Project description:To study the relevance of the phosphorylation of H4 in residues T30 and S47, we assessed the genome-wide transcriptional response of the non-phosphorylatable H4-T30A and the phospho-mimetic H4-T30D mutants upon heat stress and the phospho-mimetic H4-S47D mutant upon osmotic stress . Results point out that H4-T30(A/D) and H4-S47D mutants showed an impaired stress responsive gene expression indicating that these specific residues play a relevant role in, respectively, heat and osmotic stress responsive transcriptional regulation.

Project description:S. cerevisae cells were exposed to different series of mild stresses. Stress type include heat shock, oxidative and osmotic stresses. Microarrays were used to follow the genome-wide transcriptional response to the stresses and to identify genes that can underlie the cross protection phenotype between heat shock and oxidative stress. Experiment Overall Design: Cell sample at different time points after stress application were used for RNA extraction and hybridization on Affymetrix microarrays.

Project description:Pollen development is central for plant reproduction and is assisted by changes of the transcriptome and the proteome. At the same time, pollen is largely heat sensitive, particularly at early developmental stages. To define the changes during development and the response of different stages to elevated temperatures we investigated the transcriptome and proteome of pollen at tetrad, post-meiotic and mature stage before and after application of elevated temperatures. We used Solanum lycopersicum as an economically important crop. We demonstrate that the number of transcripts declines from tetrad to mature stage, while the protein content shows the opposite trend. Comparison of the transcriptome and proteome led to the discovery of two translational modes assigned as direct translation – transcripts increased in a specific pollen stage lead to increased translation in the same stage – and delayed translation – transcripts are produced for translation in later stages. The response to elevated temperatures is with ~5% of all genes being upregulated comparable to general plant behavior. The alteration at proteome level marked the mature pollen as least responsive and the post-meiotic stages as most responsive with altered levels of 40% of the identified proteins. For a subset of the genes with direct translation we observed a downregulation of protein levels after heat stress, whereas some of the genes with delayed translation showed a shift from delayed to direct translation when heat stress is applied. Further, Hsps, proteasome subunits, ribosomal proteins as well as eukaryotic initiation factors are the most drastically affected groups. On the example of Hsps we document that the response at transcriptome and proteome levels in e.g. the post-meiotic and mature stage does not correlate. Moreover, while all stages respond to heat stress with regulation of transcript abundance of Hsps in a comparable manner, changes at proteome level are distinct. Project description:Haploid embryos can be induced from cultured immature pollen following a stress treatment. In Brassica napus, application of the histone/lysine deacetylase (HDAC/KDAC) inhibitor trichostatin A (TSA) to pollen cultures enhances the production of differentiated embryos and embryogenic callus when applied together with heat stress (Li et al., 2014). To identify genes associated with the induction of B. napus haploid embryogenesis, we compared the transcriptomes of untreated pollen cultures and pollen cultures treated with either heat-stress or heat-stress plus TSA.

Project description:The consequences on tuber transcriptome of a short heat period during tuber development was investigated in this study with special regard to the development of secondary tuber growth. Plants were grown for 47 days in the greenhouse under ambient conditions (21°C/ 19°C, 16h light, 8h dark) before application of mild heat stress temperatures (29°C/27°C) to one group of plants for 7 days and a stress release period on control temperature for 2 more weeks until harvest. Leaves were sampled before the heat period, at the end of the heat period and at harvest, two weeks after stress release. Tuber samples were taken at harvest. Tubers grown at normal temperatures and exhibiting a normal growth phenotype were used as control. Tubers subjected to the heat treatment and exhibiting a second-growth phenotype (chain tubers) were grouped into primary (attached to stolon from plant) and secondary tubers (attached to stolon from primary tuber).

Project description:Gene expression was measured in control and heat resistance selected adult female flies before and at 8 time points after heat stress for 1h @ 36 degrees,Abstract The availability of full genome sequences has allowed the construction of microarrays, with which screening,of the full genome for changes in gene expression is possible. This method can provide a wealth of information about,biology at the level of gene expression and is a powerful method to identify genes and pathways involved in various,processes. In this study, we report a detailed analysis of the full heat stress response in Drosophila melanogaster,females, using whole genome gene expression arrays (Affymetrix Inc, Santa Clara, CA, USA). The study focuses on,up- as well as downregulation of genes from just before and at 8 time points after an application of short heat hardening,(368C for 1 hour). The expression changes were followed up to 64 hours after the heat stress, using 4 biological,replicates. This study describes in detail the dramatic change in gene expression over time induced by a short-term,heat treatment. We found both known stress responding genes and new candidate genes, and processes to be involved,in the stress response. We identified 3 main groups of stress responsive genes that were earlyâ??upregulated, earlyâ??,downregulated, and lateâ??upregulated, respectively, among 1222 differentially expressed genes in the data set. Comparisons,with stress sensitive genes identified by studies of responses to other types of stress allow the discussion of,heat-specific and general stress responses in Drosophila. Several unexpected features were revealed by this analysis,,which suggests that novel pathways and mechanisms are involved in the responses to heat stress and to stress in,general. The majority of stress responsive genes identified in this and other studies were downregulated, and the,degree of overlap among downregulated genes was relatively high, whereas genes responding by upregulation to heat,and other stress factors were more specific to the stress applied or to the conditions of the particular study. As an,expected exception, heat shock genes were generally found to be upregulated by stress in general.
