Project description:Heat stress is a very complex physiological process. At present, there are some reports about physiological function and pathological changed on chicken heat stress. We use DNA microarrays to detail the global programme of gene expression underlying heat stress and identified distinct classes of differentially expressed (DE) genes during this process. Chicken were randomly divided into two groups to study the effects of heat stress on gene expression of brain, liver and leg muscle. Twenty-four birds (12 male and 12 female) were allocated to heat stress or post-heat stress groupM-oM-<M-^H40M-BM-11M-BM-0C,3hM-oM-<M-^I and control or pre-heat stress groupM-oM-<M-^H28M-BM-11M-BM-0CM-oM-<M-^I.

Methylome patterns of cattle adaptation to heat stress

Project description:Heat stress has a detrimental impact on cattle health, welfare and productivity by affecting gene expression, metabolism and immune response, but little is known on the epigenetic mechanisms mediating the effect of temperatureat the cellular and organism level. In this study, we investigated genome-wide DNA methylation in blood samples collected from 5 bulls of the heat stress resilient Nellore breed and 5 bulls of the Angus that are more heat stress susceptible, exposed to the sun and high temperature-high humidity during the summer season of the Brazilian South-East region. The methylomes were analyzed during and after the exposure by Reduced Representation Bisulfite Sequencing, which provided genome-wide single-base resolution methylation profiles. Significant methylation changes between stressful and recovery periods were observed in 822 genes. Among these, 352 were only seen in Angus, 367 were specific to Nellore, and 103 showed significant changes in methylation patterns in both breeds. KEGG and Gene Ontology (GO) enrichment analyses showed that responses were breed-specific. Interestingly, in Nellore significant genes and pathways were mainly involved in stress responses and cellular defense and were undermethylated during heat stress. Whereas in Angus the response was less focused. These preliminary results suggest that heat challenge induces changes in methylation patterns in specific loci, which should be further scrutinized to assess their role in heat tolerance.

Project description:We investigated the root growth of several knockout mutants of heat shock protein family genes and found that heat stress response was compromised in these mutants compared to wild type plants. It suggested that heat shock protein genes including heat shock protein genes including HSP17s, HSP23s, HSP101, and HSFA2 proteins are deployed upon exposure to Cs for plant stress tolerance. Our study provided novel insights into the molecular events occurring in Cs-stressed plants.

Project description:<p>Heat stress is an important issue in dairy cattle feeding management affecting summer health and economic efficiency. In recent years, global climate change has led to an increase in atmospheric CO2 content and average daily temperature, making heat stress a major challenge in dairy farming. This experiment combined 16S rDNA sequencing, metagenomic sequencing and metabolomic analysis. In this experiment, 10 cows each of growing heifers, heifers and lactating cows were selected for sample collection in April and August. Ruminal fluid was collected and filtered through gauze, which was immediately transferred to liquid nitrogen prior to macrogenomic, 16S rDNA sequencing and metabolomic analyses.</p>

Transcriptome responses to heat stress in hypothalamus of a meat-type chicken

Project description:By using a 44k chicken Agilent microarray, we systematically analyzed the chicken hypothalamus transcriptome response to thermal stress. Twelve hypothalamus samples were chosen from three groups (four samples per group) to be used in chicken genome microarray to examine differential gene expression.We compared the expression profiles between each pairs of the three groups using the microarray data. Totally, 2474 genes were found to be differentially expressed in the three comparisons with p＜0.05 and fold change (FC) higher than 1.5 and the genes were mainly involved in self-regulation and compensation required to maintain homeostasis, including heat shock protein family, enzyme and the hormone, neurotransmitter, cell-cell signaling, metabolism and cytokines. The transcripts of heat shock protein including Hsp 40 and Hsp 90 were significantly changed respond to thermal stress and genes involved in regulation of cell morphogenesis were significantly upregulated in heat stressed group with comparison to control and temperature recovery group. Additionally, the down-regulated genes in both heat stress and temperature recovery groups compared to control group were enriched in muscle organ development, striated muscle tissue development, cardiac muscle tissue development and muscle tissue development, which indicates that muscle development was inhibited during and in short-term after heat treatment. Most of genes dysregulated in heat stress group were found to be recovered in temperature recovery group, which confirmed their roles they could play in coping with heat stress. The present study provides a broader understanding of molecular mechanisms underlying the stress response in chicken and discovery of novel genes that are regulated in a thermal stress specific manner.
