GNPS - NASTE DOM and coral tissue nutrient dosing experiment

ABSTRACT: Dissolved organic matter and coral tissue metabolites from a three, three week nutrient dosing experiment using an inorganic nutrient mixture, bird guano, or effluent. DOM and tissue metabolites were from either Porites or Montipora coral treatments. The first three weeks just dosed the corals with nutrients, the following increased temperatures to simulate future warming conditions, and the last three were a recovery period. Untargeted LC-MS/MS acquisition was performed on a Vanquish Flex UHPLC system coupled to an Orbitrap Exploris 240 (Thermo Fisher Scientific, Bremen, Germany). Chromatographic separation was performed on a Kinetex 1.7 um 100 A pore size C18 reversed phase UHPLC column 150 x 2.1 mm (Phenomenex, Torrance, CA).

The proteomic response of the coral symbiont Symbiodinium microadriaticum to thermal and nutrient stress

Project description:Abstract The coral–dinoflagellate symbiosis is increasingly disrupted by global and local anthropogenic stressors. Coral bleaching is primarily a result of high sea surface temperatures, while eutrophication is associated with reef ecosystem degradation. Excess inorganic nitrogen relative to phosphate has been proposed to directly sensitise corals to thermal bleaching and accelerate reef decline. We assessed the proteomic response of the dinoflagellate coral symbiont Symbiodinium microadriaticum to elevated temperatures under multiple nutrient conditions by mass spectrometry. Elevated temperatures resulted in reductions of many chloroplast proteins, particularly light-harvesting complexes, with simultaneous increases in chaperone proteins. N:P imbalance had a larger effect on the proteome than temperature, but the biological processes and proteins responding to each stressor largely overlapped. The proteomes were highly similar at low N:P ratios but were strongly affected by phosphate starvation. High N:P ratios inhibited cell division, reflected by changes in proteins involved in protein translation. Imbalanced N:P did not increase sensitivity to high temperatures as measured by physiological means; however, imbalanced N:P strongly upregulated cell redox homeostasis proteins at high temperatures. As redox balance is critical during thermal bleaching, these data provide insight into the mechanisms of cellular responses to thermal and multiple stresses in the coral–dinoflagellate symbiosis.

Project description:Purpose: There is a dearth of knowledge regarding the molecular pathology of growth anomaly in corals. We investigated the gene expression profile of Montipora capitata metatranscriptomes from healthy and diseased (growth anomaly) coral colonies to elucidate differentially expressed genes. Methods: mRNA profiles of coral tissue (including symbionts) were generated from three different tissue states: healthy, affected and unaffected. Healthy tissue was collected from coral colonies not affected by growth anomaly. Affected tissue was collected from coral growth anomaly lesions. Unaffected tissue was collected from coral colonies affected by growth anomaly.

Open TG-GATEs (in vivo repeat dosing)

Project description:The Toxicogenomics Project was a 5-year collaborative project (2002-2007) by a consortium comprising the Japanese government and several pharmaceutical companies. The project produced the 'Toxicogenomics Project-Genomics Assisted Toxicity Evaluation system' (TG-GATEs), a large-scale database of transcriptomics and pathology data potentially useful for predicting the toxicity of new chemical entities. Conventional in vivo toxicology data was collected from single dose and repeat dosing studies on rats, and gene expression measured for the liver (and kidney in some cases). To provide information on species differences, gene expression was also measured in rat and human hepatocytes treated with the chemicals in vitro. Approximately 130 chemicals, primarily medicinal compounds, were tested at multipe doses. Gene expression was analysed using Affymetrix GeneChip arrays. The gene expression data has been submitted in four parts: in vivo single dose (E-MTAB-799), in vivo repeat dosing, in vitro rat (E-MTAB-797) and in vitro human (E-MTAB-798). This submission comprises the in vivo, repeat dosing studies. If publishing results based on this data, please cite the full project name 'Toxicogenomics Project and Toxicogenomics Informatics Project', the database name 'Open TG-GATEs' and the URL 'http://toxico.nibio.go.jp'.
