GNPS Annulohypoxylon moriforme MA9: NP3_MS_Workflow case validation: proteasome inhibitor candidates

ABSTRACT: Ethyl acetate extract of Annukihypoxylon moriforme MA9 strain (endophytic fungi) showed potent proteasome inhibition. It was performed chromatographic steps to generated two sets of bioactive enriched fractions. Extract and fractions were submitted to UPLC-MS/MS analysis and MS data was evaluated by NP3_MS_Workflow to predict most probable m/z candidate responsable for proteasome inhibition.

PARVB overexpression increases migration capability and defines high risks for endophytic growth and metastasis in tongue squamous cell carcinoma.

Project description:Tongue squamous cell carcinoma (TSCC) varies in characteristics even in early stages and is mainly classified into three subtypes, which are superficial, exophytic and endophytic types, based on a macroscopic appearance of tumor growth.Of these subtypes, endophytic tumor has a poorer prognosis because of its invasive feature and higher frequency to have metastasis. To understand a molecular mechanism of endophytic subtype and identify biomarkers, we performed comprehensive microarray analysis for mRNAs from clinical biopsy sampleswhich were classified into subtypes and found overexpression of parvin-beta (PARVB) gene significantly related to endophytic type. PARVB is known to play a critical role in actin reorganization and focal adhesions. Knocking down PARVB expression in vitrocaused apparent decreases in cell migration and wound healing, implying that PARVB has a crucial role in cellular motility. Moreover, metastasis-free survival was significantly lowered in patients with higher PARVB expression. Therefore overexpression of PARVB is a candidate biomarker for endophytic tumor and metastasis and may be clinically applicable for decision making of an adjuvant therapy in TSCC. Twenty seven OCT embedded tissues were used to extract total RNA. Then RNAs were amplified, biotinylated, fragmented and hybridized on GeneChip Human Genome U133 plus 2.0 arrays.

Thiolutin is a direct inhibitor of RNA Polymerase II

Project description:Thiolutin is a well-known and long-used natural product transcription inhibitor with an unresolved mode of action. Recent studies have identified Zn2+-chelation activity for both thiolutin and the related dithiolopyrrolone holomycin, with direct inhibition of RNA polymerases ruled out as a mode of action. However, negative results for direct transcription inhibition of RNA polymerases in recent studies are contradicted by previously observed thiolutin inhibition of extract fractions containing yeast RNA Polymerases I, II, and III. Here, we present chemical genetic and biochemical approaches to investigate the mode of action of thiolutin. We identify diverse classes of mutant that alter sensitivity to thiolutin. We functionally dissect the multidrug resistance and thioredoxin pathways controlling thiolutin sensitivity. We provide genetic evidence that thiolutin causes oxidation of thioredoxins in vivo and suggest that thiolutin both induces oxidative stress and alters Zn2+, Cu2+ and Mn2+ homeostasis in vivo, recapitulating previously observed thiolutin interactions with Zn2+. Finally, our results resolve contradictory biochemical results for thiolutin inhibition of transcription by direct demonstration of thiolutin inhibition of RNA polymerase II (Pol II) in vitro. Inhibitory activity requires both appropriate reduction of thiolutin and the presence of Mn2+. Thio/Mn2+ inhibition is abrogated when template DNA is pre-bound to Pol II or when excess DTT is present, and renders Pol II pause prone in elongation if initiation is bypassed using a pre-formed Pol II elongation complex. Together, we propose that thiolutin directly inhibits Pol II transcription through a novel mechanism distinct from known transcription inhibitors.

Thalicthuberine: A Novel Mitotic Inhibitor in Prostate Cancer Cells

Project description:Bioassay-guided fractionation of a cytotoxic extract derived from the Australian endemic plant Hernandia albiflora (twigs) resulted in the isolation of the two known compounds: deoxypodophyllotoxin and thalicthuberine. Both compounds were tested for their inhibition of LNCaP cell viability. Thalicthuberine (TH, IC50=2.0 µM) was selected for mechanism of action studies. Ongoing investigations have shown that TH induced a strong accumulation of LNCaP cells in mitosis, severe mitotic spindle defects and asymmetric cell divisions, ultimately leading to mitotic catastrophe accompanied by cell death through apoptosis. However, unlike microtubule-targeting drugs (vinblastine, paclitaxel and nocodazole), TH did not affect tubulin polymerization in a cell-free system, while it reduced cellular microtubule polymer mass, suggesting that TH indirectly targets microtubule dynamics through inhibition of critical regulators or tubulin-associated proteins. Based on phenotypic comparisons, we excluded the mitotic regulators Aurora kinase A and Polo-like kinase 1 as potential targets of TH. TH did not directly interact with DNA (cell-free assay) or induce DNA damage in cells. Further experiments will attempt to address the molecular target(s) and inhibition mechanism of TH. This is the first report of a mechanism of action study for this natural product. Based on these results, TH has great promise for further development into a potent non-tubulin-targeting mitotic inhibitor.

Project description:IL-27 treated DCs were shown to be highly potent inhibitors of cis HIV-1, particularly of CCR5 tropic strains. Microarray studies of IL-27 treated DCs showed no up-regulation of Type I (IFN) gene expression. Neutralization of the Type-I IFN receptor had no impact on the HIV inhibition. Lastly, IL-27 mediated inhibition was shown to act post-viral entry and prior to completion of reverse transcription. These results show for the first time that IL-27 is a potent inhibitor of cis HIV-1 infection in DCs by a Type I IFN independent mechanism.
