GNPS Big 5-2 acid mine drainage water filtration test

ABSTRACT: Test run of method for acid mine drainage filtration. Water was collected from Big 5 water treatment pipeline, stored in HDPE bottle for ~8 months, thawed, filtered through a 0.22 um PES filter in a Nalgene reusable bottle top filter unit, and transferred onto SPE column via PTFE tubing and polypropylene adapter. The purpose of this test was to determine what contaminants or extractables could be expected with the above workflow.

Sample preparation procedure for mass spectrometric analysis of plant histones

Project description:The protocol for preparing samples of plant histones prior to LC-MS/MS analysis was established, using a filter-aided sample preparation (FASP) technique to remove natural contaminants and chemical additives incompatible with MS-based procedures. The protocol consists of nuclei isolation, histone extraction into acidic solvent, protein propionylation and on-membrane digestion, then peptide propionylation in solution. Two variants of the derivatization protocol, in-solution propionylation in the vial (PROP-in-SOL) and propionylation on the filter unit (PROP-on-FILTER), were compared by efficiency of the labeling and by abundance of identified histone peptide forms.

Comparative Microbial Metagenomic Analysis of Drinking Water Plants and Wastewater Treatment Plants in Istanbul

Project description:Wastewater treatment plants (WWTPs) and Drinking water treatment plants (DWTPs) are critical points for public health for persistently remaining microorganisms after treatment may pose a risk. This study aimed to conduct microbial metagenomic analyses on waters from both DWTPs and WWTPs under the Istanbul Water and Sewerage Administration (ISKI). In this study a total of 52 samples were included, comprising 18 samples from DWTPs and 34 from WWTPs. All water samples underwent pre-isolation filtration. DNA isolation was conducted using filter material, followed by library preparation and sequencing on a NovaSeq 6000 instrument following the manufacturer's guidelines.

Analysis of melanosomal, exosomal and cellular miRNAs of melanoma cells

Project description:miRNA microarray profiling was performed for exosomes and secreted melanosomes of MNT-1 cells as well as WM3682 and WM3314 cells and melanosomes isolated from cell homogenates. The aim of this study is to compare the miRNA content of exosomes and secreted melanosomes and to assess differences in the miRNA profiles of different melanoma cell lines and their melanosomes. Total RNA was extracted from MNT-1 exosomes and secreted melanosomes (2 replicates each) that were isolated from the conditioned medium of MNT-1 melanoma cells by differential ultracentrifugation. In addition, total RNA was isolated from WM3682 and WM3314 melanoma cells and melanosomes (1 replicate each). These latter melanosomes were isolated from cell homogenates by filtration through a 0.45 Âµm filter, followed by FACS sorting. Project description:Kertinocyte cultures grown in 60 mm petri dishes were placed 186 mm from the solar simulator source (Solar-simulated ultraviolet radiation 1600W Xenon short arc lamp with an Oriel Air Mass 1 Direct Filter, (AM1:D:B; model 81074) and KG2 Short Pass Filter. irradiance 9.84 mW/cm² for UVA (98.3%), 0.174 mW/cm² for UVB (1.7%) and 10 mW/cm² (0.017 mW/cm² erythemally-weighted) for the total UVR irradiance) and received a dose of either 0, 10, 20 and 150 kJ/m2 of unweighted ultraviolet radiation and 0, 10 and 150 kJ/m2 of unweighted ultraviolet radiation with SPF 15 sunscreen filtration (Homosalate 3%, Octisalate 4%, Avobenzone 2%, Titanium dioxide 0.66%) (2 mg/cm2 sandwiched between two 5x5 inch quartz plates) and were temperature controlled at 37oC using a customized water-bath. Six and Twenty-four hours post-exposure cells were harvested and RNA was extracted and subjected to microarray analysis.

The proteome data of human mesenchymal stem cell derived from healthy donors

Project description:The protein samples stored at -80 °C were thawed and treated with lysis buffer (8M urea, 1% protease inhibitor, 50mM NH4HCO3) for 5 minutes in room temperature. After ultrasonic cracking on ice and centrifugation at 14000g for 10 min at 20 °C, the amount of total protein of each MSC sample was detected by bicinchoninic acid assay (Beijing Solarbio Science & Technology Co., Ltd., Beijing, China) using NanoDrop ND-1000 spectrophotometer (Thermo Fisher Scientific, Inc., Sunnyvale, CA, USA). Protein digestion was carried out by a filter-aided sample preparation technique. Briefly, 50 μg of a sample was loaded onto a Microcon filter unit YM-30 (Millipore) and washed with 8 M urea, followed by protein alkylation with 50 mM iodoacetamide in 8 M urea in darkness for 20 min. After alkylation, the filters were washed twice with 8 M urea and twice with 40 mM ammonium bicarbonate. Trypsin (Promega, Madison, WI, USA) was added to the samples at 100:1 protein-to-enzyme ratio and incubated overnight at 37 °C. Later the same
