Project description:The timing and amplitude of reproductive effort are central life history variables for all organisms. In social insects, reproductive effort is collectively controlled at the colony level but little is known about the mechanisms that determine how much colonies invest in reproduction. As part of their female reproductive investment, honey bee colonies raise multiple new queens by feeding royal jelly to female larvae. Artificial selection for commercial royal jelly production in China has generated over the past 40 years a stock of royal jelly bees that raises an order of magnitude more queens and provisions each queen with >3x more royal jelly than unselected stock. Here we establish in a reciprocal cross-fostering experiment that this dramatic shift in social phenotype is due to changes in the nurse bees that care for the brood. We demonstrate higher electrophysiological responsiveness to brood pheromones in royal jelly bees than in unselected bees. Comparing the antennal proteome of unselected and royal jelly bees, we identify proteins involved in chemosensation and energy metabolism as candidates for the observed differences. We confirm several candidates, most prominently OBP16 and CSP4, with quantitative differences of corresponding mRNA levels and functional binding assays between the brood pheromones and the chemosensory proteins. Furthermore, we complement analyses of brood volatiles and electrophysiological recordings with behavioral attraction assays to confirm the presumed biological function of one newly discovered and two existing larval pheromones. Together, these findings help our understanding of pheromonal communication in honey bees and explain how sensory changes in nurse honey bees as alloparental caregivers have evolved in response to artificial selection, leading to a profound shift in colony-level resource allocation to sexual reproduction.

Project description:The ability of honey bees to evaluate differences in food type and value is crucial for colony success, but these assessments are made by individuals who bring food to the hive, eating little, if any, of it themselves. We tested the hypothesis that responses to food type (pollen or nectar) and value involve different subsets of brain regions, and genes responsive to food. mRNA in situ hybridization of c‐jun revealed that brain regions responsive to differences in food type were mostly different from regions responsive to differences in food value, except those dorsal and lateral to the mushroom body calyces, which responded to all three. Transcriptomic profiles of the mushroom bodies generated by RNA sequencing gave the following results: (1) responses to differences in food type or value included a subset of molecular pathways involved in the response to food reward; (2) genes responsive to food reward, food type and food value were enriched for (the Gene Ontology categories) mitochondrial and endoplasmic reticulum activity; (3) genes responsive to only food and food type were enriched for regulation of transcription and translation; and (4) genes responsive to only food and food value were enriched for regulation of neuronal signaling. These results reveal how activities necessary for colony survival are channeled through the reward system of individual honey bees.

Project description:The Varroa mite represents the main threat of honey bees (Apis mellifera). Bees from some colonies can limit the proliferation of this parasite by detecting and removing parasitized brood, such behavior is defined as Varroa sensitive Hygiene (VSH). This is an important issue for selecting colonies that can survive Varroa outbreaks. We therefore study the molecular meachnisms underlying this behavior by comparing the antennae transcriptomic profile of VSH and non-VSH bees. Those profiles were further compared to to the profiles of nurses and forager profiles involved in brood care and food collection, respectively.

Project description:Background The number of managed honey bee colonies has considerably decreased in many developed countries in recent years and the ectoparasitic mites are considered as major threats to honey bee colonies and health. However, their general biology remains poorly understood. Results We sequenced the genome and transcriptomes of Tropilaelaps mercedesae, the prevalent ectoparasitic mite infesting honey bees in Asia. The de novo assembled genome sequence (353 Mb) represents 53% of the estimated genome size because of the compression of repetitive sequences; nevertheless, we predicted 15,190 protein-coding genes which were well supported by the mite transcriptomes and proteomic dataes. Although amino acid substitutions have been accelerated within the conserved core genes in of two mites, T. mercedesae and Metaseiulus occidentalis, T. mercedesae has undergone the least gene family expansion and contraction between the seven arthropods we tested. The number of sensory system genes has been dramatically reduced; meanwhile, T. mercedesae may have evolved a specialized cuticle and water homeostasis mechanisms, as well as epigenetic control of gene expression for ploidy compensation between males and females., and water homeostasis. T. mercedesae contains all gene sets required to detoxify xenobiotics, enabling it to be miticide resistant. T. mercedesae is closely associated with a symbiotic bacteriuma (Rickettsiella grylli-like) and DWVdeformed wing virus (DWV), the most prevalent honey bee virus. The presence of DWV in both adult male and female mites was also confirmed by the proteomic analysis. Conclusions T. mercedesae has a very specialized life history and habitat as the ectoparasitic mite strictly dependsing on the honey bee inside the a stable colony. Thus, comparison of the genome and transcriptome sequences with those of a tick and free-living mites and tick has revealed the specific features of the genome shaped by interaction with the honey bee and colony environment. T. mercedesae, as well as Varroa destructor, genome and transcriptome sequences not only provide insights into the mite biology, but may also help to develop measures to control the most serious pests of the honey bee.

Project description:The samples used in this study originated from colonies of managed European honey bees (A. mellifera) that were at the epicenter of an AFB outbreak in 2020. The colonies were officially examined by the State Veterinary Administration of the Czech Republic. For our analyses, we targeted brood samples recognized as larvae before capping. Samples from two sites, namely, the villages Nevsova (denoted by N) and Tetcice (denoted by T), are included in this study.
We aimed to analyze samples of honey bee broods infected and affected by the pathogenic bacterium P. larvae. The colonies from which we collected samples had typical clinical signs of AFB. For our analysis, we selected larvae just before capping. We decided to analyze the samples using high-throughput proteomics, and to select an array of samples, it was necessary to confirm whether the samples were infected by P. larvae. Importantly, the presence/absence of P. larvae was confirmed using PCR before the proteomic analysis in the same individual sample.

Project description:There were important gaps in our knowledge of Israeli acute paralysis virus (IAPV), when IAPV was tightly linked to bee Colony Collapse Disorder (CCD), the mysterious disease that, starting in 2006-2007, has been wiping out honey bees in the US. To fill in these gaps we studied the molecular basis of transmission, pathogenesis, and genetic diversity of IAPV infection in honey bees. We investigated the impact of IAPV infection on colony losses and host transcriptional response to IAPV infections, and exploited the potential of RNAi-based strategies for treating viral diseases in honey bees. Our study clearly shows that IAPV has become established as a persistent infection and is highly prevalent in the honey bee population. The existence of both horizontal and vertical transmission pathways of the virus likely accounts for the high prevalence of IAPV in bees. While IAPV is probably not the only culprit responsible for CCD, its ability to cause increased mortality in honey bees is firmly demonstrated. The phenotypic differences in pathology among different strains of IAPV may be due to their high level of standing genetic variation. The JAK-STAT pathway, along with other signaling events such as mTOR and MAPK pathways, likely involves honey bees’ antiviral immune responses to the IAPV infection. The identification of IAPV-encoded putative suppressor of RNAi and evidence that silencing the RNAi suppressor led to a significant reduction in IAPV replication in infected bees illustrates the therapeutic potential of targeting viral suppressor protein to reduce virus replication. Our study gives direction for developing strategies to reduce colony losses due to viral diseases. Adult worker bees and brood were collected from colonies that were declining and identified with IAPV infections and its control with 6 replications per group.
