Growth factor influence on the gene expression profile of human meibomian gland epithelial cells

Project description:Analysis of growth factor influence on immortalized human meibomian gland epithelial cells at gene expression level. Growth factors play a critical role in the proliferation and differentiation of sebaceous gland epithelial cells. Given that the meibomian gland is a large sebaceous gland, we hypothesize that growth factors exert analogous effects on human meibomian gland epithelial cells. Results provide important information of the response of human meibomian gland epithelial cells to epidermal growth factor (EGF), bovine pituitary extract (BPE), and both, such as up- or down- regulated genes involved in lipid metabolic process and cell cycle. Human meibomian gland epithelial cells were immortalized in our lab with a retrovirus containing telomerase reverse transcriptase (hTERT). hTERT immortalized cells (3 wells /condition / experiment) were seeded onto 6-well plates at the density of 3.76M-CM-^W104cells /well in SFM basal medium, SFM basal medium with epidermal growth factor (EGF; 5 ng/ml), SFM basal medium with bovine pituitary extract (BPE; 50 M-BM-5g/ml), and SFM basal medium with 5ng/ml EGF plus 50 M-BM-5g/ml BPE (KGM). Total RNA was extracted from cultures in SFM basal medium at day 2, and from cultures in SFM basal medium with BPE, basal medium with EGF, and SFM basal medium with EGF plus BPE at day 7.

Human saliva modifies growth, biofilm architecture and competitive behaviors of oral streptococci

Project description:Our group recently transcriptomically characterized coculture growth between Streptococcus mutans and several species of commensal streptococci (Rose et al, 2023). However, these experiments were carried out in our lab-based experimental medium, tryptone and yeast extract (TY-). To understand whether culturing these species within a medium that more closely mimics their natural environment alters the interaction, we evaluated both monoculture and coculture growth between the dental caries pathogen Streptococcus mutans and oral commensal species Streptococcus oralis in a half TY- / half human saliva mix that was optimally chosen based on our initial characterization of oral streptococci behaviors in medium mixes containing saliva. Our results surprising show that inclusion of saliva enhances the competition of Streptococcus mutans against commensal streptococci through upregulation of carbohydrate uptake and glycolytic pathways.

Effect of overexpression of yeast Ppz1 protein phosphatase on gene remodeling in response to changes in carbon source

Project description:Our previous results (Calafi et al 2020; PMID: 32339526) demonstrated a defect in cell proliferation when PPZ1 was overexpressed under the control of a tetO-based promoter that was, up to some extent, dependent of the availability of glucose. To further investigate this phenomenon, we have determined the transcriptional response triggered by the shift form high to low glucose, or to galactose, in normal cells and in cells overexpressing PPZ1 from the tetO7–driven multicopy pCM190 plasmid. Our data suggest that, under these circumstances, overexpression of Ppz1 interferes with the proper activation of a set of genes mainly involved in carbon metabolism, thus impairing the normal transcriptional adaptation to conditions in which glucose becomes limiting.
