Triggering the polyurethane and polyethylene-degrading potential of filamentous fungi secretomes

Project description:In the present study, we screened various fungi for their ability to degrade intact polymers, such as ether-based PU and LDPE, using Impranil and a mixture of long-chain alkanes, not only as sole carbon sources but also as enzyme indicators for polymer degradation. The agar plate screening revealed 3 fungal strains belonging to Fusarium and Aspergillus genera, which were further cultured in presence of the same carbon sources, and their secretome was utilized for polymer degradation. Especially for ether-based PU, the secreted proteins of a Fusarium species reduced the weight and the molecular weight of the sample by 24.5 and 20.4%, respectively, while at the same time the secretome of an Aspergillus species caused changes in the molecular structure of LDPE. The proteomic analysis followed proved that the enzymes induced in presence of Impranil can selectively degrade the urethane bond of ether-based PU, a discovery that offers a new alternative in PU waste treatment. Meanwhile, the mechanism of LDPE degradation was not completely understood, although the presence of oxidative enzymes probably accompanied by reactive oxygen species, could be the main factors contributing to polymer functionalization.

Gene expression profiling of ramie roots during hydroponic induction and adaption to aquatic environment

Project description:Transcriptome analysis of root development related genes in terrestrial and hydroponic ramie. Ramie seedlings were cultivated in soil, and in hydroponic with the shoot-cutting propagation method. The root samples from hydrophonic ramie were collected from the early stage (5-day-old seedling) and the late stage (30-day-old seedling) of acquatic roots induction. The roots of ramie seedling cultivated in soil were also collected for comparative analysis. Our study represents the detailed analysis of ramie root transcriptomes with biologic replicates.

Expression profiling analysis of fungus Penicillium oxalicum wild type strain and its Podot1 deletion strain.

Project description:The goals of this study are to compare different gene expressions for Penicillium oxalicum wild type strain (WT), and Podot1 knockout strain (ΔPodot1) in different carbon sources. The deletion of Podot1 downregulated genes involved in the septin complex, extracellular region, and interspecies interaction between organisms when strains were cultivated with 2% glucose as carbon sources, and downregulated genes involved in cellulase activity, cellulose binding, glucosidase activity, and polysaccharide catabolic process when strains were cultivated with 1% microcrystalline cellulose and 1% wheat bran as carbon sources. We find the extracellular region was downregulated under both different carbon sources in ΔPodot1. This study provides the information that PoDot1 function are required in mycelial development and hydrolase activity of P. oxalicum.

The Drosophila immune response towards entomopathogenic nematodes and their associated bacteria.

Project description:Entomopathogenic nematodes (EPNs) of the genera Heterorhabditis are obligate and lethal insect parasites. In recent years they have been used increasingly as biological control agents. These EPNs are symbiotically associated with bacteria of the genera Photorhabdus. The bacterial symbionts are essential to kill the host (within 24-48 hours) and digest its tissues to provide nutrients for themselves as well for expanding nematodes. Drosophila larvae are suitable insect hosts and part of the tripartite model system we used before to show the importance of haemolymph clotting and eicosanoids during the infection. We used the well-established tripartite model (Drosophila, nematodes, bacteria), DNA chips and bioinformatic tools to compare gene expression in non-infected and infected fly larvae. We focused on the early time point of nematode infection and therefore infected Drosophila larvae using H. bacteriophora harbouring GFP-labelled P. luminescens bacteria. Infected (GFP positive) larvae were collected 6 hours after infection.

Oral primo-colonizing bacteria modulate inflammation and gene expression in bronchial epithelial cells

Project description:The microbiota of the mouth disperses into the lungs and both compartments share similar phyla. Considering the importance of the microbiota in the maturation of the immunity and physiology during the first days of life, we hypothesized that primo-colonizing bacteria of the oral cavity may induce immune responses in bronchial epithelial cells. Herein, we have isolated and characterized 57 strains of the buccal cavity of two human new-borns. These strains belong to Streptococcus, Staphylococcus, Enterococcus, Rothia and Pantoea genera; Streptococcus being the most represented. The strains were co-incubated with a bronchial epithelial cell line (BEAS-2B) and we established their impact on a panel of cytokines/chemokines and global changes in gene expression. The Staphylococcus strains, which appeared soon after birth, induced a high production of IL-8, suggesting they can trigger inflammation, whereas the Streptococcus strains were less associated with inflammation pathways. The genera Streptococcus, Enterococcus and Pantoea induced differential profiles of cytokine/chemokine/growth factor and set of genes associated with maturation of morphology. Altogether, our results demonstrate that the micro-organisms, primo-colonizing the oral cavity, impact immunity and morphology of the lung epithelial cells, with specific effects depending on the phylogeny of the strains.

POTENTIAL GENETIC TARGETS FOR CONTROLLING CAMPYLOBACTER JEJUNI

Project description:Campylobacter jejuni is one of the leading causes of bacterial diarrhea worldwide and the most common antecedent in peripheral neuropathies such as Guillain Barré and Miller Fisher syndromes (Blaser et al., 2000). Despite the medical and socioeconomic importance of C. jejuni, the proportion of human disease caused by different sources of infection remains unclear. Indeed, this issue has hindered effective control strategies against reducing C. jejuni levels from the food chain. Up to date, a vast number of phenotypic and genotypic typing systems and a large number of databases have been developed in order to determine the different Campylobacter infection sources. However, traditional methods are unable to discriminate strains from different sources that are responsible for causing disease in humans. Interestingly, DNA microarrays represent a technological alternative to compare entire genomes, allowing the identification of all the genes present or absent in a particular strain by comparison with the genome of the reference strain present in the microarray. Previous comparative genomic hybridazation studies of C. jejuni using DNA microarrays have focused on the genetic variability in C. jejuni (Dorrell et al., 2001), which suggest that this organism has high levels of genetic diversity and low levels of genetic plasticity (Taboada et al., 2004). On that account, the aim of this study was based on a comparison by CGH (Comparative Genomic Hybridization) using DNA microarrays for determining the degree of genomic variability of C. jejuni strains from different geographical areas and sources of isolation. This system enabled us to identify possible genes that could be used as genetic markers predictive of infection source. Furthermore, CGH data provide additional information that could be useful for the formulation of new hypotheses about C. jejuni genome evolution, virulence, pathogenicity, and host specificity.
