GNPS Skin exposed to white petrolatum

ABSTRACT: We performed an untargeted metabolomic analysis on surficial human skin samples collected with moistened cotton swabs (water:ethanol, 50:50) using LC-HR-MS/MS. Data-dependent acquisition was employed under positive ion mode. One cohort (five subjects) was recruited and white petrolatum was applied in the dominant hand (dorsal side) of the subjects for 7 days, while the other hand was not treated (non-exposed hand).

Protein metabolism drives feed efficiency in the liver and white muscle of efficient and inefficient Chinook salmon (Oncorhynchus tshawytscha) farmed in saline water

Project description:Understanding the molecular mechanisms of feed efficiency is an important step toward sustainability of salmonids aquaculture. In this study, the liver and white muscle proteomes of efficient (EFF) and inefficient (INEFF) Chinook salmon (Oncorhynchus tshawytscha) farmed in sea water were investigated by liquid chromatography-tandem mass spectrometry (LC-MS/MS) approach. In total, 2,746 liver and 702 white muscle quantified proteins were compared between 21 EFF and 22 INEFF fish. Protein synthesis was enriched in both liver and white muscle of the EFF group while conversely, pathways related to protein degradation (amino acid catabolism and proteolysis, respectively) were the most affected processes in the liver and white muscle of INEFF fish. The SOM in the INEFF group was significantly higher than EFF fish showing INEFF fish probably was the dominant group. The INEFF group (dominant) suffered stress and shifted to consume energy through protein catabolism. As the first study, the results provide a preliminary picture of the fundamental molecular landscape of feed efficiency in Chinook salmon farmed in sea water

Identification of anther expressing genes in cotton (Gossypium hirsutum)

Project description:This study was initiated with the objective of identifying the anther/tapetum specific promoters from cotton floral buds. Cotton is an important commercial crop. Hybrid cotton varieties are developed to obtain improved yield and fiber quality. Most of the hybrid seed production in cotton is carried out by hand emasculation, which requires large amount of manpower, resulting in high cost of hybrid seed. We are developing barnase-barstar based male sterility system, which would be a better alternative for hybrid development. The tapetum specific promoters are main requirement for such a system. The study was thus carried out to identify genes expressed in the anthers.

Comparative gene expression in healthy African American and White Non-Hispanic skin

Project description:The risk of atopic dermatitis, hidradenitis suppurativa, lupus erythematosus is significantly higher in African Americans (AA), while White Non-Hispanic (WNH) subjects have a greater risk of psoriasis. The mechanisms underlying disparity in inflammatory skin diseases risks are poorly understood, and the molecular landscape of skin of color including transcriptome and proteome are not well known. To assess whether and how gene expression pattern in normal skin may affect predisposition to specific skin inflammatory diseases, we performed analkysis of gene expression in full thickness skin biopsies obtained from healthy African American and White Non-Hispanic volunteers (all females, aged 22-46).

Flint glass bottles cause white wine aroma identity degradation

Project description:<p>Due to marketing recommendations, white wines are often bottled in flint glass to improve aesthetics and showcase wine color. Although this practice is known to cause a wine fault, the influence of light on the fruity and flowery aromatic profile of wine is unknown. The aim of this study was to investigate the changes to the white wine volatilome under typical supermarket shelf conditions, using 1,052 bottles of 24 white wines. After only 7 d of shelf life in flint glass bottles, a dramatic loss in terpenes (10 to 30%) and norisoprenoids (30 to 70%) was recorded, whereas colored glass bottles did not evidence such behavior even after 50 d, and darkness preserved the wine's fruity and flowery aromatic integrity. We also proposed an alternative mechanism for the insurgence of the lightstrike off-odor, which takes the varietal aroma loss into account. In light of this understanding of the flint glass negative impact on white wine aroma identity and sensorial character, this packaging should be strongly discouraged. The same findings should be valid for a wide range of several daily consumed foodstuff where transparent packaging is used.</p>

Mouse white adipose tissue HaloREVERBα ChIP-seq

Project description:The circadian clock component REVERBα is considered a dominant regulator of lipid metabolism, with global Reverbα deletion driving dysregulation of white adipose tissue (WAT) lipogenesis and obesity. However, a similar phenotype is not observed under adipocyte-selective deletion (ReverbαFlox2-6AdipoCre), and transcriptional profiling demonstrates that, under basal conditions, direct targets of REVERBα regulation are limited, and include the circadian clock and collagen dynamics. Under high-fat diet (HFD) feeding, ReverbαFlox2-6AdipoCre mice do manifest profound obesity, yet without the accompanying WAT inflammation and fibrosis exhibited by controls. Integration of the WAT REVERBα cistrome with differential gene expression reveals broad control of metabolic processes by REVERBα which is unmasked in the obese state.
