GNPS_GC_PDMS armpit samples pre-post bacterial spray treatment

ABSTRACT: A total of 63 volunteers were recruited and selected with a stronger than normal underarm malodor, as determined by a trained odor panel. The volunteers were using bacterial sprays and placebo sprays in the underarm to verify the effect on the underarm odor. Odor analysis was done using hedonic value measurement by a trained odor panel, as well as GC/MS analysis. Cleaned and degassed PDMS patches were placed on the underarm skin of the volunteer for 4h and secured in place with a large cotton bandage. After sampling, the bandage was removed, patches placed into 2 mL borosilicate vials using clean tweezers and capped with a crimp camp with silicone septum. The GC-MS analysis was carried out using the Agilent 7200 GC/QTOF equipped with robotic sampler system.

The extraction was carried out overnight at room temperature. After extraction, the sample was transferred into a 200 uL insert in a 2 mL vial and the vial capped with a crimp camp with silicone septum. The GC-MS analysis was carried out using the Agilent 7200 GC/QTOF equipped with robotic sampler system.

Serum miRNA profiles for gastric cancer diagnosis

Project description:Circulating microRNAs (miRNAs) in serum may serve as promising diagnostic biomarkers for patients with gastric cancer (GC). Using quantitative reverse transcription polymerase chain reaction (qRT-PCR) based Exiqon panel, we identified 58 differentially expressed miRNAs from 3 GC pool samples and 1 normal control (NC) pool in the initial screening phase. Identified miRNAs were further validated in the training (49 GC VS. 47 NCs) and validation phases (154 GC VS. 120 NCs) using qRT-PCR. Consequently, six serum miRNAs (miR-10b-5p, miR-132-3p, miR-185-5p, miR-195-5p, miR-20a-3p and miR-296-5p) were significantly overexpressed in GC compared with NCs. The areas under the receiver operating characteristic (ROC) curve of the six-miRNA panel were 0.764 and 0.702 for the training and validation phases, respectively. In conclusion, we identified a six-miRNA panel in serum for the detection of GC.

Project description:All samples were collected on cotton then placed into 2 mL borosilicate vials. The GC MS analysis was carried out using the Agilent 7200 GC QTOF Agilent Technologies Santa Clara CA. equipped with a robotic sampler system. The separation was conducted on an HP 5MS column (30 m x 0.25 mm x 0.25 micro m). The patch within the vial was heated for 25 min. at 200 C to desorb volatiles from the patch and 0.5 mL of headspace injected (injector temperature set at 250 C) into the instrument with a headspace syringe heated to 145 C. The GC protocol analysis included starting temperature 45 C min oven ramp (hold of 2 min), 15 C per min oven ramp to 300 C (hold of 3 min), and 50 C per min oven ramp to 320 C purge the column. The helium carrier gas was set to constant 1.2 mL per min flow and a spitless injection mode was applied. The purge flow to split vent rate was 50 ml per min at 1 min. The scanned m z range was 35-400 with the acquisition rate of 10 spectra per s. The empty vial blanks were interspersed with the samples to assess the background signal.

A novel urinary microRNA biomarker panel for detecting gastric cancer

Project description:Purpose: Gastric cancer (GC) is one of the most common causes of cancer deaths worldwide; however, reliable and non-invasive screening methods for GC are not established. Therefore, we conducted this study to develop a biomarker for GC detection, consisting of urinary microRNAs (miRNAs). Experimental Design: We matched 306 participants by age and sex (153 pairs consisting of patients with GC and healthy controls [HCs]), then randomly divided them across three groups: (1) the discovery cohort (4 pairs); (2) the training cohort (95 pairs); (3) the validation cohort (54 pairs). Moreover, 64 participants (32 pairs) with serum samples were also enrolled in the serum cohort. Result: There were 22 urinary miRNAs with significantly aberrant expressions between the two groups in the discovery cohort. Upon multivariate analysis of the training cohort, urinary expression levels of miR-6807-5p and miR-6856-5p were significantly independent biomarkers for diagnosis of GC, in addition to Helicobacter pylori (H. pylori) status. A diagnostic panel that combined the aberrant miRNAs and H. pylori status distinguished between HC and GC samples with an area under the curve (AUC) = 0.736. In the validation cohort, urinary miR-6807-5p and miR-6856-5p showed significantly higher expression levels in the GC group, and the combination biomarker panel of miR-6807-5p, miR-6856-5p, and H. pylori status also showed excellent performance (AUC = 0.885). In addition, serum levels of miR-6807-5p and miR-6856-5p were significantly higher in the GC group. Conclusion: This novel biomarker panel enables early and non-invasive detection of GC.
