ABSTRACT: Bisphosphorylated cMyBP-C following dephosphorylation of pS279,pS288,pS308,pS313-cMyBPC by PP1 or PP2A. Samples analysed by the BIOSCIENCE TECHNOLOGY FACILITY, METABOLOMICS AND PROTEOMICS LABORATORY (BTF-MAP lab) of the University of York.

INSTRUMENT(S): Enriched phosphopeptides were loaded onto a 50 cm EN C18 PepMap column with elution over a 1 h acquisition driven by a Waters mClass nanoUPLC. Eluted peptides were measured using a Thermo Orbitrap Fusion Tribrid mass spectrometer, with MS spectra acquired in the Orbitrap and MS2 spectra acquired in parallel, following HCD fragmentation, in the linear ion trap.

Immunometabolism in resolving atherosclerosis guides the development of nanometabolite therapy

Project description:A 6-month in vivo study will be carried out at Case Western Reserve University (CWRU) using an ApoE-/- mouse model of atherosclerosis (obtained from The Jackson Laboratory), which will be treated as follows: Control: Mice fed a chow diet (control) (n=12) Progression: Mice fed a high-fat diet with cholesterol (n=12) Regression: Mice fed a high-fat diet with cholesterol followed by a switch to a chow diet (n=12) At the end of the experiment, mice will be euthanized and aorta excised from each animal. They will be immediately incubated in dialyzed serum-containing medium with uniformly C13 labelled glucose for 24 h and then extracted for Stable Isotope Resolved Metabolomics (SIRM) experiments. The incubation media and tissue extracts will be sent to University of Kentucky’s RC-SIRM: Resource Center for Stable Isotope-Resolved Metabolomics for analysis. The samples will undergo the following modes of analysis: nuclear magnetic resonance (NMR), Ion Chromatography Coupled with Mass Spectrometry (ICMS)

A gene expression map of the Arabidopsis root

Project description:This experiment was donated by Philip N. Benfey's lab through ArexDB (http://www.arexdb.org). A global map of gene expression within an organ can identify genes with coordinated expression in localized domains, thereby relating gene activity to cell fate and tissue specialization. Here, we present localization of expression of more than 22,000 genes in the Arabidopsis root. Gene expression was mapped to 15 different zones of the root that correspond to cell types and tissues at progressive developmental stages. Patterns of gene expression traverse traditional anatomical boundaries and show cassettes of hormonal response. Chromosomal clustering defined some coregulated genes. This expression map correlates groups of genes to specific cell fates and should serve to guide reverse genetics.
