Intestinal β-galactosidase affects host health by regulating flavonoid metabolism

Project description:Untargeted metabolomic analysis was performed to identify the principal aglycone generated by the engineered Escherichia coli Nissle 1917 strain expressing β-galactosidase (EcN-bgal). Culture supernatants from EcN-bgal and the vector control strain were collected and subjected to LC–MS-based metabolomic profiling. Comparative analysis revealed substantial alterations in the metabolite composition of the EcN-bgal supernatant, enabling the identification of β-galactosidase-dependent metabolites and the characterization of the major aglycone produced by the engineered strain.

Genome-scale CRISPRi screen identifies pcnB repression conferring overproduction of free fatty acids in Escherichia coli

Project description:In this study, we carried out genome-scale CRISPRi modulation combining fluorescence-activated cell sorting (FACS) and next-generation sequencing (NGS) to identify genetic determinants for FFAs overproduction in Escherichia coli. The pcnBi (AP) strain that repressed the expression of pcnB produced 2992 mg l FFAs, which was 4.0-fold of the CF (A) strain. To analyze the underlying mechanism of FFAs overproduction, the engineered strain pcnBi and the control strain CF were applied to the transcriptomic and proteomic analyses.

A Metal-Dependant Formate Dehydrogenase Advances the Formate Bio-economy

Project description:In this manuscript a multi-subunit metal dependent FDH is functionally expressed in a strain of E. coli engineered to assimilate all energy and biomass from formate. The replacement of the previously described FDH with this new, faster FDH allows for improved growth parameters rivaling natural formatotrophs for the first time. This new, robsulty growing formatotrophic E. coli was engineered to produce mevalonate and achieved the highest reported titer for any bioproduct from formate in fed batch mode. Finally the authors explored bioproducion from electrochemically-derived and lignin-derived formate, demonstrating the first example of a bioproduct produced using liginin-derived formate as a feedstock.

Corynebacterium glutamicum as a powerful host for the production of 5-aminovalerate from glucose fermentation

Project description:5-aminovalerate (5AVA), L-lysine derived compound, represents a potential building block for the production of the bio-plastic nylon-5. Escherichia coli has been engineered for the production of 5AVA, but Corynebacterium glutamicum has never been engineered for the production of 5AVA, but, a lot of work was done in the last decades to optimize the production of the precursor L-lysine and more recently cadaverine. 5AVA added to the growth medium hardly affected growth rate of C. glutamicum, since, a half-inhibitory concentration of 1.1 M 5AVA was determined. While in E. coli, 5AVA production was engineered by using the DavBA pathway from Pseudomonas putida, here a pathway based on the route described in P. aeruginosa was established. C. glutamicum wild type was converted into a 5AVA producing strain by heterologous expression of L-lysine decarboxylase (LdcC), putrescine transaminase (PatA) and γ-aminobutyraldehyde dehydrogenase (PatD) genes from E. coli. 5AVA production was improved by using a strain previously engineered for high L-lysine production, by de-repressing phosphoenolpyruvate phosphotransferase system (PTS) and glycolysis and by avoiding formation of the by-product L-lactate. 5AVA accumulation by this strain was increased to 44.9 mM, representing a yield of 202 mmol mol-1 glucose, which is about three times higher than the highest yield achieved in E. coli for the production of 5AVA from glucose fermentation.

Gene expression by E. coli during the production of styrene and when exposed to externally added styrene

Project description:Transcriptomic studies of high-cell density fermentations of E. coli producing chemicals of relevance for biotechnology are limited. The aim of this study was to perform fermentations of an E. coli strain that had been engineered to produce styrene and examine the transcriptomic response. This was compared to an identical strain of E. coli that had the styrene pathway inactivated by the introduction of amino acid mutations in PAL2 and fdc1, the two enzymes required for styrene production. A further comparison was done by exposing E. coli cells to styrene added externally to the fermentation broth.

Project description:Investigation of whole genome gene expression level changes in a Escherichia coli MG1655 K-12 ?fnr mutant, compared to the wild-type strain. The mutations engineered into this strain produce a strain lacking the FNR protein. WT strains were grown under aerobic and anaerobic growth conditions. A six chip study using total RNA recovered from two separate cultures of Escherichia coli MG1655 K-12 WT (aerobic and anaerobic) and two separate cultures of the ?fnr mutant strain (anaerobic). Each chip measures the expression level of 4,661 genes from Escherichia coli MG1655 K-12 with eight 60-mer probes per gene, with each probe represented twice on the array.
