Identification of antioxidant metabolites from Five Plants of the Polynesian Pharmacopoeia and Cosmetopoeia for skin care

Project description:Oxidative stress contributes to impairment of skin health and aspects in the wound healing process for pathologies such as acne, psoriasis or skin cancer. Five Polynesian medicinal plants, among the most traditionally used for skin care (cosmetopoeia and pharmacopoeia) are studied herein for their antioxidant properties: Calophyllum inophyllum, Gardenia taitensis, Curcuma longa, Cordia subcordata and Ficus prolixa. For this purpose, plant extracts were submitted to in vitro bioassays related to antioxidant properties and their bioactive constituents were identified by metabolomic analytical approach. UHPLC-MS/MS analysis was performed leading to characterize 61 metabolites. Compounds annotated for F. prolixa and C. subcordata were reported for the first time in those indigenous trees. Antioxidant properties were evaluated by TPC, DPPH and FRAP assays. F. prolixa extract was the most active one within antioxidant properties similar to ascorbic acid and showed antioxidant intracellular activity on HaCaT model by AOP1 assay. On-Line-HPLC DPPH allowed the identification of phenolic bioactive compounds. These results highlight the potential of F. prolixa aerial roots as a source of antioxidant for skin care topical applications.

Transcriptional toxicity of imidazolium- and cholinium-based ionic liquids

Project description:Ionic Liquids are a broad group of salts with low melting points that can be specifically tuned for a broad range of applications. Despite being initially considered “green” solvents, their better environmental friendliness compared to traditional solvents has been increasingly challenged. In this study, we aimed to investigate the molecular effects of ILs exposure by using RNA-sequencing to study differential gene expression patterns. Thus, we exposed Daphnia magna to 1-ethyl-3-methylimidazolium chloride ([C2mim]Cl), 1-dodecyl chloride-3-methylimidazolium ([C12mim]Cl) and cholinium chloride ([Chol]Cl). Results suggest that the three ILs share several mechanisms of toxicity, including cellular membrane and cytoskeleton damage, oxidative stress, inhibition of antioxidant enzymes, mitochondrial affectation, changes in protein biosynthesis and energy production, DNA damage, and ultimately, programmed cell death and disease initiation. Overall, the dataset revealed that [C2mim]Cl and [C12mim]Cl were, respectively, the least and the most toxic ILs at the transcriptional level. Also, it is reinforced that [Chol]Cl is not devoid of environmental hazardous potential. Unique gene expression signatures could also be identified for each IL.

Metabolic fingerprinting of different populations of Phyllanthus niruri L. from Punjab using electrospray ionization mass spectrometry (ESI–MS)

Project description:There is growing interest of healthcare and food industry in ingredients of plant origin as they are potent sources of antioxidant, anti-inflammatory, antimicrobial and anticancer agents. In the present work different extracts of Phyllanthus niruri L. from various regions of Punjab were screened for their phenolic profiles and antioxidant properties. Crude extracts obtained by solid–liquid extraction with different solvents were tested for total anthocyanins, flavonoids, phenolic content, and free radical scavenging activity. Out of all the solvents used, methanol was regarded as best to be used for soxhlet extraction of plant metabolites, as it provided highest phenolic, flavonoid, antioxidant, and anthocyanin contents. Similarly, ESI–MS was employed to obtain mass profiles of phenolic and other metabolites present in various P. niruri populations. Out of 72 compounds detected, 51 are reported for the first time in P. niruri L. Similarly, different populations of P. niruri were discriminated through metabolic fingerprinting using ESI–MS.
