Investigation of over-expressing Annexin receptor cell line with and without agonists

Project description:The therapeutic potential of pro-resolution factors in determining the outcome of inflammatory events has gained ground over the past decade. However, the attention has been focused on the non-genomic effects of these endogenous, anti-inflammatory substances. In this study, we have focused our attention on identifying specific annexin 1 (AnxA1) protein/ALX receptor mediated gene activation, in an effort to identify down-stream genomic targets of this well-known, glucocorticoid induced, pro-resolution factor. In this study, we used four different types of samples (with 2 replicates each):1)cells transfected with the empty pGEX-4T-327 vector and vehicle (PBS) treated; 2)cells transfected with the annexin receptor ALXR (also known as FPR2) and vehicle (PBS) treated; 3)cells transfected with the annexin receptor ALXR and treated with 10uM Ac-26 peptide for 4 hours; 4) cells transfected with the annexin receptor ALXR and treated with 0.5uM Annexin 1 for 4 hours. After the treatments, cells were collected for RNA extraction.

Systematic analysis of the pro and anti-inflammatory activation of mouse myeloid cells

Project description:Inflammation is a fundamental physiological process and a key line of defense against pathogen invasion. Inflammation is exquisitely controlled: Too high and inflammation can be dangerous to the organism, but too low and the invading pathogen can escape suppression. Multiple opposing molecules initiate either a pro or anti inflammatory program, of particular interest is the pro-inflammatory bacterial endotoxins and an opposing anti-inflammatory pathway involving IL-10 and STAT3. Much work has been expended in understanding these two pathways in macrophages, key cells in the myeloid system. But other cells of the myeloid system also show a response to endotoxin and IL-10. These myeloid cell types have been less well explored. With the aim to understand similarities and differences amongst myeloid cells we performed RNA-seq analysis on 5 cells from the myeloid system: Macrophages, Neutrophils, splenic dendritic cells, mast cells and eosinophils. We treated the cells with IL-10 or LPS (endotoxin), either separately or in combination. Although the 5 cell types show much similarity in the outcome of the pro inflammatory response, the broad outline of their pro and anti-inflammatory response is highly divergent between the 5 myeloid cell types.

Proteomics of Staphylococcus aureus

Project description:Tannic acid (TA) and punicalagin(P), which both are polyphenols, have attracted much attention in recent years due to its extensive physiological activities, which include antioxidant, antitumoral, antimicrobial and anti-inflammatory actions. Previous work revealed that TA has an inhibitory effect on a variety of bacteria, including S. aureus, Salmonella, and Lactobacillus. Previous researches suggested that TA inhibited bacterial growth by chelating ions from the culture medium, and by inhibiting fatty acid synthesis through suppression of the activity of β-ketoacyl-ACP reductase, or by interfering with cell wall integrity. However, the mechanisms underlying the inhibitory effect of TA and P on S.aureus have not been assessed comprehensively. Therefore, the present study combined transcriptome and proteome data to analyze the molecular mechanisms of TA and P in S. aureus.

IDR-1018 treatment of mice infected with Plasmodium berghei, spleen and brain expression data

Project description:Severe malaria encompasses a range of syndromes manifesting systemically or in diverse organs. These are believed to represent the end-stage processes of local parasite sequestration and inflammatory cascades. Classical anti-malarial drugs target parasites only. In treatment of severe disease, adjunctive therapies capable of controlling the inflammatory processes could be beneficial. Innate defense regulator (IDR) peptides display multiple immune modulatory activities. In this study, we assessed peptide IDR-1018, which shows promise as an anti-inflammatory drug, as a lead candidate for adjunctive host-directed therapy of established disease in the P. berghei ANKA model of experimental cerebral malaria (ECM). Intravenously administered IDR-1018 partially protected mice from ECM both prophylactically and in adjunctive treatment with classical anti-malarial drugs. We used transcriptional data from spleens and brains taken early in infection (day 3) of prophylactically treated mice to investigate the protective mechanisms. The microarrays compared spleens and brains from nine IDR-1018 i.v. treated, infected mice (IDR-1018-treated infected) with three saline i.v. treated infected mice (saline-treated infected) and three uninfected untreated control mice (controls). RNA samples were hybridized in randomized order to five Illumina WG-6 v2 BeadChips . No technical replicates were performed.
