Project description:The virtual transcriptome of G. intraradices (based on >430,000 reads) served as the basis for the design of an EST expresson array and for a number of analyses of gene expression in germinating spores, extraradical mycelium (ERM) and symbiotic root tissues. The G. intraradices EST expression array (4 x 72K) manufactured by Roche NimbleGen Systems Limited (Madison, WI) (http://www.nimblegen.com/products/exp/index.html) contained three independent, non identical, 60-mer probes per sequence. Included in the oligoarray were 22,402 G. intraradices sequences, 5785 random 60-mer control probes and labeling controls. We performed 12 hybridisations with samples from germinating spores (three biological replicates), extraradical mycelium (three biological replicates), symbiotic root tissues from Medicago (three biological replicates) and rice (1 replicate) as well as from microdissected arbuscule-colonized cortical cells of Medicago and rice (1 replicate each).

Methanol is an endogenous elicitor molecule upon senescence of detached rice leaves

Project description:During senescence of detached rice leaves, tryptophan (Trp) and Trp-derived secondary metabolites such as serotonin and 4-coumaroylserotonin accumulated in concert with methanol (MeOH) production. This senescence-induced MeOH induction was closely associated with levels of pectin methylesterase (PME)1 mRNA and PME enzyme activity. Exogenous challenge of detached rice leaves with 1% MeOH accelerated Trp and serotonin biosynthesis with induction of the corresponding genes. No other solvents including ethanol resulted in a Trp-inducing effect. This MeOH-induced Trp synthesis was positively regulated by abscisic acid but negatively regulated by cytokinin, suggesting hormonal involvement on the action of MeOH. Endogenous overproduction or suppression of MeOH either by PME1 overexpression or RNAi gene silencing revealed that PME1 overexpressing lines produced twofold higher Trp levels with elevated Trp biosynthetic gene expression, whereas RNAi lines showed twofold reduction in Trp level in healthy control rice leaves, suggesting that MeOH acts as an endogenous elicitor to enhance Trp biosynthesis. Among many transcription factors induced following MeOH treatment, the WRKY family showed significant induction patterns of which WRKY14 appeared to play a key regulatory role in MeOH-induced Trp and Trp-derived secondary metabolite biosynthesis.

Project description:Illumina HiSeq technology was used to generate mRNA profiles from Oidiodendron maius mycorrhizal roots compared to free-living mycelium . Mycorrhizal roots and control mycelium were harvested after 45 days and used for RNA extraction. Reads of 2X100bp were generated and aligned to Oidiodendron maius transcripts (http://genome.jgi-psf.org/Oidma1) using CLC Genomics Workbench 6. mRNA profiles from Oidiodendron maius mycorrhizal roots and free-living mycelium were generated by paired-end (2x100bp) Illumina HiSeq2000 sequencing. Three biological replicates were sequenced for mycorrhizal and mycelium samples.
