Developmental pyrethroid exposure and age influence phenotypes in a Chd8 haploinsufficient autism mouse model

Project description:Hundreds of genes have been associated with autism spectrum disorder (ASD), including loss-of-function mutations in chromodomain helicase DNA binding protein 8 (Chd8). Environmental factors also are implicated in autism risk and have the potential to exacerbate phenotypes in genetically sensitized backgrounds. Here we investigate transcriptional and behavioral phenotypes in a Chd8 haploinsufficient (Chd8V986*/+) mouse line exposed to the pesticide deltamethrin (DM) from conception to postnatal day 22. Vehicle-exposed Chd8V986*/+ mice displayed ASD-associated phenotypes, including anxiety-like behavior and altered sociability, replicating a previous study with this mouse line. A core set of genes was altered in Chd8V986*/+ mice at multiple ages, including Usp11, Wars2, Crlf2, and Eglf6, and proximity ligation data indicated direct binding of CHD8 to the 5’ region of these genes. Moreover, a neurodegenerative transcriptional phenotype was apparent in 12 and 18 month old Chd8V986*/+ mice. Following DM exposure, the mutant mice displayed an exacerbated phenotype in the elevated plus maze, and genes associated with vascular endothelial cells were downregulated in the cerebral cortex of older Chd8V986*/+ animals. Our study reveals a gene x environment interaction with a Chd8 haploinsufficient mouse line and points to the importance of investigating phenotypes in ASD animal models across the lifespan.

Project description:The rat model was previously reported as whole-body exposure to traffic-related PM1 pollutant (PM1 group), high-efficiency particulate air-filtered gaseous pollutant (GAS group), and clean air control (CTL group) for 3 and 6 months. We performed lung function and histological examinations as well as quantitative proteomics analysis to unravel the structural and molecular changes in rat lungs upon sub-chronic and chronic exposures of PM1. Our study illustrated earlier lung injury prior to lung function decline and the underlying pathomechanism associated with traffic-related PM1-induced lung injury.

Whole brain gene expression study of dominant, subordinate and brooding females of the African cichlid fish A. burtoni

Project description:Comparison of social dominance phenotypes induced in the cichlid A. burtoni females with the goal of comparing to other gene expression profiles of social dominance Used platform GPL928 In many species, under varying ecological conditions, social interactions among individuals result in the formation dominance hierarchies. Despite general similarities, there are substantial differences across species, populations, environments, life stages, sexes, and individuals. Understanding the proximate mechanisms of this variation is an important step toward understanding the evolution of social behavior. However, physiological changes associated with dominance such as gonadal maturation and somatic growth, often complicate efforts to identify the specific underlying mechanisms. We demonstrate complementary analysis tools to allow a comparative approach to high-throughput expression profiling that allow us to both test a priori hypotheses and generate new hypotheses about the mechanisms and evolution of social dominance. Using experimental manipulation to produce female dominance hierarchies in the cichlid A. burtoni, heralded as a genomic model of social dominance, we generate gene lists, and assess molecular gene modules. We demonstrate a general pattern of “masculinization” of the female neural gene expression profile and compare expression biases between male and female dominance hierarchies. Using a threshold-free approach to identify correlation throughout gene ranked lists, we query previously published datasets from maternal behavior, alternative reproductive tactics, cooperative breeding and sex-role reversal to describe correlations among neural gene expression profiles. These complementary approaches capitalize on the high-throughput gene expression profiling from similar behavioral phenotypes in order to address the mechanism associated with social dominance behavioral phenotypes.

Persistent transcriptomic alterations in medial prefrontal cortex after prenatal exposure to maternal inflammation

Project description:Infection during pregnancy is strongly implicated as an environmental risk factor for autism spectrum disorder (ASD), with activation of the maternal immune system (MIA) identified in the causative pathway. Maternal exposure to viral and bacterial mimics at midgestation leads to the development of core ASD behaviors in rodent offspring. However, the fundamental pathogenic mechanisms coupling MIA to persistent changes in brain function and behavior are incompletely understood.The medial prefrontal cortex (mPFC) is a major locus of pathology in ASD and highly associated with behaviors dysregulated by MIA. Transcriptomic profiling of the mPFC from adult MIA and control offspring provides insight into molecular pathways persistently disrupted by prenatal exposure to maternal inflammation.

These results suggest the development of a long-term neuronal engram underlying the persistence of LID .

Stress modulates Ahi1-dependent nuclear localization of Ten-Eleven Translocation Protein 2

Project description:Three Ten-Eleven Translocation (Tet) enzymes are epigenetic regulators of gene expression that promote 5-hydroxymethylcytosine (5hmC) modification of genes. Here, we show that the loss of Tet2 can induce depression-like phenotypes in mice. Paradoxically, using the paradigms of chronic stress, such as chronic mild stress (CMS) and chronic social defeat stress (CSDS), we found that depressive behaviors were associated with increased Tet2 expression but decreased global 5hmC level in hippocampus. We examined the genome-wide 5hmC profile in the hippocampus of Tet2 knockout mice and identified 651 dynamically hydroxymethylated regions, 31 of which overlapped with known depression-associated loci. We further showed that chronic stress could induce the abnormal nuclear translocation of Tet2 protein from cytosol. Through Tet2 immunoprecipitation and mass spectrum analyses, we identified a cellular trafficking protein, Abelson helper integration site-1 (Ahi1), which could interact with Tet2 protein. Ahi1 knockout or knockdown caused the accumulation of Tet2 in cytosol. The reduction of Ahi1 protein under chronic stress explained the abnormal Ahi1-dependent nuclear translocation of Tet2. These findings together provide the evidence for a critical role of modulating Tet2 nuclear translocation in regulating stress response.
