Testis proteome of Trypanosoma cruzi infected mice

Project description:Male CD-1 mice (N=8) were intraperitoneally infected by culture-derived Trypanosoma cruzi trypomastigotes of the Brazil strain. One group (N=4) was treated with an antitrypanosomal drug (nifurtimox) on day 74 post-infection (chronic phase) for 20 days. A group of uninfected mice (N=3) was also included as control. All groups (N=11) were enthanized on day 110 post-infection, testes were collected and subjected to 2D-LC-MS/MS analysis. Label-free quantitation revealed that the testes of T. cruzi-infected mice had an altered proteome pattern relative to treated mice and uninfected controls. The analysis also demonstrated that the left and right testes had different proteomic profiles in infected animals.

Coxsackievirus B3 (CVB3) infection of 129S1 and 129X1 (heart tissue day 6)

Project description:We infected two strains of mice, 129S1/SvImJ and 129X1/SvJ, with coxsackievirus type b3 (CVB3) at a dose of 500 pfu/g. 129S1 mice developed increased cardiopathology despite equal viral replication. We hypothesized that the increased cardiopathology might result from an ongoing pathologic host response that we could characterize by global expression profiling. Gene expression was assessed in hearts from 129S1 and 129X1 mice that were uninfected or infected for 6 days. Total RNA obtained from hearts of 3 129S1 and 3 129X1 that were infected or uninfected with CVB3(H3) at 500pfu/g and collected at day 6 post infection

Transcription profiling of mouse lung from C57BL6 and DBA2 parental strains and D2.B6-Chr7 and D2.B6-Chr19 congenic mice following aerosol infection with Mycobacterium tuberculosis for 30 and 70 days

Project description:To gain insight into the host cell types, cellular and molecular pathways possibly involved in the differential permissiveness to pulmonary replication of M. tuberculosis, we carried out transcript profiling studies on M. tuberculosis-infected lungs from congenic and parental strains. We were particularly interested in two groups of transcripts. The first group consists of transcripts which expression in the lung is regulated in response to M. tuberculosis infection (global response to infection), and that is obtained by comparing transcripts profiles of infected vs. uninfected lungs. The second group of transcripts is associated with increased resistance to M. tuberculosis infection of B6 and D2.B6-Chr7 mice. That list consists in the overlap between the lists commonly expressed in response to infection between resistant B6 and D2.B6-Chr7 but that show a significant difference in modulation when compared to infected susceptible D2.<br><br> In these experiments, B6, D2 as well as the D2.B6-Chr19, and D2.B6-Chr7 congenic lines were infected with M. tuberculosis and lungs were harvested at day 30 and day 70, and RNA was prepared. Three independent RNA samples from each group were converted to labeled cDNAs and hybridized to Affymetrix oligonucleotides arrays (Mouse Genome 430 2.0 array). Hybridization results were analyzed with the Genesifter analysis program to characterize changes in gene expression.

Gene expression changes in liver upon LCMV Cl13 infection.

Project description:2-3 month old female C57Bl/6J mice were intravenously infected with 2x10^6 focus forming units of lymphocytic choriomeningitis virus strain Clone-13. Whole liver tissue was harvested in duplicated from uninfected control mice, uninfected 123d old mice, and from infected at 2, 8, 30, 60, and 123 days post infection. Three TMT 6-plex runs were carried out to monitor the changes in liver protein abundance during the entire course of infection. The first run included samples from day 2 and day 8 after infection along with the uninfected controls. Similarly, the second run included uninfected controls and samples from day 30 and day 60 after infection. The third run included samples day 123 after infection, the corresponding age matched control that was left uninfected for 123 days and the uninfected controls used in the other two runs. The uninfected controls were included in all the three runs as an internal control to monitor reproducibility between the runs.

Project description:Study of tick cells from the Rhipicephalus microplus vector (BME/CTVM6) were infceted by SFTS virus at MOI of 1 PFU/cell. We collected samples at day 3 and 6 post-infection, and added the mock infected control. This was done in 4 replicates, and allow us to perform a proteomics informed by transcriptomic study, as well as differentially protein expression analysis.
