GNPS Too Much Botrytis Co-culture

ABSTRACT: First try at GNPS. Bacillus mycoides is being investigated as a potential biological control agent against Botrytis cinerea, and we wish to elucidate what compounds are causing the inhibition of Botrytis cinerea growth. Bacillus mycoides was cultured in NB, Botrytis cinerea was cultured in NB, then B. cinerea and B. mycoides were cultured in NB together.

Project description:Bursaphelenchus xylophilus (pinewood nematode, PWN) is the causal agent of pine wilt disease, causing economic and environmental losses in pine forests. The establishment of systemic acquired resistance (SAR) offers positive prospects for PWN control. We chose Bacillus subtilis JCK-1398, which effectively mitigated disease symptoms in PWN-infected pine trees. To elucidate the molecular responses involved in increased SAR according to B. subtilis JCK-1398 treatment, we characterized the in vivo transcriptomes of pine trees infected by PWN with B. subtilis JCK-1398 treatment. Additionally, pine trees infected by PWN after Tween20 treatment were used as a negative control.

Project description:Botrytis cinerea is the causing agent of grey mould on hundreds of plants cultivated in fields, tunnels and greenhouses worldwide. Considering the central role of secretion in the necrotrophic interaction between the fungus and its host plants, non conventional secre-tion was explored through the search and characterisation of extracellular vesicles. In or-der to reveal the protein composition of these vesicles, a proteomic analysis was chosen.

Proteomic analysis of the proteins secreted by the infection cushions of Botrytis cinerea.

Project description:Gray mold disease affects fruits, vegetables and ornemental plants around the world, causing considerable losses every year. Its causing agent, the fungus Botrytis cinerea, produces infection cushions (IC) that are complex appressorial structure dedicated to the penetration of the plant tissues. A microarray analysis identified 1,231 up-regulated genes in IC-enriched mycelium, among which several genes coding for putative secreted proteins. These data were supported by qPCR analysis in vitro and in planta. They highlight a secretion of proteins involved in virulence like proteases, plant cell wall degrading enzymes and necrosis inducers. To validate these transcriptomic data, a proteomic analysis of the IC exo-proteome is required.
