MetaboLights MTBLS561 - GNPS Discovery of urinary biomarkers to discriminate between exogenous and semi-endogenous thiouracil in cattle (Cow samples)

ABSTRACT: In the European Union, the use of thyreostats for animal fattening purposes has been banned because of their carcinogenic and teratogenic effects, whereby any residues in derived food products may hold a risk for human health. In this regard, potential abuse is monitored by targeted analysis of thyreostats with the application of a zero-tolerance policy. However, such a strategy is no longer adequate because of the semi-endogenous origin of some thyreostats, including thiouracil (TU). Additionally, the use of recommended TU concentration levels (10 or 30 µg/l) is also not fully conclusive. Therefore, this study aimed at defining urinary metabolite markers by which the true origin of detected TU could be ascertained. To this end, an in vivo study was carried out in which 11 calves were subjected to either a control treatment, a rapeseed-enriched diet (known to induce semi-endogenous formation) or a TU treatment. Urine samples (n = 165) were assessed through metabolic fingerprinting, employing ultra-high performance liquid-chromatography and Q-Exactive Orbitrap mass spectrometry. Urinary fingerprints were interpreted by multivariate data-analysis.

Project description:Lipid Data Analyzer (LDA; http://genome.tugraz.at/lda2) provides automated and reliable annotation of lipid species and their molecular structures in high-throughput data from chromatography-coupled tandem mass spectrometry using embedded rule sets. Using various low- and high-resolution mass spectrometry instruments with several collision energies, the method's platform independence is proved. The software's reliability, flexibility, and ability to identify novel lipid molecular species may now render current state-of-the-art lipid libraries obsolete. LDA identified several novel lipid molecular species and novel regio-isomeric species; we consider a lipid species as “novel” if it is neither present in LIPID MAPS Structure Database, ChEBI, CyberLipid, HMDB, nor YMDB. Where evidence relies on fragments of very small intensity, high resolution HCD mode was used for final characterization. These data are presented here (CID mode data can be found in MTBLS397 - both biological experiments for Orbitrap Velos CID). Moreover, we present in this study data showing the LDA's ability to distinguish regio-isomeric standards under certain chromatographic conditions. In lipidomics, overlaps of species from isobars from other subclasses/adducts frequently occur. In this experiment, isobars from PC and PE, and from LPC and LPE are mixed in one sample. The LC-method was made worse, so that these species generated overlapping peaks. In this experiment, the LDA's propensity to detect and seperate such species was tested. The biological data was used to fine-tune the developed decision rule sets. In particular, the high numbers of isobaric species and structural isomers contained in these samples helped in the detection of decisive differences for many isobaric subclasses/adducts.
