpose of this study was to analyze and elucidate the mechanisms of non-obese diabetes-experimental autoimmune encephalomyelitis (NOD-EAE), an animal model of progressive multiple sclerosis (MS), and to compare the pathological features with those observed in human progressive MS. To achieve this, pathological analysis, flow cytometry analysis, immunohistochemical staining, and transcriptome analysis were performed at each pathological stage of the NOD-EAE mice to characterize each pathological stages in the lesion. In the chronic phase of the NOD-EAE mice, fibrosis and lymph follicle formation, characteristic of progressive human MS, were observed. We describe the pathological profile and transcriptome analysis of the NOD-EAE mice and verify that this model has similar features to those of human progressive MS. Our findings suggest that this model recapitulates lymph follicle formation, a disease hallmark of progressive MS, and fibrosis, a feature complicating the pathogenesis of MS in the chronic phase.

Project description:Investigation of whole genome gene expression level changes in a Bacteroides fragilis NCTC 9343 delta-ungD1 delta-ungD2 double mutant compared to the wild-type strain. Keywords: expression analysis A six chip study using total RNA recovered from three separate wild-type cultures of Bacteroides fragilis NCTC 9343 and three separate cultures of a double mutant strain, Bacteroides fragilis NCTC 9343 delta-ungD1 delta-ungD2, in which ungD1 (BF1706) and ungD2 (BF2848) are deleted. Each chip measures the expression level of 4,302 genes from Bacteroides fragilis NCTC 9343 and the associated plasmid pBF9343 with fourteen 24-mer probe pairs (PM/MM) per gene, with three-fold technical redundancy.
