Project description:Our understanding of cellular mechanisms by which animals regulate their response to starvation is limited despite the close relevance of the problem to major human health issues. L1 diapause of Caenorhabditis elegans, where newly hatched first stage larval arrested in response to food-less environment, is an excellent system to study the problem. We found through genetic manipulation and lipid analysis that ceramide biosynthesis, particularly those with longer fatty acid side chains, critically impacts animal survival during L1 diapause. Genetic and expression analyses indicate that ceramide likely regulate this response by affecting gene expression and activity in multiple regulatory pathways known to regulate starvation-induced stress, including the insulin-IGF-1 signaling (IIS) pathway, Rb and other pathways that mediate pathogen/toxin/oxidative stress responses. These findings provide an important insight into the roles of sphingolipid metabolism in not only starvation response but also aging and food-response related human health problems.

Besides compromising public health and food safety, sub-clinical salmonellosis is also believed to be a major problem affecting the profitability of the pig industry. Distinct responses to Salmonella infection have been observed in pigs, some recovering faster and shedding lower levels of Salmonella in faeces than others (low shedders, LS versus persistent shedders, PS). This trait variation could indicate the existence of a genetic component to Salmonella shedding and resistance that may be exploited in animal breeding and disease diagnostics. The study aimed to characterize changes in miRNA expression in response to Salmonella infection.

Project description:Metabolic and reproductive processes interact in ways that are important for survival, particularly in mammals that gestate their young. Puberty and reproduction, as energetically taxing life stages, are often gated by metabolic availability in animals with ovaries. How the nervous system coordinates these interactions is an active area of study. We identify somatostatin (SST) neurons of the tuberal hypothalamus as a cell type in the feeding circuitry that alters feeding in a manner sensitive to metabolic and reproductive states in mice. Whereas chemogenetic activation of SST neurons increased food intake across sexes, selective ablation decreased food intake only in female mice during proestrus. Interestingly, this ablation effect was only apparent in animals with a low body mass. Fat transplantation and bioinformatics analysis of SST neuronal transcriptomes revealed white adipose as a key modulator of the effects of SST neurons on food intake. Together, these studies point to a mechanism whereby SST hypothalamic neurons modulate feeding by responding to varying levels of circulating estrogens differentially based on energy stores. This research provides insight into how neural circuits integrate reproductive and metabolic signals and illustrates how gonadal steroid modulation of neuronal circuits can be context-dependent and gated by metabolic status.

Project description:Type 2 diabetes mellitus represents a major health problem with increasing prevalence worldwide. Limited efficacy of current therapies have prompted a search for novel therapeutic options. Here we show that treatment of pre-diabetic mice with mitochondrially targeted tamoxifen, a potential anti-cancer agent with senolytic activity, improves glucose tolerance and reduces body weight with most pronounced reduction of visceral adipose tissue due to reduced food intake, suppressed adipogenesis and elimination of senescent cells. Glucose-lowering effect of mitochondrially targeted tamoxifen is linked to improvement of type 2 diabetes mellitus-related hormones profile and is accompanied by reduced lipid accumulation in liver. Lower senescent cell burden in various tissues, as well as its inhibitory effect on pre-adipocyte differentiation, results in lower level of circulating inflammatory mediators that typically enhance metabolic dysfunction. Targeting senescence with mitochodrially targeted tamoxifen thus represents an approach to the treatment of type 2 diabetes mellitus and its related comorbidities, promising a complex impact on senescence-related pathologies in aging population of patients with type 2 diabetes mellitus with potential translation into the clinic.

Project description:Coxiella burnetii is an intracellular pathogen that causes Q fever, a zoonotic disease found worldwide. In humans, the bacterium is responsible for an acute and chronic disease. It is transmitted to humans by the inhalation of aerosols or contaminated dust containing bacteria from infected livestock. Recently C. burnetii re-emerged in Europe making considerable economic impact in farming industry and even causing multiple human cases. Antibiotic resistance represents a global public health problem; moreover, there are few reports about doxycycline resistance in C. burnetii. Understanding drug application consequences in tolerant bacterial strains is indispensable for the effective therapy. Herein, we report changes in proteome of C. burnetii challenged by antibiotic stress, highlighting polypeptides likely involved in the detoxification of doxycycline. These data were corroborated by gene expression analysis. Based on the identities of differentially abundant proteins we suggest that homeostasis of bacterial surface, oxidative stress defense and balanced carbon metabolism are key processes to survive doxycycline treatment.

Project description:This study aimed to use pan-viral detection microarrays to identify viruses in serum from cases of acute pediatric febrile illness in a tropical setting. Patient clinical data and serum samples were collected between 2005 and 2009 as part of an ongoing pediatric dengue virus study at the Hospital Infantil Manuel de Jesús Rivera in Managua, Nicaragua. This study focused on patients who presented with dengue-like illness but who tested negative for dengue-virus infection. We hypothesized that non-dengue viruses or previously uncharacterized viruses might be causing these illnesses. The Virochip microarray is capable of detecting known viruses and discovering novel viruses. This series includes 153 arrays corresponding to 148 cases and 5 HeLa controls. Project description:Infertility or subfertility is a widespread problem with approximately 25% of couples seeking medical help. For about half the cases it lies in the male partner. Male infertility is caused by problems that affect sperm production, maturation and/or transport. In this study the role played by the epididymis in the post-testicular maturation of spermatozoa during its transit through this organ was investigated using FT-ICR imaging mass spectrometry of metabolites and the FDR-controlled metabolite annotation platform MetaSpace for high-resolution imaging mass spectrometry. This study together with MTBLS313 provides the accompanying data for FDR-controlled metabolite annotation for high-resolution imaging mass spectrometry.
