Project description:Background: Since chemical assays of soil nutrients are unreliable the UK horticultural and agricultural industries routinely apply large amounts of inorganic fertiliser to maintain crop yields and quality. Excessive fertiliser applications are both costly and can lead to unnecessary pollution. A possible solution to this problem is to use sensor (GM or non-GM) technologies that exploit the changes in plant gene expression profiles under incipient nutrient deficiency. The aim of this project is to identify genes upregulated in the early stages of nutrient depletion. Methods: Arabidopsis ecotype Col-5 (N1644) will be grown hydroponically using established techniques. In parallel experiments NP and K will be withdrawn individually after 21 d growth. RNA will be extracted from shoots of nutrient replete (control) and nutrient depleted plants 24 h after the removal of nutrients. Shoot nutrient content will be assayed by ICP-EMS as a reference. By comparing expression profiles we will be able to differentiate between genes that are upregulated by lack of specific nutrients and those upregulated by a universal stress-response system. Promoters and transcripts of these genes will underpin the development of novel sensor technologies and knowledge of the gene expression profiles will improve our understanding of the physiology of plant mineral nutrition.

Project description:The concentrations of twenty kinds of hormones in the follicular fluid were detected by high-performance liquid chromatography–mass spectrometry (HPLC-MS/MS). An Agilent 1200 series high-performance liquid chromatography (HPLC) instrument (Agilent, USA) was utilized. A PAL autosampler (CTC, Swiss) and a Gemini-NX-C18 column (2.0 mm×50 mm, 3 μm, Waters, USA) were used. The ion source was an API-4000 quadrupole electrostatic field orbit trap high-resolution mass spectrometer (Applied Biosystems, USA). The scanning mode was multiple reaction monitoring (MRM) (Agilent-1200 LC system coupled to an API400 mass spectrometer).

Project description:For cells to grow and proliferate both a growth signal and available nutrients are required. Several lines of evidence suggest that there is recriprocal regulation of nutrients by growth signals and growth signals by nutrients. Since microRNAs are regulated by growth signals, we sought to determine if microRNAs could also be regulated by nutrient availability. We found that several microRNAs change expression following glucose starvation and that their Ago2 occupancy remains constant.

Project description:Appropriate soil nutrient management is critical for modern Canola (Brassica napus L.) varieties and hybrids to meet their yield potentials. Canola fields are typically supplemented by the application of nitrogen, phosphorus, sulphur, and to a lesser extent, potassium fertilizers to maximize yield, while deficiency in these key macronutrients can result in severe growth phenotypes and significant yield losses. To date, our understanding of canola nutrient deficiency responses is disparate, encompassing a large number of varieties using a variety of technologies and assessment criteria, with seminal understanding of the overlaps between nutrient deficiencies responses, having not yet been defined, placing limitations on our ability to increase the nutrient efficiency of this critical oil seed. To address this, we performed a comparative quantitative proteomics analysis of both shoot and root tissue harvested from soil-grown Canola plants experiencing either nitrogen, phosphorus, potassium, or sulphur deficiency. Our results show intriguing similarities in plant responses to deficiency in multiple nutrients. We also find very distinct proteome-level changes between shoot and root tissue of plants experiencing nutrient stress, suggesting the presence of highly organ-specific responses to nutrient deficiency. Our results pave the way for a more comprehensive understanding of the shared and distinct response mechanisms of plants to multiple essential nutrients.

Project description:Nutrient availability, in particular that of carbon (C) and nitrogen (N), is one of the most important factors for the regulation of plant metabolism and development. In addition to independent utilization, the ratio of C to N metabolites in the cell is also important for the regulation of plant growth including. Plants sense and respond to the balance of carbon (C) and nitrogen (N) nutrients (C/N-nutrient) available to them, a process called the C/N-nutrient response. We previously demonstrated that disrupted high C/low N stress condition promotes the senescence progression in Arabidopsis plants. However, the molecular basis of C/N-nutrient responsive senescence regulation remains unclear. In this study, we carried out proteome analysis of phosphorylation dynamics in response to high C/low N nutrient stress.
