Project description:We used 16S V3/V4 region amplification to evaluate the composition of bacteria species in mouse fecal pellets. Fecel pellets were collected from young-adult (12 weeks old) wild type C57Bl/6 mice and aged (72 weeks old) wild type C57Bl/6 mice after 21 days of vehicle or antibiotics treatment (to induce gut microbiota depletion). In one sequencing round, we sequenced a total of 12 different fecal samples (3 young control, 3 aged control, 3 young depleted gut microbiota (ABX) and 3 aged depleted gut microbiota (ABX)). Amplicons were indexed using the Nextera XT Index Kit and pooled into a library for Illumina sequencing.

Exploiting Mouse Models to Understand Female Hypersensitivity to Cocaine

Project description:Male mice, female mice, ovariectomized female mice given placebo pellets, and ovariectomized female mice given extradiol pellets were treated with experimenter-administered saline or cocaine for 7 days, and behaviorally monitored. After a 28 day withdrawal period, RNA samples were prepared from nucleus accumbens and subjected to RNA-Seq, followed by differential expression analyses using Cuffdiff, Limma and DESeq2. Selected transcripts were further investigated using quantitative polymerase chain reaction.

Effects of semaglutide on the gut microbiota of high-fat diet-fed mice lacking GLP-1Rs in T cells

Project description:Gut intraepithelial lymphocytes (IELs) are one of the few immune cell populations in the body that expresses glucagon-like 1 receptors (GLP-1R). To test the potential effects of GLP-1 on the gut microbiota through the gut IEL GLP-1R, we performed 16s rRNA seq on the DNA isolated from the fecal pellet of Lck-Cre; Glp1rfl/fl mice (Glp1rTcell-/-) or controls (Glp1rTcell+/+) fed a high-fat diet (HFD) for 12 weeks followed by 1 week of HFD plus semaglutide (10 ug/kg) or vehicle treatment. Fecal pellets from a group of age-matched, sex-matched control mice were included as a chow control group.

Liver tissue from hormone treated C57BL/6J mice versus pool C57BL/6J mice of chow diet Liver

Project description:The purpose of this experiment was to determine the expression traits in animals of inbred strain C57BL/6J, treated with gonadal hormones. (N=40, 20 males and 20 females). Liver tissue from the hormone treated group consists of 40 female and male C57BL/6J mice fed a chow diet containing 4% fat (Ralston-Purina Co., St. Louis, MO) until 8 weeks of age and then were gonadectomized at 8 weeks of life, implanted with hormone pellets at 12 weeks, and sacrificed at 14 weeks. Male and female mice of the hormone treated groups received subcutaneous implants of either 0.5 mg estradiol (E2) pellet (plasma yield of 300 picogram/ml) or a 5 mg dihydrotestosterone (DHT) pellet (plasma yield of 1-2 nanogram/ml), designed to release over 21 days, (Innovative Research of America, 17B-estradiol: Cat. No. E-121 0.5mg/pellet, 5a-dihydrotestosterone: Cat. No. A-16). Control mice were treated with the placebo pellet (Innovative Research of America, placebo: Cat. No. C-111). At 14 weeks mice were sacrificed, after a 12-hour fast, Liver tissue were dissected and flash frozen in LN2 and stored at -80Â°C. Gene profiling changes in mouse bone marrow derived macrophages upon SUMOylation inhibition

Project description:SUMOylation, a posttranslational modification, regulates protein function by covalent attachment of small ubiquitin-like modifier (SUMO) proteins to a lysine (Lys) residue on target proteins. Here we use ML-792, a selective SUMO-activating enzyme (SAE) inhibitor, to inhibit global SUMOylation in macrophages derived from bone marrow of C57BL/6 mice. Mouse bone marrow cells isolated from 6–10 weeks C57BL/6 mice were cultured in IMDM medium supplemented with 10% (vol/vol) FBS and 10 ng/mL of macrophage colony-stimulating factor (M-CSF). After 6–8 days of differentiation, the cells were treated with vehicle or ML-792 at 0.5 μM for 24 h. Total RNA was purified using miRNeasy RNA isolation kit (Qiagen) then Ribo-Zero RNA-seq was performed.
