Determination of Pseudomonas aeruginosa transcriptome induced by oleic acid or its derived oxylipins 10-HOME and 7,10-DiHOME.

Project description:Oxygenated unsaturated fatty acids, known as oxylipins, are signaling molecules commonly used for cell-to-cell communication in eukaryotes. However, a role for oxylipins in mediating communication in prokaryotes has not previously been described. Bacteria mainly communicate via quorum sensing (QS) , which involves the production and detection of diverse small molecules termed autoinducers. We showed that oleic acid-derived oxylipins 10-HOME and 7,10-DiHOME produced by Pseudomonas aeruginosa function as autoinducers of a novel quorum sensing system termed Oxylipin-Dependent QS Sytem (ODS). This experiment was designed to determine the genes whose expression is altered by these P. aeruginosa oxylipins. We found that the ODS system controls the cell density-dependent expression of a P. aeruginosa gene subset through the mediation of 10-HOME and 7,10-DiHOME oxylipins.

Project description:The bacterial quorum sensing (QS) phenomenon has been well studied since its discovery and has traditionally been considered to include signaling pathways recognized exclusively within either Gram-positive (Gm+) or Gram-negative (Gm-) bacteria. The Gm+ bacteria are known to utilize quorum sensing pathways mediated by various small autoinducing peptides (AIP), while the Gm- bacteria use small molecules known collectively as acyl-homoserine lactones (AHL). Distinct structural differences between the types of signaling molecules have historically implied a lack of cross-talk among Gm+ and Gm- QS systems. Recent investigations, however, have demonstrated the ability for AIPs and AHLs to be produced by noncanonical organisms, implying QS systems may be more universally recognized than previously hypothesized. With that in mind, our interests were piqued by the organisms Lactobacillus plantarum, a Gm+ commensal probiotic known to participate in AIP-mediated QS, and Pseudomonas aeruginosa, a characterized Gm- pathogen whose virulence is controlled by AHL-mediated QS. Both health-related organisms are known to inhabit the human gut in various instances, both are characterized to elicit distinct effects on host immunity, and some studies hint at the putative ability of L. plantarum to degrade AHLs produced by P. aeruginosa. We therefore wanted to determine if L. plantarum cultures would respond to the addition of N-(3-oxododecanoyl)-L-homoserine lactone (3OC12) from P. aeruginosa by analyzing changes on both the transcriptome and proteome over time. Based on the observed upregulation of various two-component systems, response regulators, and native quorum sensing related genes, the resulting data provide evidence of an AHL recognition and response by L. plantarum.

Expression data of enterotoxigenic (ETEC) isolate E24377A

Project description:ETEC is an important human pathogen. Although the mechanism of diarrhea is known in ETEC, the regulatory networks are less understood. This study was conducted to understand the global expression of ETEC isolate E24377A under different growth and environemental conditions. ETEC isolate E24377A was grown in the presence of several chemical signals, including bile salts, glucose, and pre-conditioned media (PCM) from other enteric pathogens. E24377A was also grown to different densities, to see if a quorum sensing mechanism was in place The isolate was grown in different types of media, with different ammendments, and at different growth densities. Quercetin alleviates ETEC K88-induced host injury by driving the circulation and fluctuation of AHL-like quorum-sensing signaling molecules in the blood–liver–colon system

Project description:Abstract:Background: Quercetin is a flavonoid with various probiotic properties, demonstrating anti-inflammatory, anti-oxidative, anti-cancer, and anti-obesity effects in humans and animals. However, quercetin’s effect on enterotoxigenic Escherichia coli (ETEC) K88-induced host body injury mediated by bacterial quorum sensing remains insufficiently explored.Results: Quercetin supplementation alleviated ETEC K88-induced body damage in weaned piglets, changed the microbial composition of the host intestinal tract, enriched the main probiotic Lactobacillus amylovorus, and inhibited microbial functions, such as virulence factor secretion. In addition, it influenced changes in AHL-type quorum-sensing signaling molecules in the host circulation. The signaling pathway alleviated ETEC K88-induced host intestinal barrier and inflammatory damage.Conclusion: This study provides a theoretical framework for further understanding and exploring the use of quorum sensing as a medium to alleviate human and animal diseases caused by pathogenic bacteria and other external adverse factors.

Salmonella enterica Typhimurium grown with and without glucose

Project description:Cell to cell communication in bacteria to regulate various cellular processes with respect to their population density is termed quorum sensing and is achieved using signaling molecules called autoinducers. LuxS, which is involved in the synthesis of the autoinducer molecule-2 (AI-2), is conserved in several Gram-positive and Gram-negative bacteria including the enteric pathogen Salmonella Typhimurium. Genes that are regulated by luxS in S. Typhimurium were identified using microarrays and RNA samples from wild type S. Typhimurium and its isogenic luxS mutant, in two growth conditions (presence and absence of glucose), and at two different time points (mid-log and early-stationary phases). Minimal differential gene expression was observed in the presence of glucose. In the absence of both luxS and glucose, a total of 1560 genes were differentially expressed and 1361 genes were identified as luxS/AI-2-regulated at the mid-log phase and 199 genes at the early-stationary phase. Quantitative real-time PCR was performed on selected genes to validate the microarray results. These results suggest that although the expression of the luxS gene in S. Typhimurium is independent of the growth condition, its role in the production of AI-2 depends on the growth condition. It was found that luxS/AI-2 plays a vital role in a variety of processes such as metabolism, virulence gene expression, motility, transcription and translation.
